1 Notes on this operating manual
Content The operating manual describes the following device model:
¡BiometraTMS
Within this document, this model will be referred to as the device or the temperature
measuring system.
The device is used to calibrate automated and manually operated thermal cyclers.
The device is intended to be operated by qualified specialist personnel who must ob-
serve all instructions given in the operating manual.
The operating manual provides information about the design and operation of the de-
vice and provides operating personnel with the necessary know-how for safe handling of
the device and its components. Furthermore, the operating manual includes information
on the maintenance and servicing of the device as well as information on potential
causes of malfunctions and their correction.
The device has a modular design. The multimeter has its own separate operating man-
ual.
Conventions Instructions for actions occurring in chronological order are numbered and combined
into action units.
Warnings are indicated by a warning triangle and a signal word. The type, source and
consequences of the hazard are stated together with notes on preventing the hazard.
Elements of the control and analysis program are indicated as follows:
¡Program terms are in bold (e.g., the System menu).
¡Menu items are separated by vertical lines (e.g., System|Device).
Symbols and signal words used
in this manual
The user manual uses the following symbols and signal words to indicate hazards or in-
structions. These warnings are always placed before an action.
WARNING
Indicates a potentially hazardous situation which can cause death or very serious (possi-
bly permanent) injury.
CAUTION
Indicates a potentially hazardous situation which can cause slight or minor injuries.
NOTICE
Provides information on potential material or environmental damage.

2 Intended use
The BiometraTMS is a temperature measuring system for calibrating thermal cy-
clers. The measured temperatures of the sample block under test are compared with
specific predefined values.
NOTICE!The temperature measuring system cannot be used for adjusting the de-
vice. The calibration can only be used to determine and document possible devia-
tions. Adjustment must be carried out separately.
If at the end of the calibration process the control software reports that the thermal
cycler has failed the test, you should contact the Service department to arrange the
temperature readjustment of the device.
Regular calibration of the sample block temperature ensures compliance with the
temperature specifications and thus reliable test results.
The temperature measuring system has a modular design. A simple plug-in connec-
tor system ensures easy replacement of the measuring tree and allows testing ther-
mal cyclers with sample blocks of different sizes. Suitable measuring trees for the
various sample blocks can be obtained via the company Analytik Jena.
The measuring trees that are available allow calibrating automated and manually
operated thermal cyclers.
BiometraTMS can also be used to calibrate the heated lid. This calibration process
can be carried out in addition to the sample block calibration.
The software provides a number of configuration and display options. The intuitive
and easy-to-use structure of the software provides a convenient way to navigate
through the individual steps of the calibration. Information boxes and tool tips make
it easier to understand the process.
The device and its components may only be used for the analyses listed in the user
manual. Only this specified use is regarded as the intended use, ensuring the safety
of the user and the device.
The device must only be operated by specialist personnel trained by Analytik Jena.

3 Security
For your own safety and to ensure error-free and safe operation of the device, please
read this chapter carefully before commissioning.
Observe all safety instructions listed in this user manual and all messages and infor-
mation displayed on the monitor by the control and analysis software.
3.1 Safety labeling on the device
Warning and mandatory action labels have been attached to the device and must al-
ways be observed.
Damaged or missing warning and mandatory action labels can cause incorrect ac-
tions leading to personal injury or material damage. The labels must not be removed.
Damaged warning and mandatory action labels must be replaced immediately!
Also refer to the information provided in the operating manual of the multimeter for
details on the warning and mandatory action labels.
3.2 Requirements for the operating personnel
The device must only be commissioned, operated and serviced by qualified specialist
personnel who have been instructed by Analytik Jena. This instruction also includes
imparting the contents of this user manual and the user manuals of the connected
system components.
In addition to the safety instructions in the operating manual, the general applicable
safety and accident prevention regulations of the respective country the device is op-
erated in must be observed and adhered to. The operator must verify that they are
referring to the latest version of these regulations.
The operating manual must be accessible to the operating and service personnel.
3.3 Safety instructions, transport and commissioning
To prevent health damage, the following must be observed when moving the device
in the laboratory (lifting and carrying):
¡The device has no carrying handles. To transport the device, grip the device
firmly at the bottom with both hands and lift it.
¡Switch off the device before transporting it.
¡Insufficiently secured components pose a risk of injury. During transport, secure
the device components as specified in this operating manual.
¡Loose parts must be removed from the system components and packed sepa-
rately.
¡Risk of damage to health due to improper decontamination! It is not possible to
disinfect the temperature measuring system itself. Only use the temperature
measuring system in thermal cyclers that have been cleaned, disinfected and
dried beforehand! Before returning the device to Analytik Jena, clean the device
with a damp cloth, allow it to dry and document the cleaning measures taken.

