This guide is intended for personnel who install, operate, and maintain ACQUITY UPLC M-Class
system modules. It gives an overview of the system’s technology and operation.
Intended use of the ACQUITY UPLC M-Class system
Waters designed the ACQUITY UPLC M-Class system to perform chromatographic separations.
The system is for research use only and is not intended for use in diagnostic applications.
Calibrating
To calibrate LC systems, adopt acceptable calibration methods using at least five standards to
generate a standard curve. The concentration range for standards must include the entire range
of QC samples, typical specimens, and atypical specimens.

When calibrating mass spectrometers, consult the instrument’s online Help system for calibration
instructions.
Quality control
Routinely run three QC samples that represent subnormal, normal, and above-normal levels of a
compound. If sample trays are the same or very similar, vary the location of the QC samples in
the trays. Ensure that QC sample results fall within an acceptable range, and evaluate precision
from day to day and run to run. Data collected when QC samples are out of range might not be
valid. Do not report these data until you are certain that the instrument performs satisfactorily.
ISM classification: ISM group 1 class B
This classification has been assigned in accordance with CISPR 11 Industrial Scientific and
Medical (ISM) instrument requirements.
Group 1 products apply to intentionally generated and/or used conductively coupled radio-
frequency energy that is necessary for the internal functioning of the equipment.
Class B products are suitable for use in both commercial and residential locations and can be
directly connected to a low voltage, power-supply network.

1ACQUITY UPLC M-Class system
Waters designed the ACQUITY UPLC M-Class system to perform a range of chromatographic
separations from nano-scale to micro-scale. With a maximum operating pressure of 103,421 kPa
(1034 bar, 15,000 psi), an analytical flow rate to 100 µL/min, and a trapping flow rate to 1 mL/min,
the system produces optimal linear velocity for columns that range in internal diameter from 75
µm to 1.0 mm.
The system is compatible with Waters mass spectrometers and optical detectors, supporting a
range of column diameters. Options include:
• NanoFlow ESI and NanoLockSpray sources for Waters time-of-flight and tandem MS, and
flow rate ranges from 200 nL/min to 2,000 nL/min.
• Micro-scale ESI probe for use in the ZSpray API and LockSpray dual electrospray ion sources
for flow rate ranges from 5 µL/min to100 µL/min.
• A 100-nL, light-guided flow cell for columns ranging from 300 µm to 1.0 mm ID with the
ACQUITY UPLC PDA and TUV detectors.
• A 10-nL, transverse-illuminated flow cell for nano-scale separations with the ACQUITY UPLC
TUV detector.
ACQUITY UPLC M-Class systems that include mass spectrometry, optical detection, or both can
analyze compounds found in low concentrations amid complex matrices, quantifying peptides, or
metabolites (when used with a tandem mass spectrometer).
1.1 Configurations of system modules
This section shows examples of the ACQUITY UPLC M-Class system stack consisting of the core
modules. Your system configuration may differ according to the number and types of modules it
contains. For example, if your system includes a mass spectrometer, the system modules may be
arranged in two separate stacks.

1.2 System components
The capabilities of the ACQUITY UPLC M-Class system depend on the components installed.
The following sections briefly describe the system components and their functions.
1.2.1 System features

• The system components are optimized for use with sub-2-µm, particle liquid chromatography,
and reduced fluid volumes.
• 100-nL, 10-mm path-length, light-guided TUV, and PDA detector flow cells deliver sensitivity
and resolution to ACQUITY UPLC M-Class applications on columns ranging from 300-µm to
1.0-mm ID.
• The columns, tubings, and fittings support flow rates for a gradient elution from 200nL/min to
100 µL/min at 103,421 kPa (1034 bar, 15,000 psi).
• With mass spectrometry, NanoLockSpray achieves exact mass measurement with columns
ranging from 75-µm to 1.0-mm ID.
• Solvent managers with flow control modules provide accurate NanoFlow rates. An optional
auxiliary solvent manager comprises two isocratic pumps. The A-side provides an additional
eluent with a flow rate range from 0.0 to 1.000 mL/min. The B-side provides NanoLockSpray
addition under flow control with a flow rate range from 0.0 to 100.0 µL/min.
• A micro-scale ESI probe in the ZSpray API interface delivers superior sensitivity. As column
ID decreases, this micro-probe reduces band-broadening to maintain chromatographic
integrity.
1.2.2 Auxiliary Solvent Manager
The Auxiliary Solvent Manager (ASM) is a high-pressure pump that provides pulse-free flow rates
at operating pressures as high as 103,421 kPa (1034 bar, 15,000 psi). The ASM consists of two
isocratic pumps, pump A and pump B. Each pump is fitted with two linear-drive actuators, which
together serve as a single reciprocating “serial” pump that delivers precise flow of a single
solvent. The solvent flows to the µSM-FL or column at an overall flow rate of 0.0 to 1000.0 µL/min
from pump A and 0.0 to 100.0 µL/min from pump B. Sometimes the ASM is also used to flow lock
mass directly into the mass spectrometer, usually from the B side.

