GeneralDescription
TheNuAireDirectHeatDecon(DHD)AutomaticCO2DirectHeatIncubatorhasbeendesignedtoprovideareliablecontrolled
in‐vitroenvironmentforoptimumtissuecellculturegrowth.Thechamberalsoprovidesanenvironmentforthestorageand
preservationofgametesandanimaltissuecellculturesintendedforresearchatornearbodytemperature.
Therearefiveparametersthatcontributetooptimumgrowthconditions,theyare:
Humidity
Precisetemperaturecontrol
PreciseCO2control
Sterility
Reliability
LikeallNuAireequipment,thisIncubatorhasbeendesignedtoprovidethehighestqualitystandardsofperformancewith
matchingcomputertechnology,precisetemperaturecontrol,andCO2gascontrolsystemcombiningstate‐of‐the‐art
technologywithyearsofdesign,quality,andmanufacturingexperience.
1.1 IncubatorChamber
ThedesignandsizeoftheDHDinnerchamberprovidesalargecapacity(7cu.ft.),andeaseofuse.Thechamberwallsare
directlyheatedbyphysicallyattachedfoilheatingelementsonthesides,bottom,top,andbackofthechamber,providing
atemperatureuniformityof+0.3C.Aspace‐agehigh‐densityinsulationthathasahigh"R"ratingcoversthecomplete
outersurfacesoftheIncubatorinnerchamber.
1.2 IncubatorClosedloopchambersamplecirculationsystem&HEPAFilter
AcontinuousoperatingairpumpdrawsenvironmentalsamplefromthechamberandcirculatesitthroughaHEPAfilter
capsulethenthroughsensorsthatmeasureCO2volume(asapercentage)inthechamberformonitoringandcontrol.
1.3 IncubatorControlElectronics
TheNuAireIncubatorControlElectronicsisastate‐of‐the‐artmicrocomputerbasedcontrolsystemspecificallydesignedto
servicetheprecisecontrolrequirementsofthechambersenvironment,providingoptimumprogrammableconditionsfor
culturegrowth.Themicrocomputeris“user‐friendly”andprovidesa5x7LCDscreenwhichprovidesoperatingcontrol
parameters,statusindicators,additionalkeyoperationalparameters,andanimbeddedtouchpaneltopermitefficient
operatorentryofdata.
ThemicrocomputerissupportedwithReadOnlyMemory(ROM)containingexecutablesoftware,RandomAccessMemory
(RAM)fortemporarystorage,andElectronicallyErasableProgrammableReadOnlyMemory(EEPROM)forcontrolset
pointsandparameters.TheEEPROMprovidesforindefinitestorageofthesevaluesduringperiodsofpowerofforpower
interruption(powerfaulttolerant).
1.4 IncubatorCO2Control
TheNuAiredirectheatIncubatorincorporatesamicroprocessor‐based,non‐dispersiveinfraredCO2inductionsensor.
TheamountofenergyreceivedatthedetectorisanapproximatelogarithmicfunctionoftheCO2concentrationinthegas
betweensourceanddetector.ThewavelengthsusedareabsorbedonlybyCO2makingthemeasurementinsensitiveto
othercomponents,suchaswatervapor.Detectorlinearizationisperformedwith32bitdigitalaccuracy.Advanceddesign
providesaverystableoutputminimizingdriftandrequiringlessfrequentcalibration.Theoutputisdigital,alleviatingerrors
broughtaboutbyanalogsignals.Calibrationofthiscontrolisaccomplishedbyazero&spancalibrationdoneonthesensor
andadisplayoff‐setcalibration.ThesecalibrationsareaccessiblethroughtheNuTouchLCDlocatedonthefrontofthe
unit.