3.4 Safety instructions: during operation
3.4.1 General safety instructions during operation
The operator must make sure that the device and its safety equipment is in sound
condition each time before starting up the device. This applies in particular after
each modification or extension of the device or its repair.
Observe the following:
¡The device may only be operated if all items of protective equipment (e.g. covers
in front of electronic components) are in place, properly installed and fully oper-
ational.
¡The sound condition of the protection and safety equipment must be checked
regularly. Any defects must be corrected as soon as they occur.
¡Protective and safety equipment must never be removed, modified or switched
off during operation.
¡The ventilation equipment on the device must be in good working condition.
Covered ventilation grilles or slots etc. may cause the device to break down or
may cause damage to it.
¡Ensure that no liquid enters the interior of the device, for example at cable con-
nections. There is a danger of electric shock.
¡Keep all combustible materials away from the device.
3.4.2 Safety instructions – protection against explosion and fire
The device may not be operated in an explosive environment.
Smoking or handling open flames are prohibited in the room in which the device is
operated!
3.4.3 Safety instructions: electrical equipment
Lethal voltages may occur in the device! Contact with live components may cause
death, serious injury or painful electrical shock.
¡Please observe the information on the electronics provided in the multimeter's
operating manual!
¡The measuring tree must only be plugged in or unplugged from the multimeter
after the multimeter has been switched off using the power button at the front
of the device.
¡Before starting any maintenance and repair work the device must be switched
off using the power button on the front of the multimeter and the power plug
must be removed from the power outlet!
¡Work on the electronics and in the interior of the device may only be carried out
by the customer service of Analytik Jena and specially authorized technicians.
3.4.4 Handling of auxiliary and operating materials and samples
The operator is responsible for the selection of substances used in the process as well
as for their safe handling. This is particularly important for radioactive, infectious,
poisonous, corrosive, combustible, explosive and otherwise dangerous substances.
When handling hazardous substances, the locally applicable safety instructions and
instructions in the safety data sheets from the manufacturers of the auxiliary and op-
erating materials must be complied with.

¡Avoid biological contamination of the device. The housing of the measuring
head cannot be disinfected and can only be wiped with a damp cloth. Clean, dis-
infect and dry the thermal cycler to be tested, and especially the sample block,
before inserting the measuring head.
¡The operator is responsible for carrying out suitable decontamination should the
device itself be contaminated with dangerous substances.
¡Splashes, drops or larger liquid spillages should be removed using an absorbent
material such as absorbent cotton, laboratory wipes or cellulose.
¡The tubes on the measuring tree must not come into contact with substances
containing oil. The operator must ensure the absence of all substances contain-
ing oil on the surface of the block and in the wells of the thermal cycler that is
supposed to be measured.
3.4.5 Safety instructions – maintenance and repair
The device is generally maintained by the customer service department of Analytik
Jena or specialist personnel trained and authorized by them.
Unauthorized maintenance can damage the device. For this reason, only the activi-
ties described in the user manual in the "Maintenance and care" chapter may be per-
formed by the operator.
¡All maintenance and repair work on the device must only be carried out when
the device is switched off (unless specified otherwise).
¡Only clean the exterior of the device with a slightly moistened, non-dripping
cloth. Use only water and, if required, customary surfactants.
¡The only suitable cleaning method for the multimeter housing is cleaning by
wiping. If the cleaning agent used has a spray nozzle, spray some of the cleaning
agent onto a suitable piece of cloth before using it on the device.
¡Do not use organic solvents or abrasives to clean the device.
¡It is not possible to disinfect the temperature measuring system itself. Only use
the temperature measuring system in thermal cyclers that have been cleaned,
disinfected and dried beforehand!
¡All protective equipment must be reinstalled and checked for proper function
when the maintenance or repair work is complete.
¡Use only original spare parts, wear parts and consumables. These have been
tested and ensure safe operation.
3.5 Behavior during emergencies
If there is no immediate risk of injury, switch off the device and the connected system
components immediately in hazardous situations or in the event of an accident and/
or disconnect the power plugs from the power outlets.

4 Design and function
Calibration of a thermal cycler using a BiometraTMS is carried out as follows:
The temperature measuring system measures the temperatures of the sample block
and, if required, of the heated lid of the tested thermal cycler. The software of the tem-
perature measuring system which is installed on a separate PC logs the captured data,
compares them with a predefined temperature program, detects any deviations, and dis-
plays them.
Before the measurement, the predefined temperature program is set on the thermal cy-
cler. Specifying a temperature program allows comparing several calibrations of the
same or of different thermal cyclers.
The software can display the temperatures that were determined during the measure-
ment in various different ways. Apart from displaying the current temperatures of the
individual sample block positions, the software can also display the progression of the
temperature in form of diagram.
The temperatures in the heated lid are measured directly after measuring the sample
block.
After completing the measurements in the sample block and the heated lid, the operator
can use the software to produce a report, save this report and print it out using an exter-
nal printer.
The temperature measuring system consists of the following components shown in the
figure:

