Installation
Site Requirements
Before the system is installed by an authorized Biotage service
engineer, the installation site should be prepared as follows:
Location Operate the system in a well-ventilated fume
hood verified as meeting the requirements of a
“non-hazardous area” with regards to potentially
explosive atmosphere and utilize the fume hood’s
safety features. If an internal system leakage
occurs, liquids are drained through drain ports
underneath the system. The system must therefore
be operated in a fume hood that has the capacity
to handle leakage of potentially flammable
liquids. Note that the maximum leakage volume
is determined by the size of the used solvent
reservoirs and that the flow rate is determined by
the flow rate setting for the run (up to 300 mL/min).
Ensure that the fume hood and location meet the
requirements of the system regarding dimensions,
weight, electrical supply, altitude, pollution degree,
ambient temperature, and humidity; see “Technical
Specifications” on page 4.
External Fire
Protection
External fire protection should be installed
according to local regulations.
Warning
» The system must be unpacked and installed by an authorized
Biotage service engineer.
» Follow regional safety practices when handling and moving
shipping boxes and containers, and when moving the system.
» Observe general and specific safety regulations for the use of
the system and its accessories and consumables at all times,
in order to reduce the risk of personal injury, fire, and/or
explosion; see “Safety” on page 5.

Software License Agreement
Biotage Sweden AB licenses the Biotage®Selekt software to
you only upon the acceptance of all of the terms and conditions
in the software license agreement supplied with the system.
By using the software, you consent to be bound by and are
becoming a party to that agreement.
Delivered Items
Note: We recommend that the boxes and packing materials are
kept by the customer in case the system needs to be returned
for service or moved to another location.
Note: The system’s internal tubing is flushed with 80% ethanol
in deionized water and then emptied before the system is
shipped to the customer.
Check the delivery against the shipping documentation to ensure
that all parts are included in the shipment. If any part of the order
is missing or damaged, please contact
Biotage®1-Point Support™.

Installation
Move the System
Warning
» At least two persons are required when lifting the system.
» Follow regional safety practices when moving the system.
If moving the system within the laboratory or between
laboratories in the same building, follow the instructions below.
If you need to ship the system, please contact Biotage 1-Point
Support for instructions.
1. When the system is not processing, shut it down:
a. Press Menu, Shut Down, and then Yes to confirm
b. When the message saying it is safe to turn off the system
appears on the touch screen, turn off the system. The
power switch is located on the left side of the system.
2. Unplug the power cord from the power outlet.
3. Remove all solvent and waste reservoirs.
4. Remove the racks.
5. If using a secondary solvent containment on top of the
system, remove the drain tube at the rear of the containment
(see Figure 4 on page 3).
6. Disconnect any external equipment connected to the ports
at the rear and on the left side of the system.
7. Carefully lift the system by the base plate and place it on a
trolley that can support the weight of the system. At least
two persons are needed.
8. Move the trolley and the rest of the equipment to the new
location.
9. Carefully lift the system by the base plate and place it in a
well-ventilated fume hood or an equivalent enclosure that
meets the site and safety requirements. Place it in a position
that allows access to the right side and the rear of the
system. At least two persons are needed.
10. If using a secondary solvent containment, connect the drain
tube to the drain port (see Figure 4 on page 3). Insert the
other end into a waste reservoir.
11. If you want to use a mouse, connect it to one of the USB
ports on the left side or at the rear of the system.
12. Connect the system to a grounded (earthed) power outlet
using the supplied power cord.
13. Carefully slide the system into a position where the
operator can access the power switch, touch screen,
column holder(s), and collection area.
14. Level the system by adjusting the height of the four feet.
15. Insert the waste outlet tube into a waste reservoir and the
solvent tubes (labeled S1 to S4) into their solvent reservoirs.
Note: We recommend having all solvent reservoirs on
the same physical height to improve accuracy in the
solvent mixing.
16. Turn on the system.
17. Once the system is operational, check all tubes and
connections for leaks using the flush function. Flush with
water or another suitable solvent. If leakage is observed,
stop the flush and check for loose fittings or damaged
tubing. For instructions and more information, see the
user manual (P/N 416183).



Power Supply and Communication Connections
Warning
» Use only a power cord supplied by Biotage. The cord should be
inspected periodically and replaced if damaged.
» The power cord plug must not be altered, and the plug should
only be connected to a grounded outlet as per local and
national regulations.