1.5 IncubatorConstruction
Theoutershelloftheair‐jacketedIncubatoriscold‐rolledsteelwithapowdercoatpaintfinish.Thefrontframesurfaceof
theoutershellisheatedwithafoiltypeheaterdirectlyattachedbehindthefrontperimeteropening.
Thefrontframeperimeterheater,aswellastheouterfrontdoorheaterisdutycyclecontrolled(manuallyadjustedfor
specificambientconditions)tobalancetheheatthatreachesthechamberandthusreducesthepossibilityofcondensation
formingontheinnerglassdoorandtheinnerchamberwalls.
Theinnerchamberis16gauge,type304L,polishedstainlesssteelusingcrevice‐freeconstruction,whichprovidesaneasily
cleanableinertsurfacethatdoesnotinitselfpromotebiologicalgrowth.Inaddition,allshelves,shelfsupports,andguide
railsareeasilyremovableandcanbeautoclavedseparatelyifsodesired.
Remember:Thechamberenvironmentisnotselective.Thegrowthenvironmentisappliedequallytoallmicroorganisms
(specimensandcontaminates)withinthechamber.
1.6 IncubatorHumidity
ModelsNU‐5800,NU5810,NU‐5830&NU‐5831
Arelativehumiditylevelofupto90%isachievedintheIncubatorbytheuseofastainlesssteelpanfilledwithdistilled
waternopurerthan1megaohm,andplacedonthebottomofthechamber.Itisnecessarytosetthedutycycleofthe
doorandfrontperimeterheaterinproperproportionstoreducethepossibilityofcondensationformingontheglassinner
doorandthechamberwalls.Itisalsoimportanttothoroughlywipethewallsandtheglassdoorcleanbeforeaddingthe
humiditywaterpan.Condensationwilloccurmorereadilyatcontaminationpoints.Thereisnoelectronicsensingandthus
noautomaticcontrolofthehumiditylevelintheairjacketedIncubator.Anairpumpinjectsfreshairintothechamberata
presetdutycycletoreducecondensationformingonthechamberwallsandfrontglassdoor.Theairinjectionsaresettable
bytheusertohelpcontrolcondensation.(Seesection8.8forthisoption.)
ModelsNU‐5820,NU‐5840&NU‐5841useahumiditymonitoring/controlsystemforchamberhumidity,thesystemis
externaltothechamber.Thechamberairisroutedthroughthewatervaporsaturatedairinanevaporatorboxtoadd
humiditywhentheRHsensorindicatesthatmorehumidityisrequiredmaintainasetpoint.
1.7 CabinetVentilatorFan
AnaxialfanismountedtothebottomcoverpaneloftheIncubatorandrunscontinuouslywhentheunitisswitchedon.
Thisfanpullsairintothecabinetshellthruthetoppanellouveropeningsanddischargesitoutofthebottomoftheunit.
Theoperationofthisfanisnecessarytoassureaccuratechambertemperaturecontroloverthe60Fto85F(15Cto30C)
ambienttemperaturerangethattheequipmentmaysee.
1.8SafetyInstructions
ThesesafetyinstructionsdescribethesafetyfeaturesoftheINVITROCELLIncubator.
Theincubatorhasbeenmanufacturedusingthelatesttechnologicaldevelopmentsandhasbeenthoroughlytestedbefore
delivery.Itmay,however,presentpotentialhazardsifitisnotusedaccordingtotheintendedpurposeoroutsideof
operatingparameters.Therefore,thefollowingproceduresmustalwaysbeobserved:

Theincubatormustbeoperatedonlybytrainedandauthorizedpersonnel.
•
Foranyoperationofthisunit,theoperatormustprepareclearandconcisewritteninstructionsforoperatingand
cleaning,utilizingapplicablesafetydatasheets,planthygieneguidelines,andtechnicalregulations,inparticular.

304L16gagestainlesssteel,usingcrevice‐freeconstruction.Allexposededgesaredeburredtoinsurenosharp
edges.Theexterioriscoldrolled16&18gaugesteelfinishedinapowdercoatedpolyurethanefinish,whichis
resistanttochemicalsandeasilycleanedusingmildhouseholddetergents.
TheDHD'smicrocomputertemperaturecontrolsystemhastwotemperaturesensorslocatedinsidethechamberat
thebackwall.Thetemperaturesensorscomparethevaluestoasetpointandexecuteadifferentialcontrol
algorithmthatenergizesasolid‐stateswitch,supplyingpowertotheheaters.
Easilyremovableinnershelvesandrailsforsterilization.
Upto21shelvescanbeplacedinsidethechamber(4shelvesareshippedwitheachDHD).
Space‐agehightemperaturematerialisusedtoinsulatetheinnerchamberwalls.
Foilheatersaredirectlyattachedtothechamberwallsandthetopandbottomsurfacesoftheinnerchamber.
Mostelectronics,motors,pumps,andvalvesarefullyaccessiblefromthebackoftheunit.
ACO2sampleportisprovidedonthefrontpaneljustbelowthedoortochecktheconcentrationofCO2inthe

