Particular attention must be paid to the following warnings:
DANGER
Risk of electric shock
−The power plug is equipped with an protective earth conductor, therefore, it has to be
used with an appropriately equipped socket.
−The protective capacity must not be rendered ineffective by the use of extension cables
with no earth conductor.
−In case of error there is a risk of contact with life-threatening mains voltage.
DANGER
Risk of damage to the device from penetrating moisture
−
Devices with a mains connection and their accessories (particularly external table power
pack) must not come into contact with moisture or liquids.
−If the housing of devices or accessories that conduct mains voltage (e.g. table power pack)
is damaged, these must be permanently taken out of service and disconnected from the
mains.
−Non-compliance in both cases results in exposure to life-threatening mains voltage.
WARNING
Risk of electric shock
−Always switch off the device and pull out the power plug before cleaning or opening the
device.
−Disconnect the power plug before changing device fuses. Failure to do so may result in
contact with life-threatening mains voltage.
CAUTION
Risk of electric shock, overheating of the power cable, in worst case, fire
Where there are detachable power cables, please note the following:
−Detachable power supply cables must not be replaced with power supply cables that do
not meet the specifications.
−Use only the specified power cables to avoid these becoming severely overheated.

CAUTION
Risk of ineffective safety measures
−
If it is determined that protective measures are no longer effective, the device must be
taken out of service and secured against inadvertent operation, to avoid the risk of injury.
−Contact ZEISS Service to have the device repaired.
CAUTION
Risk of fire from heat accumulation
−Always check the device is switched off before covering it with a dust cover.
−Clogging or covering of ventilation slats is also prohibited.
−Failure to comply with these two instructions can lead to an accumulation of heat, which
can damage the device and, in the worst-case scenario, cause a fire.
CAUTION
Risk of damage to the device due to incorrect or improvised fuses
−When changing the device fuses, only fuses specified in the technical data may be used.
−The use of other fuses and short-circuiting the fuse box are prohibited, to avoid the risk of
overheating, smoke formation or fire in the device.
NOTICE
Environmental damage due to improper disposal
−Do not dispose of defective equipment in regular domestic waste. It should be disposed of
in accordance with the prevailing national legal requirements. The manufacturer of the
device is under the legal obligation to take defective devices back.
−
Sample waste should be disposed of in accordance with the prevailing national legal
requirements.

4NOTES ON WARRANTY
The manufacturer guarantees that the instrument is free of material or manufacturing defects upon
delivery. Any defects must be reported immediately and steps taken to minimize damage. If such a defect
is reported, the instrument manufacturer shall be obliged to correct the fault, either by repairing the
instrument or replacing it with a new one, at the manufacturer's discretion. No warranty is given for
defects caused by natural wear and tear (particularly of wearing parts) and improper use of the
instrument.
The instrument manufacturer shall not be liable for damage caused by misuse, negligence or any other
tampering with the instrument, particularly the removal or replacement of instrument components, or
the use of accessories from other manufacturers. Such actions shall invalidate any warranty claims.
With the exception of the work described in this quick reference guide, no maintenance or repair work is
to be carried out on the ApoTome.2. Repairs may only be performed by ZEISS Service or individuals
specially authorized by ZEISS Service. In the event of a problem with the instrument, please contact your
local ZEISS representative.

The ApoTome.2 allows depth-discriminated images (= optical sections) of fluorescence specimens to be
produced. Compared to the conventional reflected light fluorescence methods, these optical sections
feature increased contrast and enhanced optical resolution in axial direction. Furthermore, optical
sections through the specimen are the prerequisite for the three-dimensional reconstruction of structures.