Tube Connections
Warning
» Only use a waste reservoir that is grounded or made of glass.
» To prevent leakage, inspect all tubes before each run to ensure
that they do not show signs of wear or damage and that they are
properly connected and tightened.
» Use caution when finger tightening fittings to prevent stripped
threads or crushed ferrules.
» Use only tubing designed for the Selekt system and supplied
by Biotage.
» Connect the solvent tubes labeled S1 to S4 to the S1 to
S4 ports and then insert them into solvent reservoirs.
» Connect the flow cell inlet tube to the UV port.
» Connect the column inlet tubes labeled C1 (orange
tag) and C2 (blue tag) to the C1 and C2 ports.
» Connect the column outlet tubes labeled C1 (orange
tag) and C2 (blue tag) to the R1 and R2 ports.
» Insert the waste outlet tube into a waste reservoir.
If necessary, use the waste extension tube
supplied with the system (P/N 411851).
» If using the optional secondary solvent
containment, connect the supplied drain tube to
the drain port at the rear of the containment (see
Figure 4) and insert it into a waste reservoir.

Safety
Overview
The system must be operated in a fume hood that has the
capacity to handle leakage of solvents and provide suitable
airflow to prevent the build-up of flammable vapors.
The operating temperature range for the system is 15°C to 32°C
(59°F to 95°F).
Should a leakage or solvent spill occur, your laboratory must have
local regulated processes in place to deal with these incidents in
order to prevent the possibility of a fire or explosion hazard.
Intended Use
Biotage®Selekt from Biotage is intended solely to detect,
fractionate, and purify chemical mixtures using flash
chromatography.
The system must be operated in a laboratory environment by
trained professionals. The system is not designed to be operated
in a potentially explosive atmosphere. It is the responsibility of
the customer to classify (zone) their particular environment in
order to verify that it meets the requirements of a “non-hazardous
area” with regards to potentially explosive atmosphere.
All operations must be performed:
» According to the user documentation
delivered with the system.
» According to instructions available at www.biotage.com.
» According to instructions provided through
dialogs appearing on the screen.
» According to instructions given by the
technical support staff from Biotage.
» Within limits set by the system’s technical specifications.
Failure to follow those instructions and operate within the limits
set by the technical specifications may result in personal injury
and/or equipment damage.
Education, Training, and Competence
It is your responsibility to provide all applicable health and
safety regulations to your personnel. You must also ensure that
all personnel involved in the operation and maintenance of the
system fulfill the following criteria:
» Have the necessary education, training, and competence
required for the intended use of the system.
» Observe general and specific safety regulations
for the use of the system and its accessories and
consumables at all times, in order to reduce the
risk of personal injury, fire, and/or explosion.
Static Electricity
When using Selekt flash purification systems, care must be
taken to avoid static electricity build-up. This is particularly
important when using the systems with flammable,
non-conducting solvents operating under high flow rate
conditions. Static electricity can be generated as non-polar
liquids flow through columns, tubing, pipes, valves, and filters
during chromatographic operations. It can also be generated
by the user, e.g. from friction between shoe soles and floor;
please follow your local regulations that apply to static
electricity hazards.
There are a number of simple steps that should be taken as
important safety precautions:
» Do not exceed recommended flow rates for flash
chromatography columns (this information can be
found in the column documentation) nor allow flow
rates higher than specified through any Samplet®,
dry load vessel, or pre-column that you may be using.
» When operating the system, especially when
using high flow rates, wear anti-static clothing and
shoes, and stand on an anti-static floor mat.
» Touch a grounded metal object, such as a metallic
water pipe, just before handling the column or system
to discharge any accumulated static electricity.
» Minimize the presence of air bubbles in the flow lines since
these can increase electrostatic charge significantly.
» Static electricity build-up is more common in low
humidity environments. Maintaining humidity above
65% can be helpful as a preventive measure.
» In the event of solvent leaks, immediately turn off the
system and disconnect the power cord. Do not wipe
solvent away while the system is operating. In the
event of leakage from a column, immediately turn off
the system, disconnect the power cord and allow all
solvent vapor to dissipate before removing the column.
Do not wipe away any excess solvent from the column
surface as this can generate additional static charge.
» Read and follow your local static discharge codes of
practice and available national recommended procedures
for the avoidance of hazards due to static electricity.
To guarantee safe and effective operation, it is absolutely
necessary to follow all of the safety precautions and
instructions in the user documentation supplied by Biotage.