3.0 TestPerformance&Procedures
AllequipmentisthoroughlyinspectedattheNuAireFactoryatthetimeofshipment.Qualitycontrolismaintainedby
constantsurveillanceovertheproduct,beginningatthereceiptofpurchasedmaterialandconcludingwithafinalinspection,
whichcertifiestheIncubatorperformancetothespecifications.Inallinstanceswhereproductqualitycannotbeeasily
assessedontheenditem,theproductisinspectedduringsub‐assemblyfabrication.Thefollowingtestproceduresare
conductedoneveryDHDIncubatorandacopyofthetestreportisincludedwitheachIncubator.
4.1 VisualInspection
EachIncubatorisvisuallyinspectedtoinsurethattheinterioriscleanandfreefromscratches,nicks,andburrs
andthatallwelds,bothinteriorandexteriorarefinished.
Paintedsurfacesareinspectedtobefreeofscratches,nicks,insufficientcovering,andnoticeablepaintruns.
Thedoorsopenandclosefreelywithoutbindingofthehinges.
ThegasketsealstheinnerglassdoortotheIncubatortightlyacrossitsentireopening.
Theglassdoorisfreeofscratches.
3.1 ElectricalTests

corrected,andanyfailedtestissuccessfullyrepeated.Changesfromanydefaultvalueoftheoptionalconfiguration
parametersmeanttoimproveperformancearerecordedonthisreportforyourreference.

5.0 Warranty
NuAire,Inc.warrantsthatitwillrepairF.O.B.itsfactoryorfurnishwithoutchargeF.O.B.itsfactoryasimilarparttoreplace
anymaterialinitsequipmentwithin24monthsafterthedateofsaleifproventothesatisfactionofthecompanytohave
beendefectiveatthetimeitwassoldprovidedthatallpartsclaimeddefectiveshallbereturned,properlyidentifiedtothe
companyatitsfactory,chargesprepaid.Factoryinstalledequipmentoraccessoriesarewarrantedonlytotheextent
guaranteedbytheoriginalmanufacturerandthiswarrantyshallnotapplytoanyportionoftheequipmentmodifiedbythe
user.ClaimsunderthiswarrantyshouldbedirectedtoNuAire,Inc.settingforthindetailthenatureofthedefect,thedate
oftheinitialinstallationandtheserialandmodelnumberoftheequipment.
ThiswarrantyshallnotapplytoanyNuAireproductorpartthereofwhichhasbeensubjecttomisuse,abuse,accident,
shippingdamage,improperinstallationorservice,ordamagebyfire,floodoractsofGod.Iftheserialnumberofthisproduct
isaltered,removed,ordefacedastobeillegible,theWarrantyshallbenullandvoidinitsentirety.
Thewarrantyisforthesolebenefitoftheoriginalpurchaserandisnotassignableortransferable.Priortoreturninganyitem,
foranyreason,contactNuAireforaReturnAuthorizationNumber.Thisnumbermustaccompanyallreturns.Anyproduct
shippedtoNuAirewithoutthisnumberwillbereturned,refusedshipmentorcollectfreight.
6.0Shipments
NuAire,Inc.takeseveryreasonableprecautiontoassurethatyourIncubatorarriveswithoutdamage.Motorcarriersare

6.1 DamagedShipments
6.1.1TermsareF.O.B.factory,unlessstatedotherwise.
Therefore,itisimportanttocheckeachshipmentbeforeacceptance.
6.1.2Ifthereisvisibledamage,thematerialcanbeacceptedafterthedrivermakesanotationontheconsignee'scopyof
thefreightbill.Thenaninspectionmustbemadetoverifytheclaimagainstthecarrier.Thisinspectionisthebasis
ofyourfilingtheclaimagainstthecarrier.
6.1.3Ifconcealeddamageisfound,itisabsolutelynecessarytoNOTIFYTHEFREIGHTAGENTATONCE,andrequestan
inspection.Withoutthisinspection,thetransportationcompanymaynotacceptaclaimforlossordamage.Ifthe
carrierwillnotperformtheinspection,anaffidavitmustbepreparedstatingthathewascontactedonacertaindate
andthathefailedtocomplywiththerequest.This,alongwithotherpapersinthecustomer'spossessionwill
supporttheclaim.

