INTRODUCTION
Thank you for purchasing the AnaPrep 12 system which is a fully automated,
stand-alone instrument that can purify nucleic acids within 30-45 minutes. With
advanced magnetic bead separation technology, it provides high quality extraction
results. Moreover, this easy to use interface frees users from troublesome settings
and time-consuming maintenance requirement.
This guide contains important information regarding the safe use of the AnaPrep
12 system. Please read this manual carefully, especially the Safety Information,
before operating the system for the first time,.

WARNING:
“WARNING”indicates a dangerous condition that may lead to death or serious
injury.
BIOHAZARD:
This symbol is used to indicate that certain precautions must be taken when
working with potentially infectious material.
CAUTION:
This symbol is used to indicate that non-compliance procedures from specified
instructions may lead to personal injury or may cause damage to the
instrument.
Important:
“Important”indicates that the operator/user should pay attention to the
specified instructions or prohibited actions.
Note:
“Note”indicates additional useful information.
HOT SURFACE:
This symbol is used to label potentially hot surfaces from the instrument.
For your safety and that of others, follow the guidelines provided in the following
pages for operating the AnaPrep 12 system.

About Instrument
WARNING:
Ignoring the following precautions may lead to fire or electric shock.
- In countries other than US use a power cable that meets your country’s
standard, or contact your local distributor.
- Do not use the AnaPrep 12 system with voltage other than the voltage
specified on the device.
- Do not use the AnaPrep 12 system with a damaged power plug or a loose
socket.
- If there is dust on the prongs of the power plug or on the plug socket,
remove it with a dry cloth.
- When you disconnect the plug from the outlet, be sure to hold the power
plug itself. Do not pull the power cable.
- For maintenance, disconnect the power plug from the outlet.
- Avoid contact with instrument or electrical cord and plug during storms.
Do not pour any liquid on the AnaPrep 12 system.
Do not place any objects containing liquid on the AnaPrep 12 system. Doing
so may cause a device failure, fire, or electric shock.
In the event the device overheats, starts to smoke or smells strange,
immediately unplug the power cable.
CAUTION:
Never attempt to reconfigure or disassemble the AnaPrep 12 system
without the manufacturer’s permission. Doing so may cause fire or electric
shock.
Do not place or drop objects on the AnaPrep 12 system. Also refrain from

bumping or knocking it, as doing so may cause a failure or malfunction of
the AnaPrep 12 system.
If any liquid or wet material is left inside the device, wipe it up with a soft
paper tissue, etc. Otherwise, the AnaPrep 12 system may be damaged.
Repairs to the AnaPrep 12 system should only be performed by personnel
specifically authorized by the BioChain Institute Inc.
Only original BioChain Institute Inc. replacement parts should be used.
If the equipment is used in a manner not specified by the manufacturer,
the protection provided by the equipment may be impaired.
About Reagent Kits
CAUTION:
When handling any of the kits, refer to the respective handbooks.
Reagents in each kit should be handled by observing the safety information
and precautions specific to the kit.
Extraction should be performed in an appropriate laboratory or workplace.
Note:
Kits are not included with the AnaPrep 12 system and must be ordered
separately.
About Samples
BIOHAZARD:
Always wear appropriate protective gear when handling any infectious
samples.

About Infectious Wastes
When handling or disposing of infectious materials, follow the laboratory
guidelines or the law regarding infectious waste to perform proper
incineration, sterilization, and/or disinfection.
When using a third party to dispose of hazardous waste, be sure to
outsource this work to an agency that is licensed to handle medical waste.

1.1 Composition of the AnaPrep 12 system

1.3 Operating Environment / Condition
Use the AnaPrep 12 system in a location that meets the following
conditions:

The space allocated for the AnaPrep 12 system must be at least 5-10
cm from the instrument to the side walls.
At an area where adequate power can be provided.
Where the temperature is 15 to 30°C and relative humidity is 30 to
80% (non-condensing).
On a flat and stable surface, with no vibration.
Away from direct sunlight (Close curtains or blinds if necessary).
Surrounding area must be well-ventilated and not dusty.
Temperature must be kept at a constant rate. Warming a cold room
suddenly or moving the AnaPrep 12 system from a room with low
temperature to a warm room may cause condensation inside the
device, resulting in abnormal operation.
Temperature and humidity must be kept within the specified range
(far from water taps, water heaters, humidifiers, air-conditioners and
heaters).
Far from objects which generate strong magnetic fields (motors,
transformers, TVs, audio speakers, magnets, etc.). Bringing the
AnaPrep 12 system close to any type of magnetic field may cause

malfunction.
Warning:
Do not use the AnaPrep 12 system in a location where it is wet or may
potentially be splashed with water. Doing so may cause device failure, fire,
or electric shock.
When relocating the AnaPrep 12 system, disconnect the plug from the
outlet. If the power cable is damaged, this may cause device failure, fire,
injury, or electric shock.
Caution:
Do not use the AnaPrep 12 system in an unstable place such as a slanted
surface or in a place subject to vibrations. It may cause personal injury or
device failure.
Do not use the AnaPrep 12 system in direct sunlight or close to a heating
device. It may shorten the life of the AnaPrep 12 system or cause equipment
malfunction.

1.4 Unpack the AnaPrep 12 system
(1) Open the packing box and take out the instrument and related
accessories.
Important:
The AnaPrep 12 system weighs more than 43 kg (approx. 95 lbs).
It should be lifted and moved by at least two individuals.
Hold the moving handles of the instrument from two sides to lift it
out from the box.
Do not hold the plastic outer covering.
Do not hold the front panel.
Do not hold the door.
Caution:
Improper handling or movement of the AnaPrep 12 system will
lead to instrument damages.
For proper and safe use of AnaPrep 12, install it in a location that
is near an electrical outlet and has enough space for installation
and operation of the main switches.

Important:
Keep the shipping box and packaging materials. They will be needed
again when transporting the AnaPrep 12 system.
(2) Connect Barcode Reader with the instrument.
(3) Remove the polystyrene foam block inside the instrument by following
the steps below:
i. Plug in the power cord to the instrument and connect to the
electrical outlet.
ii. Turn on the power switch.
iii. Press the “START” button on the Control Pad. After the piston
module moves up, remove the polystyrene foam block.

2 Getting Started
Biohazard:
Always wear appropriate protective gear during any biohazardous
operations. Always follow standard safety procedures and wear the
appropriate gloves, mask and safety goggles when handling specimens and
reagents.
Important:
Before starting extraction, put on appropriate gloves, mask, and safety
goggles. When preparing samples for extraction, be careful not to
contaminate the samples with sweat, saliva, etc.

2.1 Turning on the power
(1) Make sure that the power cable is connected securely to the AnaPrep
12 system.
(2) Connect the plug of the power cable to the outlet.
2.2 Preparation
The following preparations are required for the extraction operation.

- Reagent Cartridge(s)*

- Filtered tip*

For the preparation of samples, refer to the handbook of each kit.
*: Provided with the Reagent Kits.
2.3 Extraction
(1) Turn on the power switch then wait for the LCD screen to turn on and
display “AnaPrep 12 System Stand-By”.
(2) Press the “Start” button
(The system will initiate a self-testing process, “initializing”, before
entering steady mode)
Note:
The system will not respond to any information entered from the
key pad until the completion of the self-testing process.

(3) Open the sliding door and remove the sample rack from the
instrument.
(4) Load Reagent Cartridges, and all plastic disposables (Reaction
Chamber, Tip Holder, Piercing Pin, and Filtered Tip).

How to pull apart reagent cartridges
Cut the foil at the dotted line with a finger nail, then snap the cartridge apart.
