3. Using a blood collection set and a tube holder, collect blood into the PAXgene Blood RNA Tube using your
institution’s recommended procedure for standard venipuncture technique. Thefollowingtechniquesshallbe
usedtopreventpossiblebackow:
a. Place donor’s arm in a downward position.
b. Hold tube in a vertical position, below the donor’s arm during blood collection.
c. Release tourniquet as soon as blood starts to ow into tube.
d. Make sure tube additives do not touch stopper or end of the needle during venipuncture.
4. Allowatleast10secondsforacompleteblooddrawtotakeplace.Ensurethatthebloodhasstoppedowing
intothetubebeforeremovingthetubefromtheholder.ThePAXgeneBloodRNATubewithitsvacuumis
designedtodraw2.5mlofbloodintothetube.
5. Immediately after blood collection, gently invert the PAXgene Blood RNA Tube 8–10 times.
6. Store the PAXgene Blood RNA Tube upright at room temperature (18−25˚C) for a minimum of 2 hours and a
maximum of 72 hours before processing or transferring to refrigerator (2−8˚C) or freezer (−20˚C). See Procedure
for Freezing and Thawing Specimens in the PAXgene Blood RNA Tubes for details if storage temperature of
−70˚C / −80˚C is desired.
C. ProcedureforFreezingandThawingSpecimensCollectedinthePAXgeneBloodRNATubes
1. Stand the PAXgene Blood RNA Tube upright in a wire rack. DonotfreezetubesuprightinaStyrofoam™trayas
thismaycausethetubestocrack.
2. The PAXgene Blood RNA Tubes can be stored at −20˚C and below. If tubes are to be kept at temperatures below
−20˚C, freeze them rst at −20˚C for 24 hours, then transfer them to −70˚C or −80˚C.
3. Thaw the PAXgene Blood RNA Tubes in a wire rack at ambient temperature (18–25˚C) for approximately two
hours. DonotthawthePAXgeneBloodRNATubesattemperaturesabove25˚C.
4. Carefully invert the thawed PAXgene Blood RNA Tubes 10 times.
Note:If the two-hour ambient temperature incubation was not carried out prior to freezing, incubate tubes at least
another two hours after reaching room temperature before processing.
Note: The frozen PAXgene Blood RNA Tubes are subject to breakage upon impact. To reduce the risk of breakage
during shipment, frozen tubes should be treated in the same manner as glass tubes. Users must validate their own
freezing and shipping protocol for the PAXgene Blood RNA Tubes.
D. ProcedureforSpecimenPreparationforAnalysis
Blood samples must be processed according to the instructions provided with the PAXgene Blood RNA Kit.

V. Limitations
1. Under-llingofthePAXgeneBloodRNATubeswillresultinanincorrectblood-to-additiveratioandmayleadto
incorrectanalyticresultsorpoorproductperformance.
2. The PAXgene Blood RNA System is not suitable for the collection and purication of viral RNA.
3. Blood samples obtained with the PAXgene Blood RNA Tube must be prepared only with the PAXgene Blood RNA

4. The quantity of blood drawn should be 2.5 ml per PAXgene Blood RNA Tube, but this volume might vary with
altitude, ambient temperature, barometric pressure, tube age, venous pressure, and lling technique.
5. The PAXgene Blood RNA Kit is intended for purication of intracellular RNA from human whole blood with
leukocytes counts between 4.8–11.0 × 106 leukocytes/ml.
6. Endotoxin not controlled. Blood and blood components collected and processed in the tube are not intended for
infusion or introduction into the human body.
