1SAFETY INFORMATION
1.1 General
Lattice Lightsheet 7 is a stand-alone light sheet system in an inverted configuration. It consists of the Lattice
Lightsheet system proper, laser cassette, incubation components, camera with liquid cooling system and
computers.
Lattice Lightsheet 7 systems are designed for research only and not for medical diagnostic, therapeutic or
treatment purposes. Not all products are available in every country. Contact your local ZEISS representative
for more information.
The Lattice Lightsheet 7 microscopes, including their original accessories and compatible accessories from
other manufacturers, may only be used for the purposes and microscopy techniques described in this
operating manual.
If the device system is used in a manner not specified by the manufacturer, the protection
provided by the device system may be impaired.
The Lattice Lightsheet 7 microscope is intended for volumetric imaging of biological samples at high speed,
whereby the specimen is illuminated by a structured sheet of light.
Installation and commissioning of the Lattice Lightsheet 7
systems must be performed by
authorized ZEISS service
staff. The system should not be used prior to instruction by a ZEISS
representative.
The manufacturer
will not assume liability for any malfunction or damage caused by anything
other than the intended use of the Lattice Lightsheet 7 systems or individual modules or parts of
them, nor by any repair or other service operation performed or attempted by persons other than
duly authorized service staff. Any such action will invalidate any claim under warranty, including
parts not directly affected
by such action. This also includes the modification of the system
computer with new cards, etc. by the user. Any manipulation of the laser safety system will result
in the loss of warranty of laser safety.
A cooling liquid defined as a hazardous substance is used in the Lattice Lightsheet 7 to cool the
detection modules. The supplied safety data sheet with notes on hazards and safety measures
must be observed when handling the cooling liquid.

Please read the notes on device safety and manuals of additional devices, if ordered.
As the system is largely operated via menus on a computer, you should be familiar with the principles of the
WINDOWS operating system and its graphical user interface. The respective manuals are supplied together
with the programs.
The Lattice Lightsheet 7 system are laser hazard class 2 devices. The systems are equipped with
safety interlocks that comply with laser hazard class 3B. WHO recommendations concerning
health and industrial protection when handling laser devices must be observed. The operator of
the units must also observe all and any relevant statutory accident prevention regulations.
The user is referred to the safety data sheet provided together with the manual.
1.2 Explanation of Symbols
CAUTION:
Faults and hazards that might arise during operation which might cause damage to the unit or
injury to the user. Observe the operating manual.
CAUTION:
Laser radiation hazards possible when operating the system. Do not expose yourself to the laser
beam.
CAUTION:
Optical radiation hazards possible when operating the system. Do not expose yourself to the
radiation.

Pull the mains plug before opening the device housing.

1.3 Information Concerning the Use of the Device
The Lattice Lightsheet 7 systems meet the EMC requirements for EN 55011 Class A (intended use in industrial
environment). If the system is operated in a residential area or in a small trade area other devices may be
influenced by conducted or radiated disturbance. In this case special EMC measures are required.
EMC test according to DIN EN 61326-1
1. Noise emission according to CISPR 11 / DIN EN 55011
2. Noise immunity according to table 2 (industrial sector)
In the rare case of a voltage surge on the wired data lines such as USB, LVDS, Sub-LVDS (e.g. from
indirect lightning stroke), a momentary interruption of the acquisition system's functionality is possible.
The imaging can stop or a short interruption of the camera image is possible. This is no defect.
Acquisition can be continued after restarting the ZEN software. Sample position is maintained and no
damage occurs to the system. A possible protective means to minimize the probability of a temporary
disruption is a transient voltage protector in your facility's power system.

