1 INTRODUCTION
1.1 INTENDEDUSE
TheCYANHematoisaneasytooperate,fullyautomated,benchtophematologycellcounterdesigned
for
invitro
diagnosticuse.Itcombinespowerfulfeaturesinacompact,independentunitmakingitideal
forsmallclinics,andpointofcarelaboffices.ItimplementstheCoultermethodforcountingcellspassing
throughasmallaperture.Theprincipleofspectrophotometryisusedtomeasurethehemoglobincontent
ofredbloodcells.
TheanalyzerfeaturesacolorgraphicalLCDdisplaymodulewithtouchscreen,andhasaseparateSTART
button.Thesoftwareallowssendingresultstoanexternalprinter(viaUSBport),orthebuiltinthermal
printer module. Its internal memory is capable of storing 10.000 records with full histograms, and
individualpatientdata.QCmeasurementsarestoredinseparatedatabase.Thesoftwareoperatingthe
instrumentiseasytoupgradeusingaUSBflashpendrive.Makingbackups,exportingdataforprocessing
onacomputerandrestoringrecordstoandfromaUSBpendriveisalsopossible.

1.2 MEASUREMENTPARAMETERS
Theanalyzercanprocess30samplesperhourinthe3partWBCdifferentialmode.Samplescanhave
individualsampledata,and separatetestparameters.Theusercan customizethe reportformat.The
analyzerdeterminesthefollowing20hematologyparametersfroma25µLwholebloodsample:

1.3 REAGENTS
TheCYANHematoisaclosedsystem,onlyreagentssuppliedbyCypressDiagnosticscanbeusedonthe
CYANHematoandviceversa.IfotherreagentsareusedontheCYANHematoorifthereagentsareused
onadifferentanalyzerthantheCYANHematothentheaccuracycannotbeguaranteed.
1. Diluent(Code:CY006R01):Isotonicsalinesolution,usedtodilutewholebloodsamplesandtorinse
thefluidicsystembetweenmeasuringprocedures.
2. Lyse(Code:CY006R02):Createshemolysatefor3partWBCdifferentialandforHGBmeasurement.
3. Cleaner(Code:CY006R03):Forcleaningprocessofthefluidics.
CypressDiagnosticsalsooffersemergency cleaners.Theseemergencycleanersare not designedfor
regularcleaning,butshouldbeusedincaseofproblematicblankmeasurements.
4. Hypoclean(Code:CY006R04):Hardcleaner(bleachbased).
5. Clenz(Code:CY006R05):Enzymaticcleaner.
6. HypocleanCC(Code:CY006R06):Hardcleaner,usedtocleanthesampleneedle.

1.4 QUALITYCONTROLBLOOD
The analyzer allows the continuous monitoring of measurement performance with control blood
(low/normal/high). Their specifications (assay values, allowed tolerances and expiry date) are always
includedinthecontrolpackage.

1.5 CALIBRATION
TheCYANHematoarrivestoyourlaboratoryfactorycalibrated.However,calibrationisworkingcondition
dependent (temperature, humidity, reagent lot …) and thus should be repeated at instrument
installation.Use normal control bloodfor calibration.Thetargetvalues arementionedontheinsert
providedwiththecontrolblood.Thetargetvaluesforcontrolbloodislotdependent.
1.6 BLOODSAMPLES
TheCYANHematomeasureswholebloodhumansamplestreatedwithK3EDTA.Thebloodneedsto
betreatedwithanticoagulanttopreventlargegroupsofcellsformingintoclotsthatwillclogthecell
counter.Thechoiceofanticoagulantisveryimportant,assomeanticoagulantswillaffecttheshapeand
sizeofbloodcells.IngeneralK3EDTA(orK2EDTA),aretheonlyanticoagulantsrecommendedforuse
withelectronicbloodcounters.
1.6.1 Samplecollection
Alwaysfollowtheinstructionsforusethatarespecificforthesamplecollectiontubethatisused.Meeting
therequirementsofthetubemanufacturerwillguaranteegoodqualitysampling.
Thepremanufacturedsamplingtubeshavetheanticoagulantpreparedinsidethevialinformofliquid
orpowder.Toensurethecorrectamountofbloodthatcanbetreatedwiththepredosedanticoagulant,
samplevialsareshippedwithvacuuminsidethatensuresaspirationoftherightamountofbloodduring
theoriginalsamplingprocess.Itisthusveryimportanttoneveropenthepremanufacturedsampling
tubesbeforesamplecollection,asthiswouldeliminatethevacuum.Thevialsthemselvesalsohaveamark
indicatingtheoptimalsamplelevelfortheanticoagulantalreadyincludedinthesamplevial.
Whenusing“homemade”tubespredosedwithEDTA,itisimportantthattheratioofEDTAtoblood
doesnotexceed3mg/mL.Ifthisratioisexceeded,thiswillresultinosmotictransferfromtheRBCswhich
shrinks them and thereby affect the measurements. Generally, we suggest using premanufactured
sampletubescontainingthenecessaryamountofEDTA.
Aftercollectingthecorrectamountofblood,itisveryimportanttoimmediatelymixthesample.Thevial,
afterremovalfromthesamplingadapterorsamplingneedlemustbegentlyinvertedseveraltimesto
allowhomogenousmixingofthebloodandthepredosedanticoagulantalreadypresentinthesampling
vial.

