Information You Should Know Before Requesting Warranty Service
•Locate the model and serial numbers. A serial tag is located on the unit itself.
• For equipment service or maintenance, or with technical or special application inquiries, contact Technical
Services at 1-800-438-4851 or 1-740-373-4763 (USA and Canada). Outside the USA, contact your local
distributor.
Repairs NOT Covered Under Warranty
• Calibration of control parameters. Nominal calibrations are performed at the factory; typically ±1°C for
temperature, ±1% for gases, and ±5% for humidity. Our service personnel can provide precise calibrations as
a billable service at your location. Calibration after a warranty repair is covered under the warranty.
• Damage resulting from use of improper quality water, chemicals or cleaning agents detrimental to
equipment materials.
• Service calls for improper installation or operating instructions. Corrections to any of the following
are billable services:
1) electrical service connection
2) tubing connections
3) gas regulators
4) gas tanks
5) unit leveling
6) room ventilation
7) adverse ambient temperature fluctuations
8) any repair external to the unit
• Damage resulting from accident, alteration, misuse, abuse, fire, flood, acts of God, or improper

If the Anaerobic System is to be moved by forklift, leave the unit on the
skid until it has been moved to its designated location. A cardboard box

Installation and Start-Up
Location Locate the cabinet on a firm, level surface in an area of minimum
ambient temperature fluctuation. Allow adequate space at the rear and to
the side of the cabinet for connecting the gas supplies and the vacuum
pump. There should also be clearance behind the cabinet for the optional
foot switch. Penetration through the counter-top may be necessary to
accommodate the foot switch cord if the table is flush-mounted to the
wall.
Connect the interchange to an adequate house vacuum system or to an
intermittent vacuum pump. If the interchange is connected to a house
vacuum system, the system must meet the minimum requirements of 4
C M capacities to 24 in/Hg.
It is recommended that the countertop house vacuum systems be
supplied with a 1/2” non-tapering nipple. Conventional tapered barb
nipples restrict vacuum volume.
Caution If the system is to be connected to a house vacuum, refer to
Section 4 for instructions in reversing the direction of the vacuum
solenoid valve before connecting. s
1. Select a location for the vacuum pump and secure the pump as
illustrated in igure 1-2.
2. Install the nylon adapter (supplied) on the vacuum connection on the
rear plumbing panel.

3. Install the nylon adapter (supplied with vacuum pump kit) on the end
of the vacuum pump.
4. Run the vinyl tubing (1/2” ID, 3/4” OD) from the pump inlet to the
vacuum connection on the back of the cabinet and secure with the #10
SS hose clamps. Refer to igure 1-1.
5. Plug the line cord into the pump outlet (located on the back of the
plumbing panel). Check all connections fo leaks.

The gas mixture that provides optimum growth conditions for anaerobic
bacteria is an 85/5/10 mixture of N2/CO2/H1. Regulations in some
locations do not permit mixtures containing 10% hydrogen. In these
locations, a mixture containing 5% to 7% hydrogen is recommended.
ote If a 5% or less mixture of H2is used, an auxiliary
desiccant/palladium dryer may be required to maintain proper anaerobic
conditions. s
Install two-stage pressure regulators with dual indicating gauges at the
cylinder supply outlets for both the nitrogen and mixed gas supplies.
If the cylinders are to be stored in another room, manual shut-off valves
should be installed.
Low stage pressure gauges on both tanks should be adjusted between 10
and 15 PSIG.
To ensure a constant supply of both gases, it is recommended that the
operator keep extra tanks of both gases on the premises.
• Nitrogen: A “high purity” grade nitrogen, with a minimum purity of
99.99%, maximum moisture 3 ppm, is recommended.
• Tri-mixture (anaerobic gas): Component gases are recommended to be
of a “high purity grade”, the final gas mixture should result in less than
10 ppm O2.
The Anaerobic and Nitrogen gas supply fittings are located on the
plumbing panel on the back of the unit. Refer to igure 1-2. Securely
attach a vinyl line from the main source of Anaerobic and Nitrogen gas to
each appropriate serrated fitting. If a metal line is to be used, the serrated
fitting can be removed and the desired 1/8 MPT fitting can be added.
ote Gas supplies are connected one way for start-up and another way for
normal operation to save gas costs on initial start-up. s

