Analyser Components
Outside the Analyser
1. Stain table –Cover provides access to stain table and internal components. Cover is
LOCKED while analyzer in use.
2. Smear Table –Cover provides access to stain table and internal components. Cover is
LOCKED while analyzer in use.
3. Storage table –Holds cassettes containing stain slides or blank cassettes. Holds up to 100
cassettes
4. Main power switch –Turns main power ON/OFF
5. Barcode reader and cap piercer –Samples run in sampler mode have their barcodes
read and are then aspirated by the cap piercer.
6. Supply table –Holds empty dry cassettes. Slides prepared manually can be placed in the
second cassette on supply table for staining only. Holds up to 100 cassettes
7. Start-up switch - Starts analyzer up following maintenance
8. LCD Screen –User interface. Used to perform a variety of operation including manual
smear and/or stain, maintenance, shutdown etc.
9. Sampler unit –Racks are placed on the right hand side and once processed exit on the
left hand side.

Inside the Smear Table
1. Micro tube holder –Samples requiring Manual (micro) sampling are place in here for
aspiration
2. Smear drier unit –Dries smear following spreading process
3. Slide glass shift unit - Responsible for moving the slide to the dispensing/smear position
and then on to the smear drier unit
4. Smear unit –prepares the smear by adjusting the angle and speed of the spreader to one
of eight levels according to the Hct.
5. Blood dispensing unit –Once the sample has been mixed and aspirated the blood
dispensing unit delivers the appropriate amount of blood on to the slide located and at the
dispensing/smear position.
6. Slide removal unit –Removes slides from the slide towers and delivers it to the slide glass
shift unit.
7. Printer –Prints information required on frosted end of slide according to settings.
8. Drying unit –Fan dries slide after printing.

9. Slide storage and cassette rotator –Cassette rotator rotates single cassette supplied by
the supply table to the horizontal position to allow smeared and printed slides are placed
into the single cassette. Cassette is then rotated vertically to allow delivery to cassette and
slide to stain table.
10. Slide supply towers - Two supply towers are located on the smear table (A) and (B).
Sensor use weight to detect whether slide towers are empty.

Inside the Stain Table
1. Methanol pipette –Slide is lifted from cassette and methanol is dispensed into the single
cassette following which the slide is returned to the cassette.
2. Stain pipette 1 - Slide is lifted from cassette and stain 1 is dispensed into the single
cassette following which the slide is returned to the cassette. Dependent on staining
protocol the slide is then lifted again and dried by the fan.
3. Stain pipette 2 –Removes stain 1 from the cassette and dispenses the diluent or rinse
solution for stain 1.
4. Stain pipette 3 –dispenses diluent for stain 2 into cassette after the staining solution and
rinse solution have been drained.
5. Rinse pipette –Rinses smears after staining.

Smear Production and Staining
Principles
Production of stained smears is a two-step process.
1. Smear production

Smear Production
Once barcode has been read or smear request manually entered the first step in smear production
is for a slide to be removed from either slide tower (A) or (B) to the slide removal position (1). The
slide is then transferred from the slide removal position (1) to the slide glass shift unit (D) by the
slide glass supply unit (C). The slide glass shift unit (D) then transfers the slide from this position
(2) to the dispensing and smear position (3) where the dispensing pipette dispense blood evenly
over a wide area and the smear unit (E) prepares the smear by adjusting the angle and speed of
the spreader glass based on the Hct value.

Once smear is complete slide is moved back by the slide glass shift unit (D) to the smear drier unit
(4) where they are dried. From here they are moved to the slide print position (5) where the printer
(F) prints sample information on the frosted end of the slide. From the slide print position (5) the
slide is moved through a seconded drier to ensure both smear and print are dried to the slide
storage position (7). A single slide cassette moves in from the cassette supply table to the single
cassette rotator (9) where it is rotated through 90º to allow the prepared smear to be moved into it
at the slide storage position (7). The single cassette contain the prepared smear is then rotated
back through 90º to be transported to the smear table (10).

Staining Principles
Romanowsky stains, such as Giemsa, Leishman and May-Grunwald, are used widely for the
routine staining of peripheral blood films. Whichever combination of stains is used the distinction
in staining achieved due to the balance of basic and acidic components within the stains. The
colour a particular cellular component stains, is dependent on the binding that occurs between the
staining compounds and chemical structures within the cell as well as interaction between dye
molecules, pH and time.
Proteins and nucleic acids bind to the basic component of the stain, resulting in a blue-violet or
blue colour. Whereas the basic chemical structures within haemoglobin have a high affinity for the
more acidic component of the stain, resulting in a red or orange colour. Eosinophil granules and
basic compound which has a strong affinity for acidic component of the stain. Basophil granules
contain heparin, which is strongly acidic and therefore has a high affinity for basic component.

Whichever combination of stains are used the process must be carried out at the correct pH. If the
pH is too acidic the basophilic components appear too pink. If the pH is too alkaline conversely
there is over staining as too much basic stain is taken up. These reaction are also dependent on
time as DNA binds to staining compounds more rapidly than RNAand Haemoglobin iseven slower
and as a results incorrect concentration of stains and/or timings will result in poor stains.

Introduction to LCD Screen
1. Menu name - Displays name of menu currently selected

There are two types of shutdown that need to be performed on the SP-series analyzer,
•Shutdown 1 should be performed at least every 24hrs if analyzer continuously in use, six
days a week.
•Shutdown 2 Should be performed once a week on the day that shutdown 1 is not
performed.

To perform either shutdown procedure ensure that the analyzer status displays ready. NOTE: If
analyzer is on a tracked system please ensure analyzer is isolated so that no further work is sent
to the SP-series analyzer.
Shutdown 1 (Daily)
Ensure analyzer status is READY. From main menu select the [Shutdown] to open the shutdown
menu. Select the either [Shutdown1 (Daily)]. The corresponding shutdown confirmation screen will
appear. Ensure that all cassette and methanol requirements are fulfilled.
•12 Slide cassettes in supply table
•400ml of methanol
•2ml of CellClean in the sample rack in front of the aspiration position
Place 2mls of CellClean into a blank sample
tube with cap on, Place tube in position 10 of
sample rack on left hand side of sampler unit
and align with mark.

Press [OK]. The shutdown progress screen will appear and once shutdown is complete analyzer
will turn off. Turn SP-series analyzer [OFF] at mains power switch. Perform analyzer start-up
procedure (see page 22)
Check Trap Chamber
The trap chamber prevents fluid flowing back into the analyzer if a blockage should occur and
therefore should be checked on a daily basis.
While analyzer is shut down check trap chamber. The trap chamber should be free of fluid. If fluid
is found in trap chamber unscrew trap chamber and discard fluid. Clean and dryboth trap chamber.
Place float back in trap chamber with the rounded end facing down and pointy end facing up.
Replace trap chamber.
Cleaning Spreader Glass
From main menu select [Maint.] and
[Spreader glass] to open ‘Spreader glass
replacement’ menu. Press [OK] to move
smear unit forward.

Once smear unit has moved forward open smear unit cover and clean spreader glass using
moistened gauze with deionized water.
NOTE: if spreader glass moves during cleaning simply lightly push spreader back into place.
Once spreader has been clean return
press [OK] to return smear unit to home
position.
When prompted to ‘Reset the counter of
the spreader glass’ press [Cancel].
NOTE: Spreader glass counter should only
be reset if spreader glass has been
changed.
