1 Introduction
1.1 Use of the SPECORD PLUS
The spectrophotometers of the SPECORD PLUS family have been designed for measuring
the transmission, extinction, reflectance and energy of liquid and solid samples. They per-
form measurements in the spectral range of 190 to 1100 nm.
The individual devices have different optical properties/concepts and implement different
spectral resolutions:
SPECORD 50 PLUS
1.4 nm, split beam with internal reference channel
SPECORD 200 PLUS
1.4 nm, double-beam spectrophotometer
SPECORD 210 PLUS
0.5; 1; 2 and 4 nm, double-beam spectrophotometer
SPECORD 250 PLUS 0.5; 1; 2 and 4 nm, double-beam spectrophotometer with up-
stream monochromator
The optical system of the SPECORD PLUS without electromechanical modulator guarantees
a true realtime operation allowing even for the compensation of changing absorptions of the
reference samples during the measurement.
An internal wavelength calibration with holmium oxide filter ensures a very good wavelength
accuracy and reproducibility.
The SPECORD PLUS meets the regulations of DAB (German pharmacopoeia) as well as
international regulations, such as PhEur., BP, USP or JPXIII.
Together with the powerful software WinASPECT PLUS the SPECORD PLUS ist especially
suited for use in routine labs with high sample throughput, but with its versatile accessories
also provides solutions for special applications.
1.2 Notes on the manual
This manual contains the description of the SPECORD PLUS product family
SPECORD 50 PLUS
SPECORD 200 PLUS
SPECORD 210 PLUS
SPECORD 250 PLUS
If the descriptions relate equally to all three device types, only the general term SPECORD
PLUS is used. If the properties of an individual device type are to be singled out, the full de-
vice names are mentioned.
Instructions for action which occur in chronological order are numbered and combined into
action units and furnished with the corresponding results.
Lists which are not in chronological order are shown as itemized lists, sub-listings as bullet
points.

Safety notes are indicated by pictogaphs and signal words. The type and source of the
danger are stated together with notes on preventing the danger. The meaning of the picto-
graphs and signal words used is explained in the chapter "Safety notes".
The elements of the control and analysis software are indicated as follows:
Program terms are identified with SMALL CAPS.
Buttons are shown by square brackets (e.g. [OK] button)
Menu items are divided by an upright line (e.g. FILE |OPEN).
1.3 Warranty and liability
The warranty duration and liability comply with the legal requirements and the provisions in
the general terms and conditions of Analytik Jena AG.
Deviations from the intended use described in this user manual result in limitations of war-
ranty and liability during a damage event. Damage to wearing parts and glass breakage are
not included in the warranty.
Warranty and liability claims are excluded for personal injury and property damage due to
one or several of the following causes:
use of the SPECORD PLUS other than intended
improper commissioning, operation and service of the device
modifications of the equipment without prior consultation with Analytik Jena AG
unauthorized intervention in the equipment
operation of the device with faulty safety equipment
use of other than original spare parts, wearing parts or consumables
improper repairs
faults due to the non-observance of this user manual

2 Safety instructions
2.1 General notes
For your own safety and for a trouble-free operation carefully read this chapter before com-
missioning the SPECORD PLUS.
Comply with all safety instructions in the manual and pay careful attention to all messages
and notes which are displayed on the screen by the control software.
Also observe the safety instructions for system components by other manufacturers supplied
with this device (e.g. PC, printer, autosampler). The safety instructions on the labels and the
information on handling, storage and disposal of the reagents, reaction cells and packing
labels of the test kits must also be particularly observed.
Besides the safety instructions in this user manual and the local safety regulations that apply
to the operation of the device the general applicable regulations regarding accident preven-
tion, occupational health and safety and environmental protection have to be observed and
complied with.
References to potential dangers do not replace the work protection regulations which must
be observed.
2.2 Symbols and signal words used in the user manual
The user manual uses the following symbols and signal words to indicate hazards or instruc-
tions. The safety instructions are always placed before an action.
WARNING
Indicates a potentially hazardous situation.
If it is not prevented death or most serious injuries (incapacitation) can result.
CAUTION
Indicates a potentially hazardous situation.
If it is not prevented light or minor injuries and material damage can result.
CAUTION
Indicates a potentially hazardous situation.
Unless avoided, the product or items in its vicinity may be damaged.
IMPORTANT
Indicates application hints and other especially useful information without any resulting haz-
ardous or damaging situations.

