AZ-X-Y Probe

necessarytoflushforanextendedperiodoftime)themembranemay
“sweat”orappearwetthisisnormal,notduetodamageoraclog.
2. Ifnoflowisobservedviatheoutletneedle,soaktheprobeindistilled
water for a few minutes and repeat instructions in item 1. After
conrmingthedistilledwaterflowfromtheoutlet,pleaseremovethe
probefromplasticsheathandconrmnoleaking.
3. We test each probe for defect/leaking prior to shipping. If you nd
adefectiveprobeafterfollowingtheabovestepsEicomwillreplace
theprobeatnocharge.Wedonotguaranteeprobesafterinsertion
toanimals.
Removing Air Bubble
1. Ifthereisanairbubbleintheprobe,pleaseremoveit.Becausethe
airbubblewillcovertheactivemembranesiteandkeeptheperfusate
away.Thiswillresultinalowerrecoveryrate.Theairbubbleshouldbe
removedbeforestudy.Ifitissmall,itwilldisappearincaseofinvivo
studybutmaynotinvitro.
2. Perfusateneedstobedegassedbeforeuse.
3. Toremovetheairbubble,pleaseintroduceabigairtotheinletand
flushout.Donotconnectoutlettubingatthistime.Thebigairbubble
willtrapandremoveasmallairbubble.
Connecting Outlet
1. Conrmneitherleakingorairbubblesarepresentinthemembrane.
Settheflowrateonthesyringepump.Astandardflowrateis1-2μl/
min.Ahighflowratecoupledwithalongoutlettubingcanresultin
damageoftheprobemembrane.
2. Connect Teflon tubing (model JT-10) or other tubing to the outlet.
A long tubing length (more than 150 cm) can cause leaking of the
membraneortheliquidswivelduetotheinducedhighpressure.Ifyou
usetubinglongerthan150cm,pleaseavoidusingflowrateshigher
than 2 μl/min. Polyethylene tubing can cause confounding peaks
whenanalyzingsamplesusingHPLCwithelectrochemicaldetection.
Cleaning Probes and Storage
Removetheprobefromthesystemandflushwithdistilledwatertowash
out salts. Then put the probe in the distilled water and place at 4°C for
storage. Keep the membrane wet to prevent itfrom shrinking. Keep the
probeawayfromphysicalshock.
Cleaning Lines (Teflon, FEP tubing)
Bacteriagenerationandproteinpollutionoftubingcanbeacauseoflow
recoveryrates,especiallyintheoutlettubing.Inordertoavoidthisproblem,
washthelineswith50timesdilutedcommercialkitchenbleach(chlorine)in
purewater.Perfusethesolutionintothelines(nottheprobe)andleaveitfor
atleastonehour,thenthoroughlyrinsewithwater.
Perfusion Solution
Wearecommendusinganon-bufferedaCSFsolutionbecausephosphate
buffersolutionsmayprecipitatecalciumphosphateandcanresultinclogs.
Probe Check
1. Injectdistilled water using a1 or 2 mldisposable (plastic) syringe.
Connect the syringe to the inlet (longer needle) of the probe using
Bitontubing(JB-30).Theprobemaybecomecloggedordamagedif
youinitiallyflushwithasaltsolution.Werecommendusingdistilled
waterfortheinitialprobeflush.Donotconnecttubingtotheoutlet
oftheprobeduringflushing.Pleaseconrmtherearenoleaksfrom
