I. Introduction
Thank you for choosing Opto-Edu A33.1502 Biological Microscope. To ensure safety and obtain optimum performance
and familiarize yourself with the use of this microscope, we recommend that you read this manual thoroughly before
operating the microscope. Retain this instruction manual in an easily accessible place near the microscope for further
reference.
II.Caution
Attention
A33.1502 Biological Microscope can only be used inside the room. Place microscope on a stable plane and keep it in
balance. Power line must be retained in an easily accessible place to prevent any unnecessary accident.
Working Environment
As microscope is a precision instrument, improper using will make it unworkable or reduce its precision.
1. Do not expose the microscope in the sun directly
2. Temperature range is +10℃- +40℃，and the max. humidity is 85%
3. Avoid high temperature and humidification. Otherwise there will be fog or mold on the lens.
4. Avoid violent vibration as the vibration will reduce the images quality.
5. Do not place microscope in moist room to avoid short circuit. Please turn off the power supply as soon as water drop
in the microscope. If there are other things unsafe come into the microscope which may cause short circuit too, please
stop using and contact with manufacturer

V. Preparation Before Use
Assemble the Binocular Head
1．Loosen the fixing screw on the top of the microscope body.
2．Put the binocular head on the body properly, then tighten the fixing screw.
Insert the Eyepiece
1．Standard gemel binocular head. Special-processed ring is fixed in the observation tube to tighten the eyepiece.
2．Insert the eyepiece properly.
Assemble the objectives
1．Assemble the objectives clockwise. Make the magnification become bigger when assembling in clockwise
direction.
2．Condenser has been mounted, focused & pre-centered well in the microscope before shipment. There is no need
to do any adjustment.
VI. Usage of the Microscope
Illumination
1．Connect the power line to the microscope.
2．Turn on the power supply, then the light will be on. Brightness can be adjusted by turning the adjusting knob.
Iris diaphragm is designed & configured on condenser. Adjusting the
iris diaphragm to match the different N.A of different objectives.
1．Open/Close the iris diaphragm by adjusting knob.
2．When the knob is on the right, that means the iris diaphragm is fully opened.
Placing the Specimen
1．Put the specimen in the stage and make the cover glass on it. Make the clips clamp them entirely. Two slides can
be put in the stage in the same time.
2．Make the specimen under the objective by adjusting X & Y axis.
Focusing
1．Turn the nosepiece to observe through the lowest magnification.
2．Adjust the stage to the top by turning the coarse focusing knob.
3．Observe through the eyepiece and turn the coarse focusing knob slowly to let the stage down and down till there
is a image
4．Turn the fine focusing knob to get sharp image.
Adjusting of the Binocular Observation Head
1．Adjust the observation tube to make sure that view field in the right & left eyepiece is consistent.
2．Adjust the diopter of the eyepiece to “0” first. Then adjust the eyepiece according to users’ own situation to get
sharp image.

Oil Immersion
1．When observing through 100X objective, oil immersion is required. Low down the stage to the bottom. Drop the oil
supplied by the manufacturer on the observation area.
2．Turn the 100X objective on its working position.
3．Raise the stage by turning the coarse focusing knob till the front of the objective can touch the oil.
4．Swing back and forth of the nosepiece to get rid of the air bubble in the oil. Then fix the objective. Make sure that
oil should be filled between objective and specimen.
VII. Maintenance of Microscope
When not in Use
1．Handle the microscope with two hands stably, and put the instrument on a stable plane. Cover it up with dust
cover. Store the instrument in a dry place.
2．It is highly recommended that objectives & eyepiece should be kept in a separate container.
Cleaning
1．Keep all the optics clean. It is very important to maintain optical performance of the instrument.
2．If dust, or dirt is covered on the surface of the lens. Have a try of air blower before cleaning the lens.
3．It is recommended that gauze, or lens tissue moistened with methyl alcohol should be used to clean the lens.
Ethyl alcohol is forbidden.
4．When cleaning the low magnification objective, lens tissue or cotton moistened with methyl alcohol can be used.
Pay much more attention when cleaning the high magnification objective. Dry the methyl alcohol first. Do not clean so
hard. Do not use ethyl alcohol.
5．When microscope body has to be taken apart, make sure that do not touch the lens of the objective, or else it will
reduce the image quality.
6．Periodical inspection
To maintain the performance of the microscope, periodical inspection is recommended.
Changing of Bulb
A33.1502 Series biological microscope is configured with LED illumination. There is no need to change bulb if there is no
certain situation.