7.0 Installation
TheIncubatorisfastenedtothebaseskidanditisusuallythebestproceduretoleavetheskidattacheduntiltheIncubatoris
locatedinitsapproximatepositiontofacilitateeaseinhandling.Thebaseskidcanthenberemovedbyremovingthefour
boltsholdingthecabinettotheskid.Thenscrewthelevelingfeetintothesamenutsthatwereusedtofastentheincubator
totheskid.Theincubatormustbeaminimumof2.0inchesabovethefloor.Usethe1.0”spacersprovidedonthethreaded
stemofeachlevelingfootwhentheyareinstalledtoensurethisspacing.SeeBCD‐14784.
ExaminetheIncubatorcarefully.INSPECTboththeexteriorandtheinteriorforanytransitdamagebeforediscardingthe
shippingcrate.
7.1 Location
InlocatingtheIncubator,considerallpossibleconditionsthatmightaffectitsperformanceaswellaslaboratoryprocedures
foritsintendedpurpose.Donotlocatenearheatingorcoolingducts,ornexttoequipmentthatgeneratesheat(steam
radiators,stoves,ovens,autoclaves,etc.).Avoiddirectsunraysandrapidlymovingaircurrents.Theseconditionsadversely
affecttheevenheatdissipationrequiredfromtheexteriorsurfacesoftheIncubatorandmaycausethetemperature
variationinthechambertoexceedspecificationsasstatedinsection3.2.SincetheIncubatorneedsevenheatdissipation
onallsurfacesinordertomaintainaninternaltemperaturevariationoflessthan±0.2°C,aminimumof2inches(50mm)
mustbeallowedbetweentherearandsidesoftheIncubatorandanywalls,partitionsorobstructionstofacilitateadequate
convectionofairaroundtheIncubator.Confirmclearancewithatapemeasureifneeded.AdjusttheIncubatorlocation
accordingly.Inaddition,theIncubatorlocationshouldaddressaccesstoitspowercordforconnection/disconnectionif
necessary.Formaintenanceandservicepurposes,thecontrolcenterelectronicsshouldremainaccessible.Leaveatleast3
ftofclearanceinfrontfortheswingingdoor.
Note: WhenlocatingIncubatorsside‐by‐side,theyshouldbespacedatleast10”aparttopermitone

7.2 Leveling
Priortousetheincubatorshouldbeleveledusingabubblelevelonamiddleshelfinthechamber.TheIncubatorshould
haveall4levelingfeetfirmlyonthebenchorfloor.Levelfromsidetosideandfronttoback.Byturningtheadjustable
levelingfeetcounter‐clockwise,raisestheIncubator.Thelevelingfeetheightshouldallowfora2inches(50mm)space
betweenIncubatorbaseandthefloor.Thereisa1inchlongStainlessSteelspacersuppliedwitheachfoot.Besurethatit
isinplacewhenyouarelevelingtheincubator.
Note: ItisimportantthatthebottomoftheincubatorbeoffthefloorbyATLEAST2inches(50MM)

AllNuAireIncubatorsuseCO2/N2insuchquantitiesthatprecisemeteringofCO2/N2inputpressureisimportantfor

TheIncubatorisdesignedtoprovideasterile,constanttemperature,constantCO

levelandnaturallyhumidifiedatmosphere
foroptimumgrowthoftissuecellculturesandotherorganismsrequiringthispreciseenvironment.TooperatetheIncubator
properly,thefollowingparametersmustbereviewed,carefullyset,and/orprepared.
8.1 Sterility
Thechamberenvironmentisnotselective.Thegrowthenvironmentisappliedequallytoallmicroorganisms(specimens
andcontaminates)withinthechamber.Therefore,beforeplacinganyculturesintheIncubator,theshelvesandsidewall
topplenumsshouldbesterilized.Theinteriorsidewalls,top,bottom,door,aswellasthegasketshouldbewipedclean
witha70%solutionofisopropylalcoholorotherdisinfectantcompatiblewiththechamberconstruction,toremoveany
contamination.UsemilddetergenttocleantheexterioroftheIncubator.ThisIncubatorprovidesthelabprofessionalwith
achoiceof2heateddecontaminationcycles.(Seesection10.1forfurtherinformation.)
8.2 Humidity
WaterPanavailableonallModels
HumidificationoftheIncubatorchamberisachievedthroughtheprocessofwaterevaporation(vaporwaterpressure)from
astainlesssteelwaterpanplacedonthebottomofthechamber.Materialsofdifferentthermalresistance(e.g.,glass,
plastic)donotoffersufficientthermalrecoveryandarenotrecommendedforwaterpanusage.Althoughsomemetals
offerbetterthermalcoefficientsthanstainlesssteel,dissimilarmetalscauseelectrolysisintheacidatmosphere(carbonic
acid)andshouldneverbeused,orplacedwithinthechamber.
Useonlydistilledormineral‐freewater,nopurerthan1megaohm,inthestainlesssteelpan.Thewatershouldbe
changedatleastonceaweek.FLOODINGTHEBOTTOMOFTHEINCUBATORISNOTRECOMMENDEDsinceitisdifficultto
changethewaterweeklyandalmostalwaysnecessitatestheuseofchemicals,whicharenotrecommendedandmay
damagethestainlesssteel.Also,itpromotescondensationontheinnerwallsbecauseitstealsthenaturalconvection,heat
flowthroughtheinnerchamberandcondensationpointsoccur.

Thecontrolsystemusesasolid‐statecapacitancehumiditysensortomonitortherelativehumiditywithinthechamber.
Ifhumidityisrequired,thecontrolsystemwillactivateasolenoidvalveroutingthechamberairthroughahumidified
heatedenclosurejustbeforebeingreturnedtothechamber.Thecontrolwillcontinueroutthechamberairthroughthe