4.1 Multimeter
The measurement requires a Keithley multimeter configured and customized especially
for Analytik Jena.
The control elements of the multimeter can be found on the front of the device. The
functions described in this operating manual only require two of these control elements:
¡Power button: For switching the multimeter on and off
¡Terminal button: For toggling the setting of the active terminal between front and
rear. Activating the terminal on the rear for controlling the multiplexer module used
there for calibration
All further operation of the device is executed via the software.
NOTICE!Only use the mentioned elements for operating the multimeter. Operating
other elements may cause the device to malfunction.

The rear of the multimeter houses the sockets, the multiplexer module and the fuse
compartment. The sockets are used to establish the power connection and the connec-
tion between the measuring tree and the PC.

The fuse compartment on the power module can be used to adapt the operating voltage
to the mains voltage. Please observe the relevant information in the multimeter’s oper-
ating manual for setting the operating voltage.

NOTICE!The multimeter requires a warm-up time of at least 30minutes after
switching the device on to allow the temperature inside the device to stabilize. If the de-
vice has been exposed to extreme temperatures, the device requires additional time to
stabilize. Operating the multimeter outside its operating temperature range of
23°C±5°C results in an increased measurement uncertainty.
4.2 Measuring tree
Design The measuring tree consists of a measuring head and a connecting cable. The connect-
ing cable has a D-SUB connector to connect the adapter to a multimeter. The bottom
part of the measuring head contains tubes which are loosely installed in the base plate
of the measuring head. The clearance created by this allows inserting the measuring
head precisely into the sample block. The sensors are located inside the tubes. All mea-
suring trees have a total of 16sensors.

The arrangement of these sensors on the various measuring trees matches the tempera-
ture characteristics of the block, i.e. the mean value, spread and uniformity. Depending
on the block type and manufacturer, these can be distributed differently across the
block. Using the Biometra TMS system to calibrate Analytik Jena thermal cyclers guaran-
tees that the calibration is carried out in accordance with the factory settings.
The following figures show the Biometra TMS MT 96 200µl by way of example.
NOTICE!Only put down the measuring head on its top surface. The measuring head
must not be put down on its sensitive sensors.

The device designation and the serial number are indicated at the top of the measuring
head. In addition to that, a measuring head is equipped with a sensor in its upper part
for testing the heated lid.

Measuring tree variants Analytik Jena offers measuring trees with matching measuring heads for calibrating dif-
ferent types of sample blocks. These measuring trees can be connected to the multime-
ter using a simple plug-in system. The software will automatically read the serial num-
ber of the measuring tree that is connected. Measuring tree specifications are retrieved
from the measuring tree’s configuration file on the USB flash drive.
The measuring tree models available either have a cable harness on the side or at the
front.
The following table lists measuring trees with cable harness on the side as well as the

Measuring trees with cable harness at the front are suitable for thermal cyclers whose
lid closing mechanisms prevent lateral cable routing.

4.3 Software
Functional scope of the software:

Licensing The customer is provided with a single-user license protected by a USB dongle with a li-
cense key.
Refer to the file LICENSE.txt to see which open-source licenses are used in this product.
This file is stored with the software on the USB flash drive.

5 Installation and commissioning
5.1 Installation prerequisites
Climatic conditions The requirements for the climatic conditions at the installation location are set out in
the specifications. If required, ensure that the room is temperature-controlled.
Installation location require-
ments
¡This laboratory device is designed for inside use.
¡Do not use the device in wet and damp environments. Keep the device surface clean
and dry.
¡Avoid direct sunlight and radiation from heaters onto the device. If necessary, pro-
vide air conditioning.
¡Do not locate the device near sources of electromagnetic interference.
¡Avoid mechanical shocks and vibrations.
¡Do not use the device in explosion-hazard environments.
¡Place the device on a stable surface.
¡The installation site must be free of drafts, dust and caustic fumes.
¡Keep the ventilation slits free and do not obstruct them with other devices.
5.1.1 Spatial requirements
The spatial requirements of the multimeter are: 22.4×10.7×38.7cm . Additional
space is required next to the multimeter to place a PC.
Multimeter and PC can also be placed on separate tables, provided the cable of the mea-
suring tree and/or the USB cable are long enough.
5.1.2 Power supply
WARNING
Danger from electric current
¡Only connect the device to a properly grounded mains outlet.
Before connecting the device to the mains, verify that the operating voltage displayed
on the power module on the back of the multimeter matches the local mains voltage. If
this is not the case, set the correct operating voltage by adjusting the fuse to the local
mains voltage. Please observe the relevant information in the multimeter’s operating
manual for setting the mains voltage
The measuring tree must only be plugged in or unplugged from the multimeter after the
multimeter has been switched off using the power button at the front of the device.
Please observe the relevant information on the electrical connection conditions in the
multimeter manual!