1.2.3 µBinary Solvent Manager
The µBinary Solvent Manager (µBSM) is a high-pressure pump that provides steady (pulse-free)
solvent flow at flow rates ranging from 0.0 to 6.0 µL/min (under closed-loop control) or to 100.0
µL/min (under open-loop control) at 103,421 kPa (1034 bar, 15,000 psi).

1.2.4 µSample Manager - Fixed Loop
The µSample Manager - Fixed Loop (µSM-FL) manages sample injections. It uses a fixed loop-
based mechanism to inject samples. After a puncture needle pierces the covering of a well or vial,
a sample needle extends through the puncture needle and aspirates the sample. The aspirated
sample is then drawn toward the sample loop while weak-wash solvent is simultaneously
dispensed through the needle, behind the sample. The wash syringe then pressurizes the needle
from the tip, compressing the air gaps around the sample and positioning the sample completely
within the loop. When the sample is in position, the injection valve is actuated to the inject
position, and high-pressure flow from the pump impels the sample onto the chromatographic
column.
The µSM-FL employs a thermal system that maintains a set temperature in the sample
compartment. When used in conjunction with the optional sample organizer, they create a
combined, thermally controlled compartment.

1.2.5 Sample Organizer
The optional Sample Organizer enables automatic processing and increased throughput by
storing multiple microtiter or vial plates and transferring them to and from the µSM-FL.
The Sample Organizer’s storage-shelf compartment can hold a selection of ANSI plates. A
heater/cooler thermally conditions the shelf compartment and maintains the temperature at a
specified set point.

1.2.6 Trap Valve Manager
The Trap Valve Manager (TVM) comprises two trapping valves and a column-heater
compartment. The column extends from the TVM arm and plumbs into an optional optical
detector or a mass spectrometer. In addition, the TVM accommodates several columns ranging
from 75-µm ID to 2.1-mm ID, in lengths up to 25 cm.

1.2.7.1 Optical detection
The ACQUITY UPLC PDA and TUV are optical detectors that detect and quantify concentrations
of sample analyte. Low-dispersion flow cells developed with Teflon-AF are available for use with
microscale 1.0-mm and 300-mm ID columns.

1.2.7.2 Mass detection
The ACQUITY UPLC M-Class system maximizes peak capacity, extending the dynamic range of
sample analysis. The ACQUITY UPLC M-Class system is frequently used with mass
spectrometers, whether analyzing complex samples in wide-ranging matrices with a high-
resolution MS, or quantifying peptides, metabolites, or other targeted experiments with tandem
MS. Engineered for the lowest dispersion, including an improved ESI micro-probe design (and
with an extended pressure envelope) complex separations are accelerated without compromising
the chromatography.

1.2.8 Column technology
Configure columns with the TVM using no valves for direct injection, or one or two valves (when
used with two-dimensional applications, for example).
1.2.8.1 Columns
The TVM supports nano-scale (75-mm ID), capillary-scale (300-mm ID), and micro-scale (1.0-mm
ID) separations. You can choose among these options when selecting columns for the system:
• Nanoscale analytical columns:
• 75 µm to 150-µm ID
• 50 mm to 250 mm length
• Capillary-scale analytical columns:
• 300 µm-ID
• 50 mm to 150 mm length

1.2.9.1 MassLynx
MassLynx software controls the ACQUITY UPLC M-Class system. It acquires, analyzes,
manages, and processes mass-spectrometry data.

The instrument console is a software application that you use to configure settings, monitor
performance, run diagnostic tests, and manage the system and its modules.

1.2.10.1 Waters micro cart
The optional Waters micro cart provides a mobile platform for the system. The cart holds the
system modules and provides electrical outlets. Used with a mass spectrometer, the cart
positions the column outlet close to the mass spectrometer’s inlet probe, minimizing system dead
volume.
Note: You cannot use the micro cart with the sample organizer.