The ambient conditions (humidity, temperature, pressure) specified in Section 7 should be adhered to.
When documenting fluorescence preparations, do not expose the microscope to direct sunlight. This
helps minimize the influence of stray light.
For prolonged experimental procedures using the microscope (e.g. involving incubation or long image-
recording procedures) choose an installation location with minimum vibration.
Vibration stemming e.g. from forced ventilation, floor impact noise or other shocks can have an impact
on the supplementary modules of the microscope, some of which react sensitively. It can also lead to
reduced image quality and experimental errors.
To reduce vibrations at the microscope installation location, ZEISS offers various passive and active
damping options (Axio Imager (000000-0477-190 anti-vibration plate); small, air-damped system table
(000000-1984-812), table top mat TS 150).
Select an installation site which is as temperature-stable as possible for long-term observations.
For reliable long-term observations, the device must first warm up. The temperature equilibrium is
attained within 3-5 hours, depending on the light source, electronics and room temperature control. This
time must be planned into critical studies.
The ApoTome.2 system is an extension to the Axio Imager, Axio Observer, Axiovert 200 and
Axio Zoom.V16 microscopes.
The ApoTome.2 system consists of a slider, which is inserted in the plane of the luminous-field diaphragm
of the incident-light beam path, and an accompanying control box.
Before use, ApoTome.2 must warm up for at least 30 min.

9ESSENTIAL OPERATION CONTROLS
A complete ApoTome.2 imaging system comprises the following components:
−microscope
−anti-vibration table or anti-vibration microscope support
−cooled digital camera with high dynamic range
−PC with at least 1 GB RAM according to Carl Zeiss specifications plus monitor, operating system
Windows® Vista (32 bit version) or Windows®7 (32 bit version)
−ApoTome.2 slider with ApoTome control box
−Imaging software ZEN (blue edition)
This Quick Reference Guide describes the use of the ApoTome.2 hardware. For notes and instructions for
the control (calibration) of the ApoTome.2 through software, please refer to the Operating Manual of the
imaging software ZEN (blue edition).
The control elements of ApoTome.2 are reduced to the two available latch positions in the incident light
beam path and to the operating elements in the imaging software.
The ApoTome.2 hardware consists of two components:
1. slider with transmission grids changer
2. control box
The slider is directly connected to the control box via a cable. The control box is connected to the PC by a
USB cable or, optionally, it can be connected directly to the microscope by a CAN BUS cable. In this case,
the ApoTome.2 directly communicates with the PC via the electronic system of the microscope.
If an Axiovert 200 is used, the control box can only be connected to the PC by a USB cable. A
connection via CAN BUS is not possible!
Major features of the ApoTome.2 include:
−ApoTome.2 slider for the plane of the luminous-field diaphragm in the reflected light path
−ApoTome.2 slider with two click-stop positions:
Click-stop position 1:
An open passage is provided in the reflected light path. Thus, normal fluorescence observations can be
carried out in the wide field, e.g. to find interesting structures and to position the specimens.
Click-stop position 2:
In this position, a glass plate with evaporated grid structure is in the light path. The grid structure is
laterally moved in the specimen plane by means of a scanner mechanism. By capturing three (or more)
images at different grid positions and by subsequent calculation, it is possible to produce an optical
section through the specimen. Three different grid frequencies selected via software are available.
They allow the user to obtain different section thicknesses and to use different objectives.

•Carefully insert the ApoTome.2 slider in the plane of the luminous-field diaphragm of the reflected
light beam path of the microscope used (see complete operating manual of the microscope).
On insertion, a short beep signal is generated.
•Use the on/off switch (Fig. 3/3) at the front panel of the ApoTome.2 control box to switch the device
on.
•The power indicator in the switch lights up.
Switch on the device approximately 15 minutes before the first image capture in order to allow it to
warm up to the necessary operating temperature.
The slider must be inserted in the microscope stand to warm up properly.

In the first click-stop position, conventional imaging is possible (free passage). In the second click-stop
position, the transmission grid is moved into the beam path.
Click-stop position 1
In click-stop position 1 of the ApoTome.2 slider, an open passage is in the reflected light beam path. In
this position the microscope can be operated in the conventional way. The slider does not affect any
function of the microscope stand used.
Click-stop position 2
In click-stop position 2, the transmission grid is positioned in the beam path of rays. All functions for the
adjustment and control of the instrument are operated via the software. For additional information, refer
to the Operating Manual of the software used.

•After use, switch the device off by activating the on/off switch (Fig. 3/3) at the front panel of the
ApoTome.2 control box.
•The power indicator in the switch turns off.