Asamplewithclotsinsidewillnotonlyhavefalsereadings(duetothefactthatthecoagulationprocess
decreasesthenumberofbloodcellssuspendedinthebloodsamplebycollectingthemintoclots)but
canalsocauseaclogginginsidethedelicatetubingsystemofahematologyanalyzer.Additionally,the
samplemustbedrawnagainfromthepatient,asclottedsamplesarenotsuitableforanalysis.
1.6.2 Samplestorage
Theanticoagulantrequiressometime(1530minutes)tostabilizeitseffectandcompletelystopthe
coagulationprocessfromstarting.BloodsamplesshouldberunonCYANHematoonlyafterthisreaction
timehaselapsed.
Blood samples can either be collected and analyzed at the same spot (making sure that the anti
coagulant’sreactiontimeisguaranteed)orcanbeanalyzedatanoffsitelocation.
Thecollectedsamplescanbekeptatroomtemperature(15–30°C)forupto8hours.Ifthesamples
cannotbeanalyzedwithinafewhoursaftercollection,samplesshouldbestoredatatemperatureof2
8°Ctodelaythedecayofthebloodcells.Whenthesamplesarestoredat28°Cimmediatelyaftersample
collection,thesamplescanbeanalyzedupto24hoursaftercollection.
Samplesmustbeprocessedwithinthetimeperiodsmentionedabove(timeisdependingonthestorage
temperature).Readingsmadeafterthesetimeperiodscanresultinbadornoninterpretablereadingsdue
tothedecayingeffectsofbloodcells.
Notes:
a. Thebloodsamplemustneverfreeze,asthis(freezingoftheliquidincludedinbloodplasma)damages
cellstructuresandcellmembranes,resultinginbadornoninterpretablereadings.
b. Thetimeperiodsmentionedabovecanonlybeobtainedwhenthebloodsamplesarestoredona
rollermixer.Therollermixerisidealforstoringbloodsamples.Themixingissoftandthesurface
betweentheliquidandtheairislarge,sothatthecellsremainincontactwithoxygen.Thiswillkeep
thecellsalivemuchlongerandthecellskeepalltheircharacteristics.
Ifthevialsareleftuprightinarack,thebloodsamplesinvialstendtosedimentwith
time. The cells sink down to the bottomof the vial leaving blood plasma in the top.This makes
diffusionfromoxygenfromtheairtothecellsimpossibleandthecellswilllosetheircharacteristics
and die. A wellhomogenized sample is a homogenous red color liquid. A sample that has been
sedimented,isdividedintosections:PLT(thrombocyte)richplasmafloatsonthetop,andathinwhite
layerisalsoformed(WBC,leukocytes–“buffycoat”)abovethedarkredbottomhalfmadeupofRBC
(erythrocytes).