CAUTION! UV RADIATION!
Do not look directly or indirectly via a mirror into the beams of the UV lamp. The
lamp radiation can cause conjunctivitis!
CAUTION! HOT SURFACE!
The lamp covers heat up strongly when the lamps are switched on. Switch off
the lamps and allow them to cool down sufficiently before replacing the lamps or
opening the lamp covers.

2.4 Requirements for the operating personnel
The SPECORD PLUS must only be operated by qualified specialist personnel instructed in
the use of the device. The instruction must also include conveying the content of this user
manual and the user manuals of the accessories and the program WinASPECT PLUS.
In addition to the safety at work instructions in this user manual the generally applicable
safety and accident prevention regulations of the respective country of operation must be
observed and adhered to. The operator must ascertain the latest version of these regulations.
The user manual must be accessible to the operating and service personnel at any time!
Observe the following notes:
The device must only be commissioned, operated and serviced by trained person-
nel instructed in technical safety.
The operation or servicing of the device by minors or individuals under the influ-
ence of alcohol, drugs or medication is not permitted.
The operating personnel must be familiar with the dangers arising from samples to
be analyzed and excipients used. The appropriate protective equipment must be
used.
2.5 Safety instructions, transport and installation
Observe the following notes:
Only transport the device in its original packaging! Ensure that the transport protec-
tions have been fitted and the sample compartment is completely empty.
To prevent health damage the following must be observed when moving the device
in the laboratory (lifting and carrying):
– For reasons of safety 2 persons are required to transport the device and must
position themselves on both sides of the equipment.
– Because the device does not have carrying handles, grip the device firmly with
both hands at the lower end.
– The guide values and statutory limits for lifting and carrying loads without aux-
iliary equipment must be observed and adhered to.

2.6 Safety instructions - operation
2.6.1 General
The operator of the SPECORD PLUS must make sure before each commissioning that the
condition of the device is sound. This applies in particular after each modification or exten-
sion of the device or its repair.
Observe the following notes:
Free access to the device switch on the enclosure wall must be ensured during
operation.
Caution when handling glass components (cells or similar). Risk of broken glass
and therefore risk of injury!
Never look directly or indirectly, e.g. through a mirror, into the lamp beam! The de-
vice must therefore be switched off to replace the lamps! There is a risk of conjunc-
tivitis.
The ventilation equipment on the device must be in good working condition. Cov-
ered vents or ventilation slits etc. may cause the device to break down or may
cause damage to it.
Ensure that no liquid can enter the SPECORD PLUS. The device could be dam-
aged.
Do not place the SPECORD PLUS in the immediate vicinity of aggressive vapor,
e.g. strongly corrosive acid, lye or solvents! The vapor can corrode the terminals,
mechanical and optical components of the device.
Avoid contamination of the sample chamber with strongly corrosive substances or
highly volatile solvents! Use vapor-proof cells with sealing plug in this case. Do not
leave the samples longer than necessary in the sample chamber of the device after
the measurement!
2.6.2 Safety instructions - Protection against explosion and fire
The SPECORD PLUS may not be operated in an explosive environment.
The operating personnel must be familiar with the location of the fire-fighting equipment in
the operating room of the device.
2.6.3 Safety instructions - electrical equipment
Work on electrical components of the SPECORD may only be carried out by a qualified elec-
trician in accordance with the applicable electrical engineering rules. Life-threatening electri-
cal voltages occur at various locations within the system!

