Sample to Insight
Starting the instrument
Switch on the instrument using the power switch located at the rear. An
indicator light in the bottom left corner of the instrument screen lights
up when the instrument is on. The screen remains blank during startup,
which can take up to 1 minute.
Loading reagents and the PyroMark Q24 Plate
1. Fill a cartridge with the reagent volumes indicated by the PyroMark
Q24 Software and according to the instructions provided in the
PyroMark Gold Q24 Reagents Handbook.
2. If the instrument was already on, ensure that it is not performing a run
and then open the lid. An audible warning signal will alert you if the
lid is opened when it is unsafe to do so.
3. Open the cartridge gate and insert the cartridge with the label facing
you. Push the cartridge in fully and then push it down.
4. Ensure that the cartridge is properly inserted (a line should be visible
at the front) and close the gate.
5. Open the plate-holding frame and position the PyroMark Q24 Plate
with samples on the instrument heating block.
6. Close the plate-holding frame and the instrument lid.
Starting and monitoring a run
1. Insert the USB stick with the run files created with the PyroMark Q24
Software into the USB port at the front of the instrument.
2. Select Run in the main menu and press OK.
3. Use the scroll arrows to select the desired run file and press Select.
The instrument will begin dispensing reagents when all preset pressure
and temperature levels are reached (this may take several minutes).
PyroMark®Q24 Quick-Start Guide
This document briefly describes how to set up the PyroMark Q24 Instrument, start a Pyrosequencing®run, obtain run data,
and shut down the instrument. Before using the PyroMark Q24, it is essential that you read the PyroMark Q24 User Manual
carefully and pay particular attention to the safety information.

WARNING
Risk of personal injury and material damage.
To avoid contact with moving parts during operation,
the PyroMark Q24 Instrument must be operated with
the lid closed.
Do not touch the needles at the bottom of the cartridge.
Small particles and fibers may obstruct the needles.

During a run, the instrument screen displays the Pyrogram®of the
selected well. Use the scroll arrows to view the Pyrogram of other wells.
After the run
1. When the instrument confirms that the run is finished and the run file
has been saved to the USB stick, press Close.
2. Remove the USB stick. If it was removed before the run finished, insert
the USB stick. Select Administration and then Copy Unsaved Runs.
3. Remove and discard the PyroMark Q24 Plate.
4. Remove the cartridge and discard remaining reagents. If the cartridge
is to be reused, follow the instructions in the PyroMark Q24 User
Manual to clean it properly.
5. Analyze the run with the PyroMark Q24 Software (see the PyroMark
Q24 Software User Guide).
Shutting down the instrument
1. Select Shutdown in the main menu and press OK.
2. When the message “It is now safe to turn off the instrument” appears
in the screen, switch off the instrument. The power switch is located at
the rear of the instrument.
3. Press the light button at the rear of the instrument and check that the
coolant level is visible in the window. If not, please contact QIAGEN
Technical Services.
4. If the instrument requires cleaning, follow the instructions provided in
the PyroMark Q24 User Manual.