1. Introduction
The APP installed in Android Pad A59.3520 can adjust image parameter, measure the objects, capture
image & video from the camera. It also provided simple image processing feature for particles analysis.
2. Camera User Interface
1. Preview Image –show live video of the camera.
2. Tool Panel –Control panels, capture, adjust parameter, measurement, etc.

3. Capture and Playback
Click “Snap” button to take a picture. Select picture size in the setting interface.
Click “Record” button to start video recording, click again to stop video recording.
If preview size is smaller than 1080p, the video size is same as preview size. If preview
size is bigger than 1080p, the video size will down to a size smaller than 1080p.
Show picture just captured. Play video file which just saved.
The picture will be opened in Image Analysis interface.

4. Adjust image parameter of camera
When the color of image is not very good, adjust the effect of image by
“Effect” panel.
Exposure:
Auto mode: Brightness of the image will be automatically
adjusted, you can also adjust the target brightness.
Manual mode: Manually adjust exposure time and gain.
White Balance:
Once AWB: Click icon to trigger once auto white balance.
Manual WB: Manually adjust color temperature.
Color Adjustment
Contrast, Saturation, sharpness and gamma of the image can be
adjusted too.
Click the reset button to restore the paramters.
5. Measurement
5.1 Calibration
We need calibrate the ruler before measurement, Specific combination of magnification of microscope
and the preview size of camera need specific calibration.
Swap the tool pane to measurement, click “Add” to add a new calibration, Click “Edit” to recalibrate the
exist calibration. 1.

Follow the Tips：
1. Drag the yellow ruler, let endpoints of the ruler close to physical ruler’s scale. We use the 0.01mm
physical ruler, each big grid is 10μm, we pick two grid, that’s 20μm.

2. Input the name of the calibration and the physical length of the ruler. We input 40x for the name, that
means the magnification of objective is 40X,
Then input the physical length of the ruler, that is 20μm.
3. Click “Calculate” to calculate the calibration value for current objective and preview size, and save to
list.

5.3 Measure with line ruler
Choose an calibration, the selected item will show with white background and black text.
Choose line ruler from right side bar. There will be an line ruler show on the image.

Use the line ruler to measure the physical ruler.
We measured 1.5 big grid, the line ruler show 14.910μm, that represent the result is right.
6. Image Analysis
Image is an application which used to process microscopic images on Android devices. It depends on
computer vision library Open CV. Currently, version 1.0 of Image provides functions like gray scale,
contrast, binarization and particles analysis.
6.1 Examples of particle analysis
6.1.1 Load image file
Click the camera icon to the upper right corner of the screen, you can choose a picture or take picture
from Camera application.

6.1.2 Convert the image into gray scale
Choose “Grayscale” function from left sidebar, click “Apply” button to confirm conversion.
6.1.3 Adjust contrast and brightness
Adjust contrast and brightness of the gray scale image, increase the difference between the objects and
background
6.1.4 Binarization
With the binarization tool, you can change the minimum and maximum value of threshold. The intensity
of all the pixels within the threshold range will be set to red. After apply binarization, the red area’s RGB
value will set to (0, 0, 0), the other area’s RGB value will set to (255, 255, 255).

Note: Try to make the red area do not overlap each other.
6.1.5 Particle analysis
After apply binarization, open particle analysis tool:
First of all, because some noise or dirty point was not clear by binarization. You need set the range of
particle’s size. Reduce the range of particle’s size will make the result much more precision.
To restore the maximum value to infinity, click the button.
Check the “Show Outlines” checkbox to show border and number of particles after analysis.

Click “Analyze Particles” button to start analysis, the result will show blow.
Click “CSV” button to export to data to report file, the default path is: /mnt/sdcard/Image/Reports
