1 About This Manual
This Operating Manual describes the MagicPrep™NGS instrument and provides all the information
required for operating it safely and for maintaining it in good working order. This manual must be read
carefully before performing any work on the MagicPrep NGS and before using it.
This chapter outlines the purpose of this manual and specifies the product referred to. Furthermore, it
explains the use of symbols and conventions as well as further general information.
1.1 Scope of This Manual
This manual applies to:

1.3 Intended Use
The MagicPrep NGS System is a stand-alone instrument and associated reagent kits that allow the
unattended and fully automated preparation of next generation sequencing (NGS) libraries from
nucleic acid samples (i.e., RNA or DNA previously extracted and purified from biological samples)
using selected proprietary Tecan Genomics, Inc., chemistries.
The system is For Research Use Only, Not for use in diagnostic procedures.
1.4 Areas of Use / Area of Application
The MagicPrep NGS system can only be used for the fully automated preparation of next generation
sequencing (NGS) libraries using the specific Tecan Genomics MagicPrep reagent kits.
In each environment the individual laboratory is responsible for the validation of the MagicPrerp NGS
instrument together with the specific MagicPrep NGS Reagent Kits for the laboratory’s application
workflow or method(s).
1.5 Improper Use
Improper use may prejudice the MagicPrep NGS instrument safety concept.
The MagicPrep NGS instrument must not be used with reagents or components that are not
approved by Tecan.
The MagicPrep NGS instrument is not explosion-proof and should not be installed in locations
where there is a risk of explosion.

The MagicPrep NGS instrument should not be used in the absence of functional safety
devices.
1.6 Warranty
The MagicPrep NGS instrument must not be used with components that are not approved by Tecan
The use of unapproved components may impair the safety concept of the MagicPrep NGS instrument.
The use of unapproved components would invalidate any warranty of safety and compliance to
national and international standards, as required for NRTL certification, by EU directives, etc.
1.7 Trademarks
The product names, whether registered or unregistered trademarks, mentioned in this manual are
reproduced for identification purposes and remain the exclusive property of their respective owners.
For simplicity reasons, the trademark symbols such as ® and ™are not repeated in the manual.

This chapter describes the safety concept of the MagicPrep NGS instrument, provides general rules
of correct behavior, and warnings concerning health hazards associated with the use of the
MagicPrep NGS instrument.

2.2 General Safety consideration
MagicPrep™NGS is designed and built-in accordance with the present state-of-the-art
technology and the recognized technical safety regulations. Nevertheless, risks to users,
property and the environment can arise if the Magic Prep NGS is used without due care and
attention.
The safety of all users and personnel depends on the strict observation of these safety
instructions and awareness of the safety-related warnings provided in this manual.
•Please pay great attention to the following general safety information.
•This manual must always be available to all persons performing the tasks described herein.
•Legal regulations, such as local, state and federal laws concerning the use or application,
as well as the handling, of dangerous materials in connection with the MagicPrep NGS must
be strictly followed.
•The operating company is responsible for defining instructions in accordance with company
procedures and local legal requirements. The instructions provided by the operating
company must be strictly observed.
•Observe the correct environmental conditions for storage and operation.
•Structural changes to the safety devices are forbidden.
•Damaged safety devices must be replaced immediately as described in this manual.
•The MagicPrep NGS must not be modified in any way without prior consultation and written
approval of Tecan. Tecan will reject any claim resulting from unauthorized modifications.
•Fire hazard caused by the improper use of the MagicPrep NGS. The MagicPrep NGS
should not be installed in locations where there is a hazard of explosion.

•Chemical, biological, and radioactive hazards can be associated with the substances used
or the samples and reagents processed with the MagicPrep NGS (e.g., during loading and
unloading). The same applies to waste disposal.
–Always be aware of possible hazards associated with these substances.
–Use appropriate protective clothing, safety goggles, respirators, and gloves.
–The handling of substances and the disposal of waste may be subject to local, state, or
federal law, or to regulations with regard to health, environment, or safety. Strictly
observe the corresponding provisions.
–If the user is working with biohazardous substances then a biohazard safety sign
should be applied by the user. Apply the label on the front door in a position visible to
the user and convenient for the application.
•Any contamination must be dealt with immediately as described in this manual.
•The user is responsible for ensuring that the MagicPrep NGS is always operated under
proper conditions.