1.4 Regulations
Extensive knowledge of the hardware and the system is indispensable for safe operation of the
Lattice Lightsheet 7 systems.
Read this operating manual and all device publications belonging to the system conscientiously
before operating the Lattice Lightsheet 7 system! You can obtain additional information on the
hardware configuration delivered and on optional system extensions from the manufacturer or
via the service hotline.
The Lattice Lightsheet 7 systems have been designed, built and tested in conformity with the following
regulations and guidelines:
−DIN EN 61010-1 (IEC 61010-1) "Safety requirements for electrical equipment for measurement, control
and laboratory use", taking relevant CSA and UL specifications into account,
−DIN EN 60825-1 (IEC publication 60825-1) "Safety of laser equipment",
−21 CFR §1040.10: "Performance standards for light emitting products – laser products",
−DIN EN 61326-1 "Electrical equipment for measurement, control and laboratory use – EMC
requirements",
−Low voltage directive: 2014/35/EU,
−EMC directive: 2014/30/EU, KN 61000-6-2, KN 11.

ZEISS has installed a process for taking back and recycling the instruments within the member states of the
European Union, which takes care of the appropriate utilization according to the said EU guidelines.
For details on the disposal and recycling please refer to your relevant ZEISS sales or service organization.
The product must not be disposed in the household waste or through the municipal disposal organizations.

1.5 Power Requirements
The following components must be connected to the AC mains grid:
−power supply unit
−3 point powerstrip (multipoint connector 2 or incubation components power strip)
−3 point powerstrip (multipoint connector 3 or storage components power strip)
The following components must be inserted in the 6-point power strip (multipoint connector 1 or system
components power strip, see Fig. 1), which itself is connected to the power supply unit:
−flat screen monitor
−control computer (system PC)
−sCMOS camera
−stage motion controller
−camera liquid cooling system
The following components must be inserted in the 3-point power strip (multipoint connector 2 or incubation
components power strip, see Fig. 1):
−gas mixer controller
−temp module controller
The folowing components must be connected to the 3-point power strip (multipoint connector 3 or storage
components power strip)
−analysis computer (storage PC)
−monitor
The following components must be connected to the power supply unit (PSU):
−laser module (LM)
−Lattice Lightsheet 7 module
−6-point power strip (multipoint connector 1 or system components power strip)
The storage and data analysis PC and respective monitor must be separately connected to the AC network by
further power cables.

The Lattice Lightsheet 7 system must be connected to the AC network via the Power Supply Unit (PSU) by a
country-specific mains power cable (Fig. 1).
The 6 point power strip must be separately connected to the AC network by further power cables.
Power connectors and fuses of the PSU, the laser module and the Lattice Lightsheet 7 are displayed in Fig. 2,
and Fig. 3, respectively.

Power supply unit (PSU) and laser module (LM) unit can be housed in the component rack (see Fig. 3). The
components 6-point power strip is connected to the multipoint connector for components and is attached
to the rack.

The PSU and the LM are pre-mounted, the MC will be mounted on-site.
The mains plug may only be inserted into a socket with a protective contact. The protective effect must
not be overridden by an extension cable without a protective conductor.
Detachable power cords should not be replaced with inadequately assessed power cords. Only the
specified power cords may be used.
Only the connection cables supplied with the device may be used. The use of commercially available
cables for use with the LLS7 is prohibited.
A safe disconnection from the power supply network is only guaranteed by pulling the power plug.
The switch on the controller only switches to standby mode.
Danger of voltage from contact with live parts. This can lead to burns and a fatal electric shock.
Switch off the microscope before cleaning. Do not touch any live parts. Disconnect live parts from
the power supply.

1.6 Environmental Requirements
To ensure peak performance and avoid possible damage, the system should be given some time to acclimate
if it was stored at very low temperatures prior to setup. Otherwise, air humidity might condensate on the
system’s components, or mechanical stress in optical components may cause damage or misalignment. The
system has to be stored at room temperature for 24 hours prior to unpacking. Opening the top of the
shipment crates will help to speed up the process. As abrupt temperature changes must be avoided in any
case, ensure to open the shrink-wrapped plastic packaging of the component rack only after a sufficient
warm-up time. For the same reason, keep the cardboard packaging of other major components, such as the
Lattice Lightsheet 7 module and user PC sealed during acclimation.
For operation the system must be placed in a closed room.
1. Operation, specified performance without
incubation T = 22 °C ±0.5 °C without interruption (24 h a day independently whether system is operated or
switched off)
It has to be ensured that the air-flow of the air-
conditioning is not directed at the system.
2. Operation, reduced performance without
incubation T = 15 °C to 30 °C, any conditions different from item 1. and 5.
3. Storage, less than 16 h T = 4 °C to 30 °C
4. Storage, less than 6 h T = -20 °C to 55 °C

