AccuStrata Fluorotek 200 User’s Manual
1. Introduction
1.1. Description
The Fluorotek 200 is a small form factor handheld fluorometer that is optimally designed for use
in the detection of bacteria. Dual channel design allows users to test for a variety of bacteria
using various assays.
1.2. Quick View Diagram
1.3. Inspection and Setup
1.3.1. Inspection
Upon receiving your device, please inspect everything carefully and make sure all accessories
are present. All devices include:
The Fluorotek 200 handheld unit.
Light-blocking cap for sample holder
A USB drive containing PC software and pdf documents for quick start and user’s
manual.
1.3.2. Setup
Battery installation:
1. On the back side of the instrument, remove the battery panel by pressing slightly on the
corrugated tab with thumb and pulling down.

3 | P a g e
AccuStrata Fluorotek 200 User’s Manual
2. Install 4 - AA batteries as indicated at the bottom of the battery housing.
3. Replace the battery protection panel by inserting the panel into the groves and sliding up
into position.
1.4. General Information, Precautions and Cleaning
The sample compartment is designed to accommodate standard polypropylene 0.5 ml
Polymerase Chain Reaction tubes as shown below:
Any other sample holder besides those displayed above are not guaranteed to fit
properly in the sample holder. Do not use solvents inthe test tube that may penetrate or
degrade the integrity of the tube. See tube manufacture instructions for further
information.
If a sample is accidentally spilled inside the sample compartment, invert the Fluorotek
200 unit to drain the fluid from the compartment. Then carefully wipe the inside of the
compartment with a clean cotton swab or cloth. Allow the unit to dry before using.
The Fluorotek 200 is NOT waterproof and should be kept away from water and moisture.
Do not expose the Fluorotek 200 unit to temperatures below 5 ⁰C or above 40 ⁰C.
2. Instrument Operation
2.1. Instrument Power Up
To turn on the Fluorotek 200, toggle the power switch on the top right side of the unit to the left.
To turn the unit off, toggle the power switch right.
If the Fluorotek 200 is left on for more than 30 minutes, the unit will power off automatically.
Once the system is turned on, a LOADING SCREEN will appear followed by the HOME SCREEN:
Once the HOME SCREEN appears, the system is ready for use.

3. Sample Analysis Guidelines
3.1. Handling Samples
Keep the sample chamber dry. Do notspill liquid into the sample chamber.
If a spill occurs, immediately invert the unit to remove the liquid.
Allow to fully dry before proceeding.
Condensation on the outside of test tube may result in read errors. Ensure that the
sample temperature has reached ambient temperature before placing the tube into the
sample chamber.
Never reuse test tubes for multiple samples
Bubbles or aeration within the sample may result in read errors. To remove air bubbles
from the sample, hold vertically and rapidly tap the outside of the tube with fingers.
3.2. Positioning Samples
The orientation of the test tube may result in slightly different readings, especially for low
fluorescence readings. This is due to inconsistency in quality of the polymer test tube. For
proper use, ensure that the tab of the tube is oriented the same way for every measurement.

3.3. General Data Acquisition Instructions
To take a measurement press the Measure button on the HOME SCREEN. This will load the
CHANNEL SCREEN. Select the desired channel to use for the measurement to load the
MEASUREMENT SCREEN.
To ensure that thetest tube does not influence the result, the tube and media must be zeroed.
To do this, insert the blank test tube (tube with media absent of fluorescing material) and press
Blank. This will bring up the TESTING SCREEN.
The value measured through this process will be subtracted from any future measurement
results. To reset the blank value to zero, use the Return button to load the HOME SCREEN.
Once the sample has been blanked, a measurement can be taken using the Measurement
button. This button will initialize the TESTING SCREEN followed by the RESULTS SCREEN.

6 | P a g e
AccuStrata Fluorotek 200 User’s Manual
The top of the TEST SCREEN displays the number of minutes at which the measurement was
taken after the test was initiated. Once the measurement is complete, the RESULTS SCREEN is
displayed. The RESULTS SCREEN displays the time of the reading and the measured values,
which is:
𝑅𝑒𝑎𝑑𝑉𝑎𝑙𝑢𝑒 = 𝑅𝑎𝑤𝑅𝑒𝑎𝑑 − 𝐵𝑙𝑎𝑛𝑘𝑅𝑒𝑎𝑑
After the first read, a 10 minute timer is started. This is the average amount of aging required for
specific bacteria detection reagents to sufficiently fluoresce. Once the 10 minute time is
reached, a second measurement is automatically taken. While the timer is counting, the user
can save the first read to memory using the Save button or cancel the read by using the Return
button. To the right of the timer is the data save location. Each light channel has a set of
memory slots that are numbered. When a new reading is saved to the system, the value is
assigned to a memory slot that has an integer value associated with it. This integer value that
represents the data slot that the measured value will be stored in is displayed to the right of the
timer value. This number also corresponds to the table position of downloaded values on the
software application. The values can also be reviewed on the system itself.

3.4. Reviewing Data
3.4.1. Review Data on Fluorotek 200
To review data that has been saved to the Fluorotek 200, press the Data button on the HOME
SCREEN. This will display the CHANNEL SCREEN. Select the wavelength that the data to be
reviewed was taken with. Finally, the DATA SCREEN will be displayed. The DATA SCREEN allows
the user to scroll through all of the datasets currently saved on the unit by pressing the <- and -
> position on the <-0000-> button.
3.4.2. Review Data with PC Software
First attach the Fluorotek 200 unit to the computer. Next, open the software application. Finally,
press the Read All Data button. The data will be displayed in tabulated form on the right of the
application screen. If the Fluorotek 200 unit is connected to the computer after the application