1.6.3 Samplepreparation
1. Beforeanalysis,thesamplesneedtobehomogenized.
a. Theidealwaytohomogenizesamplestakenoutisputtingthemonarollermixerfor2030
minutesensuringagentlemixingofthecellsandplasma.
b. Immediatelybeforesampling,itisalsorecommendedtogentlyinverttheclosedsampletubeat
least8times.
c. Avoidshakingoraccidentallydroppingthesamplesasmeansofmixing,asthisactioncanbreak
thebloodcellsandmightintroducemicrobubblesthatwillalsoinfluencereadings.
2. Theanalysistemperaturedependsonthestoragetimeperiodofthesamples.
a. Ifstoredforlessthan6hoursthesamplesshouldbetakenfromthesamplecooler(orrefrigerator)
andgentlywarmeduptoroomtemperature.Avoidwarmingthemupimmediatelyinyourhand
asthismayintroduceaheatshocktothecellsandinfluencereadings
b. Ifstoredformorethan6hours,itisessentialthattheanalysisisperformedoncoldsamples.
Homogenize the samples and analyze them immediately. Do not let them warm to room
temperature,thiswouldresultitrapiddecayofthecellsandinfluencetheanalysis.
3. Alwaystakeoffthecapofthetubes!Otherwise,theneedlewillcrash!Theneedlecannotpiercecaps.
Note:
Ifthesamesampleneedstobemeasuredmultipletimes,pleasemixthesamplegentlybeforeevery
sampling(invertthetubeatleast8times).Makesuretowipethecapcleanbeforeplacingitagainon
thevial.
1.6.4 Samplehandling
Bloodsamplesmustalwaysbeconsideredasbiohazardandinfectiousbiologicalmaterial.
Itisrecommendedtowearprotectiveglovesandgoggles.
Spilledbloodmustbecleanedupassoonaspossible,andthesurfaceincontactwithit
mustalsobecleanedwithasuitablesanitizer.

A LED (light emitting diode) is located above theSTART button. Its color indicates the status of the
analyzer.
LEDCOLOR ANALYZERSTATUS
GREEN Theanalyzerisreadyforsamplemeasurement.Analysisisinitiatedbypressing
theSTARTbutton.
REDBLINKING Blood sample can be removed when the LED blinks red 3 times and the
instrumentbeeps3times.
RED Theanalyzeriscurrentlyperformingananalysis.Nonewmeasurementcanbe
started.
ORANGE Theanalyzerisperformingamaintenanceprocess.
ORANGEBLINKING Theinstrumentisinstandbyandthedisplaylightisoff.Hitthescreentowake
upfromstandby.
1.7.3 LCDTouchscreen
Thedisplayisa320x240dots,highcontrastbacklithighcolorgraphicLCDmodule,withintegratedtouch
screen.Afingerorsmallerbluntobject(styluspen)shouldbeusedtooperatethetouchscreen.Ifthe
operatorgentlytouchestheLCD,theanalyzerwillrecognizethepositionwherethescreenwaspressed;
thesoftwarewillactivatethecorrespondingfunction/menu/button.
!ATTENTIONPOINT!Thescreencanonlyreacttoonetouchpointatthesametime.Ifsomethingelseis
touchingorpressingthescreen(onanemptyposition),itwillnotrespondtoyourtouch!
1.7.3.1 KEYBOARD
Whendataneedstobeentered,akeyboardwillappear.Thekeyboardwillbeadaptedtothedatathat
canbeenteredinthatspecificarea;alphanumericornumericonly.

Press“Enter”toconfirmaninsertionandreturntothepreviousmenu,press“OK”.PressCancel”toreturn
tothepreviousmenuwithoutusingtheinsertion.
1.7.3.2 TOUCHSCREENCALIBRATION
Incasethe touch screendoesnotrespondcorrectlyto your tap(youtapa specific location,stillthe
requiredfunctionkeyisnotactivated)youhavetocalibratethetouchscreen.
Procedure:
1. Gentlytapandcontinuouslyholdanyemptylocationonthetouchscreenforatleast30seconds.
2. Afterca30seconds,acalibratingscreenappears.Tapthemiddleofthecrossofthereferencepoints
oneafteranother.
Ifyoumadeanerror,youwillhearanerrorbeep,andtheprocessrestarts.
Ifthecalibrationwassuccessful,thesoftwarewillreturntotheoriginalscreen.

1.7.4 Buildinthermalprinter
TheCYANHematoisequippedwithabuiltinthermalprinter.The
textorgraphisprintedoncoatedthermochromicpaper.Thispaper
isselectivelyheatedwhenpassingoverthethermalprintinghead,
andtherebythecoatingontheseplacesturnsblack,producingan
image.Oneofthebigadvantagesofathermalprinteroveranormal
printer is that it does not require expensive consumables like a
toner.
Pressthefeedbuttontomovethepaperforwardwithoutprinting.
Thefiguresbelowshowthebackviewoftheanalyzer,withcontrolsandconnectors.
