The source exhaust system is designed to be robust and leak-tight. Waters recommends you
perform a hazard analysis, assuming a maximum leak into the laboratory atmosphere of 10% LC
eluate.
Flammable solvents hazard
Notice: To prevent instrument damage, a gas fail cable must be fitted between the gas fail
connection on the rear of the Xevo TQD IVD and the stop flow connection on the I-Class BSM
module (see the rear panel connections section of the ACQUITY UPLC I-Class / Xevo TQD IVD
System Guide, part number 715005210IVD).
Overload hazard
Warning: To avoid electric shock, use the SVT-type power cord in the United States and
HAR-type (or better) in Europe. The main power cord must only be replaced with one of
adequate rating. For information regarding what cord to use in other countries, contact your
local Waters distributor.
Warning: To avoid exposure to toxic substances and biohazards from O-ring leaks in the source
exhaust system, observe these precautions:
• Renew the source O-rings at intervals not exceeding one year.
• Prevent chemical degradation of the source O-rings, which can withstand exposure only to
certain solvents (see page 143), by determining whether any solvents you use are chemically
compatible with the composition of the O-rings.
Warning: To prevent ignition of flammable solvent vapors, Nitrogen gas must flow
continuously through the mass spectrometer’s ion source when solvents are flowing into
the source. The Xevo TQD IVD monitors the nitrogen gas input pressure, and MassLynx
software displays a nitrogen gas fail warning if the pressure falls below 400 kPa (4 bar, 58
psi). Additionally, the capillary voltage and desolvation temperature are switched off, the
fluidics divert valve switches to the waste position, and a stop flow signal is sent from the
Xevo TQD IVD to the UPLC system, preventing solvent from flooding the source.
Warning: To prevent personal injury, ensure equipment placed on top of the Xevo®TQD
IVD does not exceed 15 kg.

Hazards associated with removing an instrument from service
When you remove the instrument from use to repair or dispose of it, you must decontaminate all of
its vacuum areas. These are the areas in which you can expect to encounter the highest levels of
contamination:
• Source interior
• Waste tubing
Warning: To avoid injuries from broken glass, falling objects, or exposure to biohazards or toxic
substances, never place containers on top of the instrument or on its front covers.
Warning: To avoid burn injuries, before performing maintenance operations that involve
handling components inside the mass spectrometer's ion source, allow the source interior
to cool.
To avoid personal contamination with biohazards, toxic materials, and corrosive materials, wear
chemical-resistant gloves during all phases of instrument decontamination.

The need to decontaminate other vacuum areas of the instrument depends on the kinds of
samples the instrument analyzed and their levels of concentration. Do not dispose of the
instrument or return it to Waters for repair until the authority responsible for approving its removal
from the premises specifies the extent of decontamination required and the level of residual
contamination permissible. That authority must also prescribe the method of decontamination to
be used and the appropriate protection for personnel undertaking the decontamination process.
You must handle items used to carry sample into the source area in accordance with laboratory
procedures for contaminated vessels and sharps. To avoid contamination by carcinogens, toxic
substances, or biohazards, you must wear chemical-resistant gloves when handling or disposing
of used oil.

Audience and purpose
This guide is for operators of varying levels of experience. It gives an overview of the instrument
and explains how to prepare it for operation and maintenance.
Operators of the Xevo TQD IVD should be professionally trained and qualified laboratory
personnel.
Intended use of the Xevo TQD IVD
Waters designed the Xevo TQD IVD for use with the ACQUITY UPLC IVD system and the
ACQUITY UPLC I-Class/Xevo TQD IVD system to perform mass spectrometry for in vitro
diagnostic use.
Compliance with Medical Device Regulations
Important: The ACQUITY UPLC I-Class/Xevo TQD IVD complies with the European Union In
Vitro Diagnostic device Directive 98/79/EC only when the mass spectrometer is fitted with a
Waters ESI source. Do not fit the instrument with other Waters or third-party sources.
Environmental requirements
Temperature
The ambient temperature range required for normal operation is 15 to 28 °C (59 to 82 °F).
The optimum temperature range is 19 to 22 °C (66 to 72 °F).
Short-term (1.5 h) variations must be no more than ±2 °C (3.5 °F).
Humidity
The relative humidity in which the instrument and pumps are to operate must be in the range of
20% to 80%, non-condensing.
The Waters ACQUITY UPLC I-Class/Xevo TQD IVD system is CE-marked
according to the European Union In Vitro Diagnostic Device Directive 98/79/EC.