Nachstehende Hinweise sind besonders zu beachten:
GEFAHR
Gefahr durch elektrischen Stromschlag
−Der Netzstecker des Gerätes ist mit einem Schutzleiterkontakt ausgeführt, deshalb darf
dieser nur in eine Steckdose mit Schutzkontakt eingeführt werden.
−Die Schutzwirkung darf nicht durch ein Verlängerungskabel ohne Schutzleiter außer Kraft
gesetzt werden.
−Im Fehlerfall kann lebensgefährliche Netzspannung berührt werden.
GEFAHR
Gefahr der Beschädigung des Gerätes durch eindringende Feuchtigkeit
−
Geräte mit Netzverbindung und deren Zubehör (insbesondere externe Tischnetzteile)
dürfen nicht mit Feuchtigkeit und Flüssigkeiten in Berührung kommen.
−Ist das Gehäuse von Netzspannungsführenden Geräten oder Zubehör (z. B. Tischnetzteil)
beschädigt, sind diese dauerhaft außer Betrieb zu nehmen und von der Netzspannung zu
trennen.
−
Bei Nichtbeachtung kann in beiden Fällen lebensgefährliche Netzspannung berührt
werden.
WARNUNG

−Vor Reinigung oder Öffnen des Gerätes
ist dieses auszuschalten und der Netzstecker zu
ziehen.
−Vor dem Wechseln von Gerätesicherungen muss der Netzstecker gezogen werden! Bei
Nichtbeachtung kann lebensgefährliche Netzspannung berührt werden.
VORSICHT
Gefahr durch elektrischer Schlag, Überhitzung des Netzkabels, Extremfall Brand
Wenn abnehmbare Netzkabel vorhanden sind, dann ist folgendes zu beachten:

Abnehmbare Netzkabel dürfen nicht durch unzulänglich bemessene Netzkabel ersetzt
werden.
−Es dürfen nur die vorgeschriebenen Netzkabel ve
rwendet werden, weil diese sich sonst
stark erwärmen können.

Wird festgestellt, dass Schutzmaßnahmen nicht mehr wirken, so ist das Gerät außer
Betrieb zu setzen und gegen unbeabsichtigte Benutzung
zu sichern, damit keine
Verletzungsgefährdung eintritt.
−Zur Instandsetzung des Gerätes ist Verbindung mit dem ZEISS-Service aufzunehmen.
VORSICHT
Brandgefahr durch Wärmestau
−
Vor Abdecken des Gerätes mit einer Staubschutzhülle muss immer geprüft werden, ob
dieses ausgeschaltet ist.

4GARANTIEHINWEISE
Der Gerätehersteller leistet Garantie dafür, dass das Gerät bei Übergabe frei von Material- und
Fertigungsfehlern ist. Aufgetretene Mängel sind unverzüglich anzuzeigen und es ist alles zu tun, um den
Schaden gering zu halten. Wird ein solcher Mangel gemeldet, so ist der Gerätehersteller verpflichtet, den
Mangel nach seiner Wahl durch Reparatur oder Lieferung eines mangelfreien Gerätes zu beheben. Für
Mängel infolge natürlicher Abnutzung (insbesondere bei Verschleißteilen) sowie unsachgemäßer
Behandlung wird keine Gewähr geleistet.
Der Gerätehersteller haftet nicht für Schäden, die durch Fehlbedienung, Fahrlässigkeit oder sonstige Ein-
griffe am Gerät entstehen, insbesondere durch das Entfernen oder Auswechseln von Geräteteilen oder
das Verwenden von Zubehör anderer Hersteller. Hierdurch erlöschen sämtliche Garantieansprüche.
Mit Ausnahme der in dieser Kurz-Gebrauchsweisung aufgeführten Tätigkeiten dürfen keine Wartungs-
oder Reparaturarbeiten an ApoTome.2 ausgeführt werden. Reparaturen sind nur dem ZEISS-
Kundendienst oder durch diesen speziell autorisierten Personen gestattet. Sollten Störungen am Gerät
auftreten, bitte zuerst an die für Sie zuständige ZEISS-Vertretung wenden.

Mit ApoTome.2 ist es möglich, tiefendiskriminierte Bilder (= optische Schnitte) von Fluoreszenzpräparaten
zu erstellen. Diese optischen Schnitte weisen im Vergleich zu konventionellen Auflicht-
Fluoreszenzmethoden einen erhöhten Kontrast und eine verbesserte optische Auflösung in axialer
Richtung auf. Außerdem sind optische Schnitte durch das Präparat die Voraussetzung für die
dreidimensionale Rekonstruktion von Strukturen.