Observe the following notes:
The SPECORD PLUS and its system components (PC) must always be switched
off before being connected to the mains.
The mains plug must only be inserted into a shock-proof socket to guarantee pro-
tection class I (protective conductor connection) for the device. The protective ef-
fect must not be invalidated by the use of an extension line which does not have a
protective conductor.
Before connecting the SPECORD PLUS to the mains check that the operating
voltage specified on the power rating plate at the rear of the device matches the
mains voltage at the intended socket. Operation with a voltage other than the spec-
ified operating voltage can destroy the device.
Before opening the device it must be switched off from the device switch and the
mains connector must be disconnected from the mains outlet!
Any work on the electronics may only be carried out by the customer service of
Analytik Jena AG and specially authorized technicians.
The device must be switched off immediately at the device switch (on the enclo-
sure wall) and the power supply disconnected from the mains if there is any inter-
ference with the electric components.
2.6.4 Handling of samples and reagents
The operator is responsible for the selection of substances used in the process as well as for
their safe handling. This is particularly important for radioactive, infectious, poisonous, corro-
sive, combustible, explosive and otherwise dangerous substances.
When handling dangerous substances local safety codes and guidelines must be observed.
The following general notes do not replace the specific local regulations or the regulations in
the EC safety data sheets of the manufacturers for the auxiliary and operating materials.
Observe the following notes:
The operator is responsible for carrying out suitable decontamination should the
device be contaminated externally or internally with dangerous substances.
Spots, drops or larger spillages should be removed and cleaned using an absor-
bent material such as cotton wool, laboratory wiping cloths or cellulose. The affect-
ed areas are then to be wiped with an Incidin Plus solution. Before using a cleaning
or decontamination procedure other than that prescribed by the manufacturer, the
user shall check with the manufacturer that the intended procedure will not damage
the device.

2.7 Safety instructions - service and repair
The SPECORD PLUS is usually serviced by the customer service department of Analytik
Jena AG or its authorized and trained specialist personnel.
Independent servicing can maladjust or damage the device. Therefore, the operator may
only carry out the tasks listed in the chapter "Service and care".
Observe the following notes:
The exterior of the SPECORD PLUS may only be cleaned with a damp, not drip-
ping, cloth after the device has been switched off.
Any service and repair work at the device may usually only be carried out in the
switched-off condition (unless stated otherwise).
Service tasks and the replacement of system components (e.g. lamps) must only
be carried out after a sufficiently long cooling down phase.
Only use original accessories and original replacement parts from Analytik Jena
AG. The notes in chapter "Service, maintenance, lamp replacement" p. 36 must be
observed.
Do not touch the glass body of halogen and deuterium lamps. Contamination of the
glass body, in particular of the deuterium lamp's beam outlet, will damage the lamp
properties.
Remove the protective cap from the halogen lamp after insertion! There is a risk of
fire.
2.8 Behavior during emergencies
If there is no immediate risk of injury and if this is possible immediately switch the
device switch of the SPECORD PLUS and the connected system components (PC,
accessories) in the "0" position during dangerous situations or in case of accidents
and disconnect the mains plugs from the mains outlets!
Note: For the PC there is a risk of data loss and damage to the operating system!

The SPECORD PLUS is built and tested according to standards that meet the requirements
of EC directives 2006/95/EC and 2004/108/EC. The device leaves the factory in a sound
condition as far as technical safety is concerned. To maintain this condition and to ensure
safe operation, the operator must strictly observe the safety and operating instructions con-
tained in this manual. For accessories which have also been supplied, and system compo-
nents from other manufacturers, their operating instructions should be referred to.

Device description and principle of operation

4 Device description and principle of operation
The SPECORD PLUS mainly consists of the modules:
Radiation sources
The halogen lamp and deuterium lamp generate the light radiating
through the sample after monochromatization.
Upstream monochroma-
tor
(only
SPECORD 250 PLUS)
The upstream monochromator is a holographic grid moving syn-
chronous with the monochromator grid. By first dispersing the light
the stray light is reduced.
Spectrometer system
The spectrometer system includes the monochromator which
disperses the light of the radiation sources. It is protected against
extraneous light and dust by its cover.
Photometer chamber
This is used to divide the light emitted from the monochromator
into a measuring and a comparison channel. The optics generate
the beam cross section in the sample chamber. The photometer
chamber is also protected by the cover.
Sample chamber
SPECORD 200/210/250
The samples are measured in the sample chamber. The sample
chamber has two sample chamber rods to accept various acces-
sory units. The two brackets for the cell adapters are mounted to
the adapter plate on the right sample chamber wall.
Sample chamber
SPECORD 50 Only the measuring beam path is directed through the sample
chamber. The accessories are mounted on the sample chamber
rods. The two brackets for the cell adapters are mounted to the
adapter plate on the right sample chamber wall.
Receiver circuit board
The radiation receivers are two photo diodes which are tempered
by a Peltier element in the SPECORD 210/250 PLUS.
Power supply modules
The power supply modules provide the electrical voltage for the
electronics and the fans for the thermoelectric tempering of the
receivers in the SPECORDPLUS.
Device controller
The device controller coordinates the modules involved in the
measuring process.
