Before using the MACSxpress Separator, read the chapter Important safety
information and all other information contained in this user manual, including
any safety and operating instructions. Pay special attention to all warnings
displayed on the instrument. Failure to read and follow these guidelines could
lead to improper or incorrect usage and result in damage to the instrument.
Improper usage could also cause severe personal injury, death, unpredictable
results, instrument malfunction, and premature wear to components shortening
the lifetime of the instrument. Such actions may void your warranty. Keep the user
manual and any other safety and operating instructions provided with the
instrument in a safe place accessible to all users for future reference.

• Use this instrument only as indicated in this user manual to avoid injury to
personnel and damage of the equipment.

• In case of severe accidents, damages to the instrument, or if smoke or flames
appear, cut the power supply immediately.

Liquids inside the instrument
Liquids inside the instrument can cause short circuits.
• Do not allow liquids to enter the inside of the instrument.
• Do not use or clean the instrument without sealing the USB connection
with the blind plug.
• Use only small amounts of cleaning agents on a soft cloth to wipe the
instrument. Do not spray or pour liquid cleaning agents onto or into the
instrument.
• Protect the external power supply against spilled liquids.
• Clean up spilled liquids immediately.
• Do not use the instrument if liquids have entered the inside of it.
• Do not use the external power supply if liquids have entered it.
Humid and dusty environments
Humidity, condensation, or dust inside the instrument can cause short circuits.
• Do not use or clean the instrument without sealing the USB connection
with the blind plug.
Biological hazards
BIOLOGICAL HAZARD
Contamination or infection may lead to severe personal injury or
death, depending on the used material.
Biological material
All biological material must be considered potentially infectious.
• Follow local working area safety instructions and laboratory policies and
standards for health, safety, and prevention of accidents.
• Wear personal protective equipment (such as gloves, safety glasses, etc.) as
indicated in the documentation for the particular substance.
• Decontaminate the instrument after processing hazardous or potentially
infectious material.
• If hazardous or potentially infectious material has been spilled or leaked
from the system, decontaminate the affected area.
• Use a disinfectant or cleaning agent that is appropriate for the specific
pathogen or substance.

• Do not use bleach.
Sample processing
Incorrect sample processing may lead to contamination or infection.
• Use only specified sample container.
• Follow the protocol in the respective data sheet.
• Process maximal 30 mL of blood product sample in one isolation step.
• Do not pour off the supernatant, as the tube will fall out of the MACSxpress
Separator.
Equipment damage
Defective or inadequate equipment can cause a biological hazard.
• If parts of the instrument are damaged, unplug and do not use the
instrument until damaged parts are replaced. Contact Miltenyi Biotec
Technical Support, if necessary.
Waste
All liquid and solid waste must be considered hazardous.
• Follow local regulations regarding waste disposal.
• Autoclave or decontaminate waste with an appropriate disinfectant.
• Follow the general laboratory safety regulations when handling liquid and
solid waste.
Chemical hazards
CHEMICAL HAZARD
Substances and reagents can be hazardous.
• All safety measures in section Biological hazards also apply to any
hazardous substances and reagents that may be present in the sample.
• Use any substances and reagents only as stated in the respective data sheet.
• Operate the instrument in a fume hood if hazardous substances and reagents
are processed.
• Only use reagents provided by Miltenyi Biotec if not otherwise stated in this
manual.

Magnetic field hazards
STRONG MAGNETIC FIELD
Strong magnetic fields can influence the functioning of pacemakers or
electronic medical implants.
• If wearing pacemakers or electronic medical implants keep a distance of at
least 30cm.
Magnetizable objects can suddenly move towards the magnet.
• Keep all magnetic storage devices, electronic equipment, and magnetizable
objects at a distance of at least 30cm.
Mechanical hazards
CRUSH HAZARD
Risk of crushing.
The instrument may fall if it is not placed on a flat and even surface. This may
cause injury.
• Place the instrument only onto stable, even, and vibration free tables or
laboratory benches.
SHARP ELEMENTS
Risk of cutting.
Tubes
Unspecified sample containers (e.g. made of glass) may break. This may lead to
injury.
• Do not use sample containers made of glass.
• Do not pour off the supernatant, as the tube will fall out of the MACSxpress
Separator.
• Follow the protocol in the respective data sheet.

