Terms and abbreviations
Digital slide / Slide Sli e igitize with a scanner microscope, for example Pannoramic SCAN. Consists of an
MRXS file an a fol er with the same name as the MRXS file.
Donor block The onor block is a stan ar tissue block that may be from surgical pathology, autopsy,
or research material.
Recipient block The tissue cores are remove from the onor an inserte into a “recipient” paraffin
block.
Clone Block A uplicate (triplicate, qua ruplicate, etc.) of a recipient block with i entical layout an
core or er.
TMA Tissue microarrays – The tissue microarray consists of an array of cylin rical cores of
paraffin-embe e tissue that are remove from preexisting “ onor” paraffin blocks.
PCR Polymerase Chain Reaction, a biochemical technology use for a variety of applications,
but the primary aim of the metho is to exponentially amplify particular DNA sequences.

1 Intended Use
TMA Grand Master is a software-controlle electronic laboratory evice that automates the creation
of tissue microarrays (TMAs) an also the process of PCR sampling.
The creation of TMA blocks (by taking tissue core samples from onor blocks an inserting them into
recipient blocks) an the insertion of extracte tissue cores into PCR tubes is controlle by using a
graphical user interface. The built-in igital camera units of TMA Grand Master give you a clear image
of each onor block, so you can aim the puncher more accurately; an a itional camera takes
pictures of the block labels. You can also save an image of each onor block with markers showing
the positions from where the tissue cores have been extracte .
You can export block-relate ata to ifferent types of sprea sheet files through the TMA Grand
Master software application to track associations between recipient block positions an the ID of the
onor block from which the core tissue was inserte into the recipient block.
1.1 Main eatures of TMA Grand Master
•It has a robust an stable mechanical main unit.
•It has an intuitive operation control via the keyboar an the mouse of a PC connecte to
the main unit.
•The evice measures block height base on which the software calculates the epth of
punch, rill, an insertion. Thus, embe e cores are place correctly into the recipient
block resulting in an even surface level.
•It makes image creation possible when a TMA block is create either for reference or for
archiving uring a research project.

•Block i entifiers are store in a atabase from which each block ata within a Project is
automatically loa e after the ID is entere , furthermore the whole project assembly ata
can be restore after exiting an restarting the software without saving any project ata.
•Maximum TMA han ling capacity:
−Up to twelve recipient blocks an 60 onor blocks at same time.
•It is capable of receiving 6 PCR cassettes with a hol ing capacity of 10 PCR tubes. The
software allows you to transfer four cores into each tubes, so a maximum number of 240
tissue samples can be collecte in 6 PCR cassettes uring a project if not interrupte .

2.2 Cabling
For require cable connections, the supplie cables must be plugge in as shown in the figure above.
To connect the main unit with the computer any of the three USB ports can be use .

2.4 Installing and Configuring TMA Grand Master Hardware

2.5 Transporting the TMA Grand Master Main Unit
Warning!
If you nee to move TMA Grand Master to another location, transport it in its
original packaging only.
1. Switch off the TMA Grand Master, the control computer an the monitor, an unplug all cables.

The following installation proce ure is for information purposes only – follow the
below steps only if you nee to re-install the software, or upgra e to a newer
software version.

Do NOT install any other application than the TMA Grand Master software
application an its auxiliary rivers on the control computer.
If the application is running, make sure there is no high traffic communication over
USB ports use by other evices.

1. Install the following rivers from the 3DHISTECH Installation Disk:
•TMA Gran Master USB river (CDM river)
•Camera river – VRmagic
2. Launch the TMAGrandMaster_2.0_RTM.exe file that you receive on a 3DHISTECH
Installation Disk or ownloa e in a compresse file.
3. Follow the instructions in the installation wizar .
4. Accept the En -User License Agreement, an click Next.
