Principles of Operation:
The Scienceware®Colony Counter marks, counts, and gives an audible and visual alert each time a count is
registered. The lightweight, pen-style design reduces the likelihood of error, combining all aspects of counting into
one motion. The Scienceware®Colony Counter can store up to 35 individual samples in its memory.
Operation Instructions:
BEFORE OPERATION open battery compartment using a small Phillips-head
screwdriver. Pull metal Battery Release Tab back, away from battery, and
pull up on Paper Tab to remove the battery. Discard Paper Tab. Reinsert
battery into housing, pushing firmly to click into place. Reassemble battery
compartment.
Power button – Turns colony counter on and off.
Note: turning off the colony counter will clear the memory
Power will shut off automatically after 3 minutes of inactivity, but will NOT clear memory
LED – Visual indication when a count has been added or subtracted
Lock – Locks and unlocks the counting function. When the lock is on, a lock symbol will appear in the display
Button 1 – Count Up or Down/Clear memory
The colony counter default setting is counting UP but can be set to DOWN to verify count.
Button 2 – Sample Identification Selection
The colony counter can store up to 35 plates under separate sample identification numbers
Sample ID number will scroll in a continuous loop from 1 through 35.
Sum Button – The counter will display the total number of colonies counted
under all Sample Identification numbers
Sound Button – Turns audible sound on and off.
To Count Samples:
1. Insert Scienceware®pen into colony counter and remove pen cap.
2. Press tip onto surface above colony. When a count is successfully recorded, the following things will occur:
Pen tip will mark surface of plate cover
Audible beep will sound (if sound is ON)
LED will flash once
Counter total will INCREASE
3. To change to a new plate, press Button 2
The Sample ID number will advance to the next number
Note: the counter maximum for each sample ID
is 9999. When the maximum is reached,
the LED will flash 3 times and count will
not increase

Caution! Do not use Colony Counter if:
1. The unit is damaged or broken
2. The battery lid is not closed
3. The unit or the end-user’s hands are wet

To verify a sample count:
1. After counting colonies using one pen color, switch to other pen color.
(We recommend starting with red and switching to black)
2. Press Button 1 one time. The Up arrow will disappear and the Down arrow will appear.
Note: If BOTH arrows appear and flash in the display, do NOT press Button 1 again until the arrows
STOP flashing. Pressing Button 1 again while both arrows are flashing will clear the memory for that
sample identification number.
3. Recount colonies in plate. During recount:
Pen tip will mark surface of plate cover
Audible beep will sound (if sound is ON)
LED will flash once
Counter total will DECREASE
Note: The original sample count (1st) will be continuously shown in the upper right hand corner of the
display. The TOTAL count (2nd) will be shown in the lower right hand corner. The TOTAL
count is the sum of the original count plus the recount.
Changing SAMPLE ID number:
To advance the sample ID number: Press Button 2
Audible sound will indicate change
Sample ID number will INCREASE (e.g. 01->02->03->…35)
To review the sample ID numbers:
1. Press and hold BOTH Button 2 and the Back Light button. Audible beep will sound
2. Press Button 2
Audible sound will indicate change
Sample ID number will DECREASE (e.g. 35->34->33->…1)
To clear a specific SAMPLE ID memory:
1. Scroll through the Sample Identification numbers by pressing Button 2 repeatedly until
the correct Sample ID number is shown in the display.
2. Press and hold Button 1 until both Up and Down Arrows flash in the display.
3. While both arrows are flashing, press Button 1.
LED will flash once
Audible Beep will sound (if sound is ON)
Counter display will show +0000
To clear ALL Sample ID memories:
1. Press and hold Power button for 3 seconds. Display becomes blank. Unit is turned off.
2. Press Power button to turn unit back on. Memory has been cleared.
Battery Replacement:
1. Locate and loosen retaining screw on battery cover on the underside of the unit.
2. Depress battery retaining clip to release battery from holder
3. Insert new battery with + side facing UP.
4. Slide battery cover back into place and tighten retaining screw to secure cover in place.
Cleaning:
To clean unit, wet paper towel with 70% ethanol solution and wipe down unit.
Helpful Hints:
1. Press firmly to make sure pen depresses with each count. The counter will emit an
audible sound and the LED will flash each time a count is registered.
2. To preserve pen life, cap the pens when they are not in use.
3. Operating Temperature: 5˚C to 40˚C @ 50-80% RH.
4. Storage Temperature: -10˚C to 40˚C @ 80% RH max.
