Select the image tab and check the
live box

2Position plate Insert the plate holder, ensuring the
plate holder which gives the best
contrast between the colony and
background is used
Place plate onto plate holder
Adjust the exposure by increasing/
decreasing the sliding scale

5Plate configuration
Select Plate Configuration to set
plate size, sample volume and
threshold. Defaults are Circular
plate 90mm, Spiral volume 100µl
and threshold of 20. Change if
necessary

colonies to be counted

Protos 3 is now ready to make the first measurement

Select colony type from the dropdown,
either Light or Dark Colonies

Move slider to the left to accept small
particles or to the right to exclude
small particles
If any colonies are touching select the
Split function

detection sensitivity
10 Enter a plate ID

tab. Autoincrement default is “on”, to
change this click on the Configuration