•Risk of incorrect measuring results. After system care or maintenance has been performed,
operation must only be resumed after the correct system operating conditions have been
verified.
•Always use recommended reagent kits within expiration date to assure good system
performance and reliability.
•Always wear protective clothing according to GLP.
•Heavy load! Use two people to lift the instrument.
•Fire hazard caused by flammable liquids or system liquids.
–Avoid the formation and accumulation of flammable vapours.

2.3 Operating Company
Responsibilities
The operating company must ensure that the MagicPrep NGS instrument and in
particular the safety features, function properly and that all the personnel in
contact with the instrument are adequately trained.
Method and process validation

Ensuring that all people in contact with the MagicPrep NGS are adequately
trained
Ensuring the availability of appropriate protective clothing and equipment
Ensuring the maintenance and safe operation of the MagicPrep NGS
instrument.

2.4 Method and Process Validation
Responsibility
of the Key
Operator
While performing method and process validation, pay attention to the following.
Only use Tecan Revelo MagicPrep NGS Reagent Kits.
Check that the pipette used to dispense the sample and magnetic beads
meets the accuracy and precision requirements for the process or workflow.
Remove completed library samples from the MagicPrep NGS instrument
within 65 hrs. of the preparation being completed.
2.5 User Qualification
The laboratory personnel must be fully qualified and trained to operate the MagicPrep NGS
instrument. The work described in this Operating Manual must only be performed by authorized
personnel with the qualifications prescribed below.
Laboratory personnel must:
have suitable technical training
be familiar with the laboratory safety regulations and directives
be familiar with the instructions for the safety elements of the instrument

use protective clothing and equipment
be familiar with and adhere to good laboratory practices
and have read and understood the instructions in the Operating Manual
2.5.1 Operator

2.6 Safety Elements
When the front door of the MagicPrep NGS instrument is open, do not insert your hand(s) into the instrument.
When opening the Magic Prep NGS front door and inserting or removing the reagent
cartridge and/or sample deck, please take care not to insert your hand(s) into the
instrument.
When the front door is open, there is no power to any of the moving parts within the
MagicPrep NGS instrument, however, there are instrument components that could
potentially pinch or catch on a hand when inserted into the instrument

When opening the Magic Prep NGS front door and inserting or removing the
reagent cartridge and/or sample deck, please take care not to insert your
hand(s) into the instrument
When the front door is open, there is no power to the thermal PCR
component within the MagicPrep NGS instrument, however, there may be
residual heat that could potentially burn a hand when inserted into the
instrument
2.7 Decontamination Declaration
In addition to regular system care, and in accordance with standard laboratory regulations, the
MagicPrep NGS instrument and its parts must be thoroughly decontaminated in the following
circumstances:
In the event of accidents (e.g., spillage, etc)
Before returning the MagicPrep NGS to Tecan (e.g., for repair)
Prior to storage.
Prior to disposal.
In general, before moving the MagicPrep NGS from its location
The owner of the instrument has full responsibility for the effective decontamination of all the
equipment.
Before returning the MagicPrep NGS instrument to to Tecan, the owner of the instrument must
complete and sign the Decontamination Declaration form, confirming that the decontamination has
been performed in accordance with good laboratory guidelines. Contact Tecan Genomics support to
obtain this form and refer to section Decontamination.
Tecan reserves the right to refuse to deal with any MagicPrep NGS instrument or reagent
kit parts that is not accompanied by the Decontamination Declaration form.

The MagicPrep NGS instrument has been designed to be able to operate under the above

The MagicPrep NGS instrument has been tested and found to comply with the
limits for a Class A digital device, pursuant to part 15 of the FCC Rules and
CISPR 11/EN 55011. These limits are designed to provide reasonable protection
against harmful interference when the equipment is operated in a commercial
environment. This equipment generates, uses, and can radiate radio frequency
energy and, if not installed and used in accordance with the instruction manual,
may cause harmful interference to radio communications. Operation of this
equipment in a residential area is likely to cause harmful interference in which
case the user will be required to correct the interference at his own expense.
It is the operator’s responsibility to ensure that a compatible electromagnetic
environment for the MagicPrep NGS can be maintained in order that the
MagicPrep NGS will perform as intended. Do not operate the MagicPrep NGS in
close proximity to sources of strong electromagnetic radiation (e.g., unshielded
intentional RF sources), as these can interfere with the proper operation.