Supplies

Connect the main supply of nitrogen to the serrated fitting, labeled
Anaerobic Gas, on the rear of the cabinet. The anaerobic gas should not
be connected during start-up.
ollowing the start-up procedure, reconnect the gas supplies for normal
operation.
Connect main supplies of both pure nitrogen and mixed gas to the
appropriate gas inlets at the back of the system.
Plug the foot switch line cord into the outlet labeled “ oot Switch”, on the
rear plumbing panel. Refer to igure 1-2.
1. Put the cuff into the rubber glove with the narrow end toward the
fingers. Pull the glove up over the cuff until the rolled edge slips over
the edge of the cuff.
2. Cut four 6-inch strips of the clear plastic tape provided. Run a piece of
tape from the middle of the glove cuff over the edge and half-way
down into the inside of the cuff. Use the other three pieces of tape in
the same manner, equal distance around the cuff.
3. Put your hand in the glove and carefully push it down through the
sleeve until one-third to one-half of the cuff portion is protruding
from the end of the sleeve.
4. With the plastic tape, tape the glove tightly into the sleeve in a circular
manner. Cover approximately 3/4 of the cuff area with tape. old over
the end of the tape to make replacement of the glove easier.
Plug the system’s line cord into a suitable electrical outlet. See the
electrical data plate mounted on the unit for exact electrical specifications.

To help conserve gas, put as much of the equipment as is practical into the
chamber at this time. Any equipment that must be put through the
interchange with both doors open should be put in before the gas is turned
on. The layout of the equipment should be well planned. 6 to 8 BBL Gas
Pak™ disposable anaerobic indicator strips (Mfr. #R65055, available
through a variety of suppliers) should be placed in the work chamber at
this time for use during the start-up procedure.
The following is a list of other possible equipment:

• Microscope - a binocular type with a “high eye point” is recommended
• Electrical Loop Sterilizer - SEPCO Electrostat Loop Sterilizer is recom-
mended

1. Install the wafers before start-up.

To prepare the chamber relief bubbler, fill the plastic bottle shipped with
the unit, with mineral oil (readily available where health care products are
sold) to a level of approximately 1-1/2”. Clip the bottle into the chamber
relief with the tubing extending approximately 1/8” into the mineral oil.
Once a month, change the mineral oil and clean the bottle with soap and
water. Check the bottle daily to ensure that the tube extends into the
mineral oil.
Caution Before starting the anaerobic system, it is extremely important
that the installation procedures have been followed correctly. If the system
has not been prepared correctly for operation, malfunction or damage to
the system may result. s
1. Before starting up the system, carefully check all gas connections and
seal both the inner and outer doors.
2. Make sure that the gloves are properly installed.
3. Make sure that the charcoal, catalyst and desiccant wafers are installed
in the cabinet before beginning the start-up procedure.
4. Check the vacuum pump by pulling a vacuum on the interchange with
both interchange doors closed.
5. Check the vacuum gauge to make sure that the interchange is holding
a vacuum.

Start-Up

The purpose of this procedure is to remove all atmospheric gas, particularly
oxygen, from the work chamber and replace it with a special anaerobic gas
mixture.
Caution Start-up takes three to four hours. Do not leave the system during
this procedure. s
1. Connect the N2gas supply to the anaerobic (special) gas connection at
the back of the unit.
ote Do not connect the anaerobic gas at this time. s
2. Turn on the N2supply at the source, adjust the low stage pressure to
10-15 PSIG and check all the connections for leaks.
3. Close and secure the outer door.
4. Open the inner door.
5. Open the incubator doors.
6. Turn the Vacuum, Nitrogen and Equalize switches to the Auto
position and turn the Gas System switch On.
7. Turn the Manual ill knob fully counterclockwise. Do not overturn.
8. Push the Start button to cause the vacuum system to evacuate the
atmospheric gas. It automatically shuts off and the work chamber fills
with nitrogen.
9. When the chamber relief bubbler bubbles, push the Start button again.
10. Repeat Step 9 a total of twenty times.
11. Turn the Manual ill knob O when the procedure is completed.
12. Turn the nitrogen O at the source.
13. Remove the N2from the special gas compression fitting, and connect it
to the N2fitting.
14. Connect the special gas to the appropriate fitting on the back of the
unit. Refer to igure 1-2.