Calibrating
To calibrate LC systems, follow acceptable calibration methods using at least five standards to
generate a standard curve. The concentration range for standards must include the entire range of
QC samples, typical specimens, and atypical specimens.
When calibrating the mass spectrometer, consult the calibration section of the ACQUITY UPLC
I-Class/Xevo TQD IVD System Operator’s Guide (part number 715005210IVD).
Quality control
The following quality controls are required when using Waters in vitro diagnostic (IVD) LC-MS/MS
Systems.
To prevent incorrect reporting of results, verify proper sample vial placement and positioning
against the sample list, prior to sample injection.
Routinely run three matrix-matched QC samples at the Lower and Upper Limit of Quantitation and
the mid-point of the Measuring Range for each analyte. Additionally, QCs at decision points may
speed detection of critical deviations. If sample trays are the same or very similar, vary the location
of the QC samples in the trays. Ensure that QC sample results are within acceptable ranges, and
evaluate precision from day-to-day and run-to-run. Data collected when QC samples are out of
range might not be valid. Do not report these data until you are certain that the instrument
performs satisfactorily.
TargetLynx™ processing includes calibration curve, internal standard, and quality control flagging
features. Waters recommend including these capabilities when validating quantitative clinical
methods, to automate detection of erroneous results. Nevertheless, a manual inspection of data
should be performed prior to reporting results.
To reduce the possibility of an incorrect patient result due to carryover caused by exhaustion of the
wash solvent, run appropriate blank samples during the sample analysis.

When analyzing samples from a complex matrix, note that the matrix components can adversely
affect LC/MS results, enhancing or suppressing ionization. To minimize these matrix effects,
Waters recommends you adopt the following measures:
• Prior to the instrumental analysis, use appropriate sample pretreatment such as protein
precipitation, liquid/liquid extraction (LLE), or solid phase extraction (SPE) to remove matrix
interferences.
• Verify method accuracy and precision using matrix-matched calibrators and QC samples.
• Use one or more internal standard compounds, preferably isotopically labeled analytes.
• To ensure data integrity, Waters recommends you run QC samples before and after test
samples within a single analysis. Additionally, you can periodically run known or PT/EQA
samples as unknowns to verify assay results for a single analysis.
Analytical methods for use in reporting clinical results should be evaluated and validated according
to local regulations and should conform to internationally recognized guidelines, for example,
Clinical and Laboratory Standards Institute guideline C50-A, Mass Spectrometry in the Clinical
Laboratory: General Principles and Guidance; Approved Guideline, C62-A Liquid
Chromatography-Mass Spectrometry Method; Approved Guideline and related documents.
Always follow Good Laboratory Practice (GLP) for method development, method validation, and
quality control.

1Specifications and Operating
Modes
1.1 Uses and compatibility
The Waters®Xevo®TQD IVD is a tandem quadrupole, atmospheric pressure ionization (API)
mass spectrometer. Designed for routine UPLC®/MS/MS analyses in quantitative applications, it
can operate at fast acquisition speeds compatible with UltraPerformance LC®applications.

1.1.1 IntelliStart technology
IntelliStart™ technology monitors LC/MS/MS performance and reports when the instrument is
ready for use. It automatically tunes and mass calibrates the instrument and displays performance

readbacks. Integrated with MassLynx®mass spectrometry software and ACQUITY UPLC®
Console software, it enables simplified setup of the system for use in routine analytical and open
access applications. (See page 20.)
The IntelliStart fluidics1system is built into the instrument. It delivers sample directly to the MS
probe from the LC column or from two integral reservoirs. The integral reservoirs can also deliver
sample through direct or combined infusion so that you can optimize instrument performance at
analytical flow rates. See the instrument’s online Help for further details of IntelliStart.
1.1.2 Software and data system
MassLynx software, version 4.1, controls the mass spectrometer, and the OpenLynx™and
TargetLynx™application managers are included as standard software.
Note: OpenLynx is not supported on IVD systems.
MassLynx software acquires, analyzes, manages, and distributes mass spectrometry, ultraviolet
(UV), evaporative light scattering, and analog data. It enables these major operations:
• Configuring the system
• Creating LC and MS/MS methods that define operating parameters for a run
• Using IntelliStart software to automatically tune and mass calibrate the mass spectrometer
• Running samples
• Monitoring the run
• Acquiring data
• Processing data
• Reviewing data
• Printing data
Consult the user documentation for MassLynx, version 4.1, and the online Help for more
information about using MassLynx software.
1.1.3 Instrument Console software
You use the Instrument Console, a software application, to configure settings, monitor
performance, run diagnostic tests, and maintain the system and its modules. The Instrument
Console functions independently of MassLynx software. As such, it does not recognize or control
MassLynx data systems. Consult the Instrument Console system’s online Help for details.

components and the fluid pathways within and among instruments or devices. It also appears in
the product name “IntelliStart fluidics” where it describes a mass spectrometer’s integral apparatus
for delivering sample and solvent directly to the instrument’s probe. Finally, the term can arise in
the context of a component part name, as in “fluidics drawer”.
