1 Safety
1.1 Introduction
With the purchase of the Medite Tissue Stainer COT 20 you decided for a high
quality product of medite Medizintechnik GmbH. These instructions shall help you to
use the instrument. Please read it thoroughly and follow the advices.
If you see this symbol in the instruction manual, it indicates dangers
(including warnings)
1.2 Intended use
Unauthorized changes and technical modifications of the instruments are not
permitted for safety reasons. For the exchange of defective parts only original medite
spare parts have to be used.
The conditions for operation, maintenance and service mentioned in this instruction
manual have to be strictly observed.
1.3 Authorized operator
The COT 20 must be used by those persons only, who have been authorized by the
owner. In his working area, the operator is responsible with regard to third persons.
The owner must give the operator access to the instruction manual and make sure
that the operator has read and understood its contents.
1.4 Safety on site
The COT 20 must be placed on an even and solid basis. An overturning instrument
means an accident risk.
Any failure to comply with the approved technical regulations for the
operation of technical equipment or any non-intended use of the
instrument will void the manufacturer’s liability.

2.2 Transport/ Storage
The medite Tissue Stainer COT 20 is delivered in a protecting box. Please move it
only upright and avoid shock.
2.3 Unpacking
Put the instrument on an even, solid surface and remove the protecting box and
plastic wrap. If possible, keep the packing material to avoid damages in case of a
later transport of the unit. Check whether the equipment is complete. The COT 20 is
supplied with one power cord and one instruction manual.
2.4. Installation and Connection
Before starting the instrument, please make sure that your mains voltage
corresponds to the value indicated on the instrument (example: for an instrument
requiring 240 Volts, 200 - 240 Volts should be available; for a 115 Volts instrument,
there should be 100 - 125 Volts.) The instrument is supplied ready to use. For
connection to mains the included power cord must be used. It must be connected to
a socket with intact ground wire.
Furthermore pay attention that no inflammable objects are in the heating
area of the instrument
3 Operation
Operating conditions:
The operation is only allowed in closed rooms
Laboratory bench free of vibrations and strong enough for the instrument’s
weight
Environmental temperature from + 15°C to + 45°C
(without large variation in temperature)
At maximum 80% relative humidity of air, not condensing
The instrument must not be exposed directly to the sun
Guaranteeing a sufficient air circulation for cooling, the instrument needs
enough space (minimum 15cm) to all sides

The medite COT 20 is a linear staining machine which because of its continuous
working principle can be adapted to the individual requirements of histology and
cytology laboratories.
The stainer has 23 troughs with a volume of 250 ml each. Corresponding to the
employed staining method, between station 2 and 17 running water can be used,
supplied by a central water connection. The instrument is constructed in such a way
that up to 4 slide holders can be placed in the start position. Additionally, two
separate removable solvent containers are integrated in the working plate, taking up
10 slide holders each. The slide holders are capable of 20 slides each.
The Tissue Stainer COT 20 is supplied with an instrument cover, reducing the fume
contamination and avoiding a pollution of the staining solvents. Additionally, there
are two covers made of stainless steel which can be put on the staining baths when
the instrument is out of use. The medite Tissue Stainer COT 20 is supplied with fume
extraction as standard equipment.
According to the staining time which can be regulated by means of a timer from 20
seconds to 10 minutes, the slide holders are transported and pass the baths. The set
time is always the same for every bath. Normally, a staining time of 2 minutes is
chosen. In order to keep the carryover of the solvents as low as possible, the COT
20 is equipped with a drain mechanism, adjustable at a timer from 3 to 60 seconds.
By using the agitation, i.e. the up and down movement of the slide holders in the
bath, the effectiveness of the solvents can be increased and the staining time may
be reduced. However, in order to avoid that sections of sensitive specimens detach
from the slides, it is recommended to work without agitation.

Put the transport bars „ A “ on the guide rolls „ B“ in such a way that the folded
edges are located to the inside of the instrument and that the parts with the holes
extend from the left side (Fig. 1).