applicable WEEE and hazardous waste disposal legislation, which may differ by
country or region. Electrical equipment may contain hazardous substances
that may have a serious detrimental effect on the environment and/or human
health. All equipment must be specifically collected and treated by
designated waste facilities and by qualified WEEE compliance schemas. By
ensuring that you dispose of your unwanted electrical and electronic
equipment according to the applicable WEEE and hazardous waste disposal
legislation, you are helping to preserve our natural resources and protect

INTRODUCTION
2.1 Intended use
The MACSxpress Separator is a laboratory equipment for manual separation of
magnetically labeled cells. The device contains a permanent magnet for cell
separation and uses LEDs with a rechargeable battery for illuminating of the
target cell fraction. The MACSxpress Separator is used for untouched isolation
of leukocytes from blood products without density gradient centrifugation.
Erythrocytes are sedimented, while non-target cells are removed. Up to 30 mL
of prepared sample can be processed at a time. The instrument is for research
use only and not for therapeutic or diagnostic use. It is to be used by
professional laboratory personnel only.
2.2 Separation principle
The MACSxpress Separator is developed for the untouched isolation of
leukocytes from up to 30 mL anticoagulated whole blood without density
gradient centrifugation. Erythrocytes are aggregated and sedimented, while
non-target cells are removed by immunomagnetic depletion with
MACSxpress Beads. The tube containing the labeled sample is placed in the
magnetic field of the MACSxpress Separator. The magnetically labeled cells
will adhere to the wall of the tube while the aggregated erythrocytes
sediment to the bottom. While the tube is still inside the MACSxpress
Separator, the supernatant containing the non-labeled target cells can be
carefully pipetted into a new tube (Figure 2.1).

3.3 Assemble the instrument
The instrument has powerful magnets.
Strong magnetic fields can damage equipment susceptible for magnetic
fields.
• Keep all magnetic storage devices, electronic equipment, and
magnetizable objects at a distance of at least 30cm.
1Connect the USB cable with the power supply.
2To attach the power supply for charging, remove the blind plug from the
MACSxpress Separator USB port at the rear side of the instrument.
3Connect the USB cable to the USB port.
4After charging, unplug the power supply from the electrical outlet.
5Disconnect the MACSxpress Separator from the USB cable and re-attach
the blind plug to the USB port.
The blind plug provides protection against spillages and splashes.

OPERATE THE INSTRUMENT
4.1 Charge the instrument
Overloading of batteries can lead to leakage of battery fluid.
• Only use the included power cable.
1Before using the MACSxpress Separator fully charge the integrated NiMH
battery.
2Connect the MACSxpress Separator to the power supply using the
provided cable (refer to Assemble the instrument on page16).
3Plug the power supply into an appropriate electrical outlet. The LED on
the MACSxpress Separator indicates the battery charge status (Table 4.1).
4After charging, unplug the power supply from the electrical outlet.
5Disconnect the MACSxpress Separator from the USB cable and re-attach
the blind plug to the USB port.

4.2 Switching the light on or off
1Press the button labeled IO for a few seconds to switch on the light.
2The light will turn off automatically after 4 minutes.
3Alternatively,pressthe button again for a few secondsto switch offthe light.

4.3 Perform a separation
Insufficient light can lead to handling faults and impair experiment
results.
• Provide for sufficient light.
1Position the MACSxpress Separator stable on a laboratory bench.
2Open the tube containing the labeled sample.
3If necessary, remove air bubbles on top of the sample with a pipette.
4Place the open tube in the magnetic field of the MACSxpress Separator
for 15 minutes. The magnetically labeled cells will adhere to the wall of
the tube while the aggregated erythrocytes sediment to the bottom.
5Switch on the light.
Do not pour off the supernatant, as the tube will fall out of the
MACSxpress Separator.
6While the tube is still inside the MACSxpress Separator, carefully pipette
supernatant containing the non-labeled target cells into a new tube.
