1.1 Important user information
Read this before operating the
product
All users must read the entire Operating Instructions before installing, oper-
ating, or maintaining the product.
Always keep the Operating Instructions at hand when operating the product.
Do not install, operate, or perform maintenance on the product in any other way than
described in the user documentation. If you do, you may be exposed or expose others
to hazards that can lead to personal injury and you may cause damage to the equip-
ment.
Intended use
The Media Wand and Media Handling Unit are intended for use in the preparation and
handling of bulk chromatography resin solutions for the separation of biomolecules
such as proteins, peptides and oligonucleotides in GMP-regulated environments.
Use of the Media Wand and Media Handling Unit facilitates the preparation of chroma-
tography resin solutions and the transfer of chromatography resin solutions to the
column or slurry tank. Media Wand can also be used together with Chromaflow™
Packing Station 100, see Section 5.6 Use of Media Wand with Chromaflow Packing
Station 100, on page 47.
The products are specifically developed for the simplification of the following proce-
dures:
•removal of transport solutions from resin containers.
•addition of buffer and preparation of resin slurry in a resin container.
•transfer of resin slurry to a process column or slurry tank.
The Media Handling Unit is not approved for use in a potentially explosive environment
or for other purposes than those described in this manual.
The Media Wand and Media Handling Unit are intended for production use only and
must not be used for diagnostic purposes in any clinical or in vitro procedures.
Prerequisites
In order to operate the products in a safe way and according to the intended purpose
the following prerequisites must be met:
•The user must be acquainted with the use of general process equipment and with
handling of biological materials.

•The user must read Chapter 2 Safety instructions, on page 9 of these operating
instructions.
•The products must be installed according to the instructions in Chapter 4 Installa-
tion, on page 26 of these Operating Instructions.
•The user must be trained in handling chromatography resin in general.

2.1 Safety precautions
Introduction
Before installing, operating or maintaining the instruments, you must be aware of the
hazards described in the user documentation.
Follow the instructions provided to avoid injury to the operator or other
personnel, damage to samples or other substances handled by the equipment,
to the product, or to other equipment in the area.
The safety precautions in this section are grouped into the following categories:

This user documentation contains safety notices (WARNING, CAUTION, and NOTICE)
concerning the safe use of the product. See definitions below.
WARNING
WARNING indicates a hazardous situation which, if not avoided,
could result in death or serious injury. It is important not to proceed
until all stated conditions are met and clearly understood.
CAUTION
CAUTION indicates a hazardous situation which, if not avoided,
could result in minor or moderate injury. It is important not to
proceed until all stated conditions are met and clearly understood.

General precautions
WARNING
Risk assessment. Perform a risk assessment for the process or
process environment. Evaluate the effects the use of the product
and the operational processes may have on the classification of the
hazardous area. The process can cause the hazardous area to
increase or the zone classification to change. Implement the risk
reduction measures needed, including use of personal protective
equipment.
WARNING
Only properly trained personnel are allowed to operate and main-
tain the product.
WARNING
Pressure may remain in the column and tubing for an extended
period after shutdown. Make sure the system is depressurized
before opening or disconnecting.
WARNING
Reduce the pump setting prior to using the RESET button, to avoid
an immediate high pressure increase in the system when it is reac-
tivated. A sudden pressure increase may cause rupture of piping
connections and consequent leakage.
Personal protection
WARNING
Always use appropriate Personal Protective Equipment (PPE)
during operation and maintenance of this product.

WARNING
Hazardous substances. When using hazardous chemicals, take
all suitable protective measures, such as wearing protective
clothing, glasses and gloves resistant to the substances used.
Follow local and/or national regulations for safe operation and
maintenance of the product.
WARNING
Use ear protection whenever working close to the system in opera-
tion.
Flammable liquids
WARNING
Explosive environment. The product is not approved for work in
a potentially explosive atmosphere.
WARNING
Some of the chemicals used with Media Wand and Media Handling
Unit may be flammable under certain conditions. Make sure to use
chemicals only under conditions where they are not flammable.
Refer to local and/or national classifications of liquids under flam-
mable conditions.

2.3 Emergency procedures
Introduction
This section describes emergency shutdown of the Media Wand and Media Handling
Unit in an emergency situation, and the procedure for restarting the Media Wand and
Media Handling Unit.
Precautions
WARNING
Pressure may remain in the tubing for an extended period after
shutdown. Make sure the system is depressurized before opening
or disconnecting.
Emergency shutdown
In an emergency situation, follow the steps below to stop the run.
Step Action
1Push the EMERGENCY STOP button on the Media Handling Unit.
2Remedy the cause of the emergency stop.
Restart after emergency shutdown
To reset the emergency stop, follow these steps:

Step Action
1Push the EMERGENCY STOP button.
2Turn the EMERGENCY STOP button in the direction indicated by the arrow.
3Reduce the pump setting.
WARNING
Reduce the pump setting prior to using the RESET
button, to avoid an immediate high pressure increase in
the system when it is reactivated. A sudden pressure
increase may cause rupture of piping connections and
consequent leakage.
4Set the valve control switches to 0.
5Press the RESET button.
Note:
The pump restarts immediately when the valves are re-opened.

Pump
The pump is designed to be able to be run dry. The pump can be stopped either by
setting the pump switch to OFF or by setting the valve controls to 0. The pressure from
the system automatically stops the pump. This does not damage the pump.
Only use dry, clean, oil-free compressed air with a minimum pressure of 5 bar g (max
7 bar g).
The capacity curve of the built-in pump is shown below.

Example: External pressure (air pressure) is 4 bar g.
To obtain a pump flow of 40 L/min, the pump pressure is set to 3 bar g, which results in
an air consumption of 0.3 m3/min.