These conditions do not include the requirements for high precision measurements. Please refer to the
operating manual of the microscope concerning special requirements.
At an environmental temperature of 22 oC the covered sample area will heat to about 25 oC. If sample
temperatures of 22 oC are required, the room temperature needs to be set at 20 oC. Note that
temperature control does not include cooling.

1.7 Notes on Setting up the System
Installation and commissioning of the Lattice Lightsheet 7
system must be performed by
authorized ZEISS service
staff. The system should not be used prior to instruction by a ZEISS
representative.
The Lattice Lightsheet 7 systems are delivered in several crates.
The Lattice Lightsheet 7
system must be set up so as to ensure that the minimum clearance
between the wall and the rear of the system is no less than 0.5 m. This clearance is needed for
adjustment and maintenance operations.
The safe disconnection of the Lattice Lightsheet 7
system from mains supply is performed by
removing the power supply cables of the PSU and the multipoint power strips
. The
Lattice Lightsheet 7 system must be set up and operated so that removing the connectors is not
difficult.
The liquid cooling system must be placed such that the air flow of the systems is not coupled.
The device must be placed securely on the table surface to prevent slipping and falling.
The system contains components with dangerous voltage. The
system may be opened only by
authorized ZEISS service staff. Before opening a system component the power plug must be
disconnected.
Do not set up the unit in the proximity of heat sources such as radiators or direct sunlight. To
avoid heat build-ups, the v
entilation louvers or slots on the microscope system must not be
covered up. Do not position the electronics parts in a closed container.
The system must not be set up in areas with potential danger by explosives.
The unit must be connected to a properly installed socket outlet with earthing contact by means
of the mains cables supplied. Continuity of PE connection must not be affected by the use of
extension leads.
Maintenance, repairs, modifications, removal or exchange of components, or other interference
with the equipment beyond the operations described in this manual may only be carried out by
the manufacturer ZEISS or by persons expressly authorized by ZEISS to do so.
This applies especially to the Lattice Lighsheet 7 system, incubator, stage, stage controller, lasers,
the PC system, the power supply units, cable connections and other system components.
Please note that the Lattice Lightsheet 7 system is a high-precision opto-
electronic instrument.
Inexpert handling may easily impair its function or even damage it and will invalidate any claim
under warranty.
For reasons of laser safety, all ports must either be equipped with the corresponding device
(camera or safety plate) or covered with the counterpart of the laser safety kit provided. The user
is not allowed to remove any parts.

The openings for ventilation must not be covered.
Before connecting the main cables and switching on the device, please check whether your mains
voltage is suitable for this device and corresponds to the voltage specified on the rating plate of
the laser module.
Do not allow the table power supplies to come into contact with moisture.
Make sure that no liquids get inside the device.
If the housing is damaged, the power supply unit must be taken out of service. The microscope
may only be operated with the desktop power supply unit supplied.
Note that the power of the motor drives of the scanning stage is strong enough to squeeze your
fingers e.g. between the housing and the scanning stage.
After installation or after conversion of the Lattice Lightsheet 7 system, authorized specialized staff must
carefully check that it is in a proper condition and, particularly, that covers protecting against laser radiation
are provided.
By establishing a corresponding workplace environment, please ensure that the formation of electrostatic
charges is avoided.
To avoid vibrations during operation, the Lattice Lightsheet 7 systems should only be operated in conjunction
with the system table (vibration damping).
The Lattice Lightsheet 7 should be placed in a room suitable and dedicated for microscopes.
Do not operate the devices included in the scope of delivery in a potentially explosive atmosphere,
in the presence of volatile anesthetics or flammable solvents such as alcohol, petrol or similar
substances.
If it is determined that protective measures are no longer effective, the device must be taken out
of operation and secured against unintentional use. Contact ZEISS Customer Service or Carl Zeiss
Microscopy Service to have the device repaired.