Connect front and back bar by supplied spacer bolts and screws „D“ on the left
side

If necessary, align transport bars (the centers of the scores must be in the
centers of the staining troughs) and fix angle joint „ C“

Connect supplied tubes for water in- and outlet at the bottom (Fig. 2) of the
instrument
CAUTION: In order to avoid back pressure, outlet tube must be declined
towards the drain!

Connect instrument to mains by means of the supplied lead

Check whether mains voltage corresponds to the voltage indicated on the
instrument

The timer for staining works in the range from 12 seconds to 10 minutes, the timer
for the drain cycle in the range from 3 to 60 seconds. The desired times are set by
turning the control knob of the timer. Time range and unit are easily variable (see
enclosed diagram).
When the instrument is delivered, with factory setted staining time is preset to 2
minutes, draining time to 15 seconds. Please remember, that the time is the same
for every bath.

5 Operation
Particular attention must be paid to the arrangement of the staining troughs. They
must be placed exactly in sequence so that they connect to each other with no
space, even small, between them!
Water Supply
Having chosen the staining programme and having determined the stations requiring
water rinsing, the troughs with base outlets are connected to the water supply bar.
This is done by removing the screws on the bar at the required positions and by
inserting the small tube of the trough in the hole. Then, the staining troughs are
placed and filled with solvents according to the desired programme. Now, water is
turned on and water flow is regulated by the control valve.
CAUTION: In order to avoid back pressure, outlet tube must be declined
towards the drain!

Program Determination and Start
When the first slide holder is placed in start position, the stainer is switched on
(green switch „Power“). Staining is started with the 1st red switch „Stain“. In order to
increase the effectiveness of the solvents and to be able to reduce immersion time,
the slide holders can be moved slightly up and down in the bath. For this purpose,
the 2nd red switch „Agitate“ is pressed. However, to prevent that sections of sensitive
specimens detach from the slides, agitation is not used. The drain mechanism, used
to avoid carryover of the solvents from one bath to the next, is operated by the yellow
switch „Drain“.
Stain
The staining programme now runs completely automatically. When the first holder
has arrived at the end position, an alarm is sounding and the staining process is
stopped. In order to avoid an overstaining, the holders in process stay above the
baths and their transport is automatically continued when the ready slide holder has
been removed.
6 Cleaning and Maintenance
The slide stainer can be cleaned easily as it is completely made of powder coated
aluminium and stainless steel.
Staining troughs and slide holders:
Cleaning with usual detergents may prove unsatisfactory. Therefore, we recommend
to soak the troughs and holders for several hours in a mixture of 10 ml of
concentrated hydrochloric acid and 1 litre of 70% ethanol.
7 Putting out of operation
After end of the total period of use please deliver the equipment to a regular disposal
facility. The company medite as manufacturer is also prepared to take back the
equipment for disposal against payment for the disposal fee.

The following instructions have to be observed by all means:
1.
As a precaution, due to evaporation particularly in case of the use of Xylene, it is
recommended to place the stainer under a fume hood or to use the charcoal filter
supplied as an optional extra.
2.
Before starting the staining process, water flow must be regulated by means of
the valve at the water supply tube.
3.
The slide holders must be put exactly in the scores of the transport bar.
4.
The drain tube must be purged or changed, respectively, as it can be blocked by
residues.
5.
In case of a major spillage, especially if fluid could have been spilled inside the
instrument, switch off the stainer and pull out the mains plug immediately!
Before starting again, make sure that all moisture inside the instrument is
completely removed. If possible, the stainer should be checked by a competent
service engineer. We recommend to have the instrument repaired only by our
authorized service engineers.
6.
In case of failure, which f.ex. could be caused by voltage oscillations, please
check the fuses inserted in the socket at the instrument back.
If it is not possible to repair the failure by changing the fuses, please contact our
service department.
CAUTION: Before taking out the fuses, pull out the mains plug!
