Introduction

Operating Nexera CL System
nIntended Use
This system is designed to perform qualitative and quantitative analysis of target
compounds in a sample matrix and can be used as a liquid chromatograph for general
in vitro diagnostic applications. This instrument operates with reduced technician
involvement and automation of some, but not all, procedural steps. However, only
personnel who have received appropriate training on use of the system can use it for
these purposes.
nCalibration
A calibration curve should be generated for each analyte in appropriate methods. At
least four out of six non-zero standards should meet the appropriate criteria, including
the calibration standard lower than the assumed LOQ and the calibration standard at
the highest concentration.
nQuality Control
Implement quality control of the instrument by routinely measuring at least one each of
four types of quality-control samples (a sample with a normal concentration level, a
sample with a concentration level higher than normal as well as a sample lower than
normal, and a blank sample). Check the selectivity/specificity in the matrix used for the
actual sample, the accuracy, correctness, recovery rate, dynamic range, linearity, and lower
limit of quantitation.
Based on these assessment results, check that the results are within the permissible range.
If the assessment results are out of range, the measured data may be invalid, so do not
use analysis results obtained from the instrument in question until it can be confirmed
that it is functioning normally. When, for example, analyzing samples that contain
complex sample matrices such as serum, plasma or urine, it may be possible to obtain
stabilized data by using an appropriate pre-treatment or an internal standard.

1.1.1 LC-40D XR CL, LC-40D X3 CL, or LC-40B X3 CL Solvent Delivery Unit
Micro-plungers with a discharge volume of 10 µL per stroke enable solvent delivery with
minimal pulsation. Newly developed plungers and plunger seals provide a longer service
life for each pressure region than previous models.
Two gradient methods are available, depending on objectives. A high-pressure gradient
can be selected to minimize the gradient delay volume or a low-pressure gradient can
be selected for low-cost delivery of up to four solvents.
Furthermore, an optional solvent selection valve (built into the pump) or reservoir
selection valve can be used to switch between multiple mobile phases for each solvent
delivery unit. Using such a mobile phase selection system allows samples to be
continuously analyzed using different analytical systems or columns to be automatically
rinsed after analysis.

These autosamplers provide the low carryover required for high sensitivity analysis, high
reproducibility for trace quantity injection, and high expandability for satisfying a wide
variety of analytical needs.

The standard injection method involves no sample losses and is a total injection method
with high injection reproducibility.
Equipped with automatic pretreatment functionality for diluting samples online, adding
internal standard samples, or derivatizing samples online, these autosamplers help
increase throughput.
Functionality for rinsing the exterior surface of the needles is also included standard to
ensure low carryover levels. By using the rinsing functionality, the exterior surface of
needles can be rinsed with two types of rinse solutions and, using the multi-rinsing
functionality, the interior surface of needles can be rinsed with three types of rinse
solutions or the injection port can be rinsed.
Sample racks can hold three microplates, deep-well plates, 1.5 mL vial plates, or other
plates, for processing up to 1152 analytes (three MTP384 plates).
Furthermore, up to three optional plate changers can be connected to load up to 44
microplates. In combination with temperature-control functionality, that enables
continuous multianalyte analysis with high reliability.

1 Overview

1.1.3 CTO-40C CL Column Oven
These ovens use an air circulation method that provides high temperature control
accuracy, which improves analysis reproducibility and peak separation capability by
ensuring a constant temperature in columns and flow channels.
The CTO-40C CL can control temperature up to 100 ℃ include a cooling mechanism that
can cool the oven down to 10 ℃ below room temperature. The temperature gradient
within the column can be reduced to improve separation capabilities by adding an active
preheater (optional) that can control the temperature of individual mobile phases flowing
through the column.
1.1.4 SPD-40 CL UV-VIS Detector
The SPD-40 CL includes a D2 (deuterium) lamp and additional functionality that can be
used in a wide variety of situations, such as a simultaneous dual wavelength measurement
function and wavelength scanning function (for measuring absorbance spectra).
By controlling the flow cell temperature, the mobile phase and sample temperature can
be kept constant even if the room temperature varies, which enables stable analysis.
The detectors also include an internal cell ID memory that remembers the type and ID
number of cells and a lamp ID memory that remembers the illumination time and ID
number of lamps. Information about the cell and lamp used for analysis is automatically
recorded in analytical data to improve analysis traceability.

Optional Part P/N Features
System Anchoring Strap for
Earthquakes 228-54756-02
By strapping stacked components together,
this strap secures the system in the event
of an earthquake (4 m long, with buckle).
L-Type Retainer Plate for
Earthquakes 228-77035-41
This part is used to fasten the strapped
stack of system components to an
installation base stand. It is fastened
directly to the base stand with bolts.
System Base Kit A 228-77036-41
Systems are placed on this base stand.
Used in combination with a system
anchoring strap for earthquakes, this kit
can be used to secure the system to the
base stand. It also enables liquid waste to
be drained from the back of the system
(420 mm wide).
System Base Kit B 228-77036-42 Same as Kit A, except 290 mm wide
System Base Kit C 228-77036-43 Same as Kit A, except 170 mm wide for
plate changers
For IVDR systems (for Europe),MPM-40 cannot be used.