5.2 Installation
WARNING
Danger from electric current
¡Only connect the device to a properly grounded mains outlet.
¡When installing the device, refer to the respective instructions on the electrical con-
nection conditions and the correct setting of the operating voltage in the multimeter
manual.
NOTICE
Keep the original packaging
Transport damage can only be avoided if the device is transported in its original packag-
ing.
¡Keep the original packaging for transport, e.g., in case the device must be returned
to the manufacturer for repair.
NOTICE
Also observe the information provided in the operating manual of the multimeter!
Proceed as follows for installing the device:
}Remove the multimeter and the connecting cable from the transport
case. Wait until the device has reached room temperature before putting
it into service.
}Remove the measuring tree from the transport lock inside the transport
case.
NOTICE!Only put down the measuring head on its top surface. The mea-
suring head must not be put down on its sensitive sensors.
}Verify that the delivery is complete. Check all components of the device
for transport damage.
If the delivery is incomplete or in case of transport damage, contact Ana-
lytik Jena.

}Check which operating voltage is set on the multimeter(1). If required,
set the correct operating voltage via the slot in the fuse compartment.
When doing this, observe the information provided in the operating
manual of the multimeter!
}Plug the provided power cord into the multimeter(2). Insert the plug into
the mains socket.
When connected to the mains power, the state of the multimeter is the
same (soft power-on/soft power-off) as it was when the device was last
disconnected from the power supply.

}When the multimeter is switched on: Use the power button at the front
of the device to switch the multimeter off (3, soft power-off).

}Use the D-SUB connector to connect the measuring tree to the multime-
ter(4).
}Switch on the PC.
}Insert the USB flash drive with the software provided with the device into
the PC. Launch the installer file. The installer file installs the software
and all drivers for the device on the PC.
}Insert the provided USB dongle with the license key into the PC.
}Use the shortcut in the Start menu or on the PC’s desktop to launch the
software.

}Switch on the multimeter. To turn the device on, press the power button
on the front of the multimeter(5).
NOTICE!The multimeter requires a warm-up time of at least 30minutes
to achieve its nominal accuracy.
}If the terminal on the front of the multimeter is active when the device is
started up (green status LED for the terminal “Front”): Press the terminal
button(6) to activate the multiplexer module on the rear of the multime-
ter.
üThe status LED for the terminal on the “Rear” lights up in orange
color.

}Use the USB cable to connect the multimeter to the PC. The USB port is
located on the back of the multimeter(7).
}Open the software to set up the interface to the PC.
– To set up the interface, click the button Settings and checks on the
start screen.
– Click the button Interface multimeter on the tab Settings.
– On the page Connection settings, click the button Search multime-
ter.
– If required: Use Test connection to test the interface.
– Click Continue to save the settings.
üThe device is ready for operation.
When a measuring tree is connected to the multimeter for the first time, the
software will prompt the user to load the configuration file for this measur-
ing tree when starting a calibration. The configuration file can be found on
the USB flash drive provided with the measuring tree. Observe all further in-
formation in the chapter “Operation”.

6 Operation
6.1 Switching the device on and off
Switching the device on ðThe multimeter is connected to the mains supply and the PC via the mains cable and
the USB cable.
ðThe measuring tree is connected to the multimeter.
ðThe software is installed on the PC.
}Use the power button on the housing front to turn the multimeter on.
}Boot the PC.
}Use the shortcut in the Start menu or on the PC’s desktop to launch the software.
üAfter a 30-minute warm-up time, the device is ready for operation.
Switching the device off }Exit the software by closing the window.
}Shut down the PC.

6.2 Overview of basic software functions
The software guides the user through the calibration process in simple steps. Various
configuration options ensure that the operator can customize the software according to
their needs and visualize results.
Starting the software Use the shortcut on the desktop or in the Start menu to launch the software.
Starting the software in an-
other language
The software is started in the language set for the operating system it was installed on.
If you want to start the software in a different language, you have the option to enter a
command line parameter in the software link. To do this, proceed as follows:
}Select the shortcut for the installed software on the desktop or in the Start menu.
}Open the shortcut menu and click Properties.
}Open the tab Shortcut.
}Add the following command to the specified link in the input field under Target :
–-language=de (German software interface)
–-language=en (English software interface)
üWhen using this link to launch the software the next time, it is started in the
specified language.
If the software is not available in the desired language, the system will automati-
cally start the English version of the software.

Exiting the software Click the button X in the top right corner of the window to exit the software. Clicking this
button will close the window and stop the execution of the software.