15. With the Gas System switch ON and the Vacuum, Nitrogen, and
Equalize switches in the Auto position, repeat Steps 6 through 8 five
more times.
16. Turn O the Manual ill knob when the procedure is complete.
17. Turn the Catalyst an switch ON.
18. After one hour, open a BBL strip and check to see if the chamber is
anaerobic. The cabinet should now be below 1% oxygen.
Caution Let the cabinet sit overnight before adding the cultures. This
permits the catalyst wafer to remove any traces of oxygen that may be
present. s
The interchange is used to transfer items to and from the work chamber
without contaminating the work chamber with atmospheric gas. The
vacuum pump is programmed to operate until it pulls a 20 in/Hg vacuum.
Nitrogen is injected until the interchange reaches atmospheric pressure and
the interchange equalizes with anaerobic gas mixture until it reaches work
chamber pressure.
The transfer cycle is factory-set for three evacuations, two nitrogen purges
and one equalizing purge of anaerobic gas mixture. The operator may alter
the transfer cycle to one nitrogen purge, two vacuum functions, and one
equalizing purge of anaerobic gas mixture. Refer to Section 4.9 for
instructions in making this adjustment.
The duration of the transfer functions may be manually controlled by
setting any one of the three function switches to the Manual position.
ote Only one function switch may be in the Manual position at any
given time. Having two or more switches in the Manual position at the
same time wastes gas. s
Caution When using any of the function switches on Manual, the operator
must remain in the room and pay strict attention to system operation. s
Both the inner and the outer door must be securely closed during the
transfer cycle. The doors should not be reopened until the cycle is
completed and the appropriate door indicating light goes out.

Caution The inner and outer interchange doors should not be open at the
same time while the work chamber is operating under anaerobic
conditions. s
1. With the inner door securely closed and clamped, open the outer door
and place the material to be transferred into the interchange.
2. Secure the outer door.
3. or an automatic transfer cycle, press the Cycle Start button.
4. When the cycle is complete, the Equalize and both door lights go out,
and the Cycle Complete light comes on. Then it is safe to open the
inner door and remove the material.
5. After removing the material, be sure to secure the inner door.
Caution Always keep the inner door securely closed to prevent atmospheric
contamination due to accidental outer door openings. The outer door
should also be kept closed at all times. s
To remove the material from the chamber:
1. With the inner and outer doors secure, press Start Cycle.
2. When the Sequence Complete light comes on, open the inner door
and transfer the material to the interchange.
3. Close and secure the inner door, then open the outer door to remove
any material.
4. Close and secure the outer door.

In some cases, it is desirable to manually operate the interchange. To do
so, depress the function switches to Manual for the desired duration and
sequence.
If, for any reason, the unit circuit board should malfunction, the unit can
still be operated manually by following these procedures.
Caution rom the Auto sequence cycle, do not turn more than one
function switch to Manual at a time, or leave the system unattended while
a function switch is in the Manual position. When operating any of the
function switches on Manual, make sure both the inner and outer doors
are securely closed. The inner door must be securely latched shut, not
simply closed. s
1. Turn the Gas switch O , then ON. This will automatically transfer
cycle, during any function, back to the beginning of the program.
2. Put the Vacuum switch in the Manual position until the vacuum
gauge reads 20 inches. Then switch it back to the Auto position.
3. Put the Nitrogen switch in the Manual position until the vacuum
gauge drops to 2 inches. Then switch it back to the Auto position.
4. Repeat Steps 2 and 1.
5. Repeat Step 2 again, then switch the Equalize switch to Manual until
the vacuum drops to 0.
The interchange is now anaerobic. However, the Sequence Complete and
Sequence Not Complete indicating lights will not function correctly
because of Manual operation.
Operating the interchange manually has no effect on the glove chamber.
The glove chamber will always have a positive pressure of mixed gas to
maintain an anaerobic atmosphere.