4 Description of Function
This chapter explains the basic functions of the MagicPrep NGS instrument.
4.1 Overview
The MagicPrep NGS instrument and associated reagent kits provide an automated, hands-free
method of library preparation for NGS sequencing. The workflow starts with end repair of fragmented
DNA or enzymatic fragmentation of genomic DNA followed by adapter ligation, PCR amplification and
bead purification to produce the final library.
Samples of DNA or RNA along with magnetic beads are placed onto a sample deck which is inserted
into the MagicPrep NGS instrument. This sample deck contains the bulk reagents, pipette tips, and
several rows of PCR tubes which may contain additional reagents. A reagent cartridge specific for the
library preparation method is also inserted into the MagicPrep NGS instrument. This contains the
reagents specific for the library preparation method. Samples are manipulated fully automatically in
the MagicPrep NGS instrument using conventional technology which includes an 8-channel pipetting
engine with disposable tips, magnetic bead separation using a conventional magnet system and PCR
using a thermal cycler.
The operator is not able to access the internal parts of the MagicPrep NGS at any time. Insertion and
removal of the reagent cartridge and sample deck are the only manual interactions with the
instrument.

4.2 Sample Deck
The Sample Deck is an assembly containing all the commonly required single use components for a
library preparation workflow. It uses a re-usable sample deck frame supplied with the instrument,
along with a reusable deck stand. The sample deck is loaded with a set of single-use disposable
assemblies containing the generic reagents used in all the MagicPrep NGS system workflows.
4.3 Reagent Cartridge
The Reagent Cartridge is a single use consumable containing pre-metered volumes of reagents
required for a specific library preparation protocol.
The reagent cartridges should be stored in a freezer at ≤-20 °C until required for use.
The MagicPrep NGS instrument reads the 2D bar code on the upper surface of the reagent cartridge,
which determines the specific library preparation protocol to be employed.

When handling the sample deck parts and reagent cartridges, follow your institution’s
safety procedures for working with chemicals and handling of biological samples. Follow
good laboratory practices and safety requirements. Wear a lab coat, disposable gloves
and protective glasses where necessary. Changing gloves between handling samples is
recommended to avoid cross-contamination between samples orreagents.

5 Installation

5.2 Unpacking
The MagicPrep NGS instrument has been designed to be operator installable.
The MagicPrep NGS instrument weighs 18.8 kg and should be lifted by two people
On receipt of the shipping package, cut the black or white plastic straps securing the
MagicPrep NGS shipping package to the wooden palette.
Using 2 people at each end of the package, lift the shipping package onto the laboratory
bench or similar.
Inspect the shipping package for any severe damage such as rips, tears, holes etc. If in any
doubt as to the nature of the damage, contact Customer Support. Lightly deformed edges,
scuff marks etc., are considered normal wear and tear through the shipping/transport process.
Remove any protective shrink wrap from around the packaging (if used) and cut the black or
white plastic packaging strips and remove and dispose.
Remove the top brown cardboard tray and place to one side.

Remove the cardboard insert and power cable from inside the box and place to one side.
Lift the outer shipping brown cardboard casing up off the bottom cardboard tray and place to
one side.
Inspect the interior sales packaging around the MagicPrep NGS instrument and note if there is
any damage. The foam packaging supports are designed to protect the MagicPrep NGS
instrument.
Using two people, lift the MagicPrep NGS box up, remove the lower shipping cardboard tray to
one side and remove the protective foam supports from each end.
Cut the black or white plastic packaging strip and remove and dispose
Lift the sales packaging cover up and off the MagicPrep NGS instrument. Take the small
cardboard box containing the re-usable sample deck and sample deck bench support and
power cable away from the instrument and place to one side.
Remove the foam packaging/protective material from the front and rear of the instrument.
Using two people, one at each end, lift the MagicPrep NGS instrument out of the white
cardboard tray, remove the plastic packaging and place in position on the laboratory bench.
5.3 Installation
Air flow
Take care to position the MagicPrep NGS instrument in a location that allows sufficient space from
walls or other objects that might limit airflow. It is recommended to have an unobstructed gap of at
least 15 cm (6 inches) to the sides and the rear of the instrument.
Ensure that the MagicPrep NGS instrument is installed on a level surface.
Having placed the MagicPrep NGS instrument in a suitable location on the level laboratory
bench, remove the protective film from the front touch screen panel.
Remove the power cable from the small cardboard box and insert into the power socket at the
rear of the instrument.
Plug the power cable into the laboratory power socket (complying with the power requirements
noted in section 1.5 –Power Supply).
Switch the power switch at the rear on.