1.8 Warning and Information Labels
The warning and information labels attached on the Lattice Lightsheet 7 system (Fig. 4 and Fig. 5)
must be observed. Check whether all of the labelsshown below are provided on your instrument,
and contact ZEISS Germany or one of the service agencies if you should discover that any of the
labels should be missing. You will receive a free replacement.

1.9 Notes on Handling the Laser Components and Illumination Systems
The Lattice Lightsheet 7 system is a laser hazard class 2 instrument.
Caution: Use of controls or adjustments or performance of proceduresother than those specified
herein may result in hazardous radiation exposure.
The following laser types are currently intended for use in the Lattice Lightsheet 7 system. The use of any
other lasers as the ones listed below is not authorized.
1 Laser 488 nm, 10 mW, laser class 3B
2 Laser 561 nm, 10 mW, laser class 3B
3 Laser 640 nm, 5 mW, laser class 3B
Please contact ZEISS if you intend to use a laser type with a wavelength or output power other
than the ones above.
If used properly, the Lattice Lightsheet 7 systems will not pose any laser radiation risks for operating staff.
Nevertheless, you should observe the following warnings:
•
If necessary - insofar as specified by law -
inform the laser protection officer before
commissioning the laser.
•The laser module is equipped with a key switch.
•
The laser module is equipped with an electric interlock at the rear of the laser module (see
Fig. 4). This can be used to connect remote interlock.
•Always keep the keys for the laser module in places which are not accessible to persons who
are not authorized to operate the laser.
•A white LED (Fig. 5/1) o
n the front of the component rack lights up, once the lasers are
enabled to emit light. Please note that the LED does not light up when just turning the laser
module on with the key. Additionally, the ZEN software has to be started to enable the lasers.
•At the rear of the laser module a port is located which is used to connect an optional external
signal lamp with a green/red LED light that lights up for better visibility when the lasers are
activated for light emission.
•The system may only be used by persons authorized to operate the laser.
•Do not place any reflecting objects into the beam path.
•Never open any covers or paneling (except the user interface described in this manual, i.e. the
incubator covers or doors for the immersion medium).
Suitable protective measures must be taken if gases, dust or vapors hazardous to health,
secondary radiation or explosive objects should arise on the specimen because of laser radiation.

As standard, your computer has one hard disk drive and one CD-ROM drive.

system, please ensure that the operating system is
installed including initialization and start file setup and that the ZEN program is also installed
When working with the hard disk, it is important to know that the more data it contains, the
slower its operation will become. Therefore, data that you do not need permanently should be
stored on a DVD, a CD-ROM or an external HD drive.
When handling CDs, CD-ROMs or DVDs, do not touch the data side of the disc. Do not apply
paper labels or write on any part of the disc, data side or label side. Wipe dust or fingerprints on
the disc with a softcloth fromthe center to the edge. Do not usebenzene, thinner,record cleaner,
or antistatic clothes. This may damage the disc. Do not expose the disc to direct sunlight or high
temperatures. Backup your data on a regular basis. Do not install any other software without
consulting your ZEISS representative.
Never turn your computer off before exiting the ZEN program and shutting down the WINDOWS
operating system. Otherwise, program files and/or data files may get lost.
The Lattice Lightsheet 7 computer is a system computer which controls a high-end microscope
system. The computer must not to be used as a general workstation with frequent updates of
windows or security patches. The ZEN software is released with a specific windows version and
patch and this must not be changed unless approved by Carl Zeiss Microscopy.
Necessary data transmission rates of the network are dependent on the tasks to be performed. For standard
data transfer and online condition monitoring a data transmission rate of 1 MBit/s is sufficient. For Remote
Desktop Support a data transmission rate of at least 5 Mbit/s is required. To transfer big data sets we
recommend a data transmission rate of at least 10 Mbit/s.