Safety
Service
All service or upgrades must be performed by an authorized
Biotage service engineer. Before handing over the system for
service, it should be emptied of liquid and cleaned from
harmful residues.
It is the responsibility of the customer to inform Biotage
1-Point Support representatives if the system has been used
for analysis of hazardous biological, radioactive, or toxic
samples and/or solvents, prior to any service being performed.
When returning equipment to Biotage, this should be done
in accordance with the material return procedures supplied
separately by Biotage.
Labels Used on the System
The following labels are used on the system.
In accordance with all the essential requirements of
all applicable European product directives; see the
Declaration of Conformity document.
In accordance with both U.S. and Canadian safety
standards; see the Declaration of Conformity
document.
RoHS In accordance with the Restriction of Hazardous
Substances Directive; see “Restriction of Hazardous
Substances (RoHS) Directive” on page 7 and the
Declaration of Conformity.
This device complies with part 15 of the FCC Rules.
Operation is subject to the condition that this device
does not cause harmful interference. For details, see
the Declaration of Conformity document.
Subject to the Waste Electrical and Electronic
Equipment (WEEE) Directive; see “WEEE Compliance
Statement” on page 7.
Manufacturer.
Drain ports for leakage of potentially flammable
liquids. If leakage is observed, shut down the system
immediately, disconnect the power cord, and follow
the instructions in the “Leaks” section in the user
manual (P/N 416183).
Ultraviolet (UV) light can injure your eyes.
To prevent exposure, always turn off the system
before removing the flow cell. Never have the
system turned on when the flow cell is removed
or when the retaining nut (see A in Figure 5 on
page 8) is loosened.
Consult accompanying user documentation.

operating the system. Failure to install or use the system in the
manner specified by Biotage may result in personal injury and/
or equipment damage.
If the system has been damaged or does not function properly,
shut it down immediately and contact Biotage 1-Point Support
(www.biotage.com).

shipping boxes and containers, and when moving the system.
» Keep the mains plug easily accessible in case the system
needs to be disconnected quickly from mains power.
» Ensure that the power cord, cables, and tubing connected
to the system cannot come in contact with chemicals.
Corrosives and solvents can degrade the cord/cable
insulation and dissolve the tubing causing a risk of
electric shock, fire, and/or equipment damage.
» External fire protection should be installed
according to local regulations.
» Operate the system in a well-ventilated fume hood verified
as meeting the requirements of a “non-hazardous area”
with regards to potentially explosive atmosphere and utilize
the fume hood’s safety features. If an internal system
leakage occurs, liquids are drained through drain ports
underneath the system. The system must therefore be
operated in a fume hood that has the capacity to handle
leakage of potentially flammable liquids. Note that the
maximum leakage volume is determined by the size of the
used solvent reservoirs and that the flow rate is determined

» Do not connect any accessory to any port other than
what is specified in the user documentation.
Operation
» The system shall be used in accordance with the instructions
found in the user documentation supplied by Biotage.
» The system may be pressurized when
paused or when shut down.
» Never flush the system without a column mounted
on the system or without the column inlet and outlet
tubing coupled together. To prevent leakage, check
all tubes and fittings before flushing the system.
» Always ensure that the settings for the column
selected in the software are correct for the column
to be used before starting a run. Never exceed the
maximum pressure or flow rate for the used column.


Safety
Power Supply and Fuses
» Use only a power cord supplied by Biotage. The cord
should be inspected periodically and replaced if damaged.
» The power cord plug must not be altered, and
the plug should only be connected to a grounded
outlet as per local and national regulations.
» Use only exact replacement fuses specified
by Biotage (P/N 411916SP). Incorrect fuses
create a potential fire hazard.
Solvents and Waste
» To avoid damaging the system, ensure that all solvents
used with your system are free of particulate matter.
The filters on the solvent inlet lines should be cleaned
(sonicated) or replaced every 1000 hours of operation
or every 12 months, whichever comes first.
» Do not place solvent reservoirs larger than 5 liters
in the optional secondary solvent containment.
» Only use a waste reservoir that is grounded or made of glass.
» Electrical equipment can introduce ignition
hazards. Ensure that all solvent manufacturers’
recommendations are followed with respect to
handling, ventilation and operating environment.
» Monitor the waste reservoir to prevent
overflow during operation.
Lab Safety Procedures
» Follow all generally-accepted lab safety procedures
and applicable laws and regulations.
» Personnel working with or near the system
must wear protective clothing, safety gear, and
eye protection in accordance with applicable
local and national safety regulations.
» The system uses solvents. Always follow local
and national safety regulations and the solvent
manufacturer’s safety, handling, storage, and disposal
recommendations; refer to the safety data sheets (SDS).
Collection Arm
» To avoid being struck by the collection arm (see C in
Figure 5 on page 8), keep hands away from the
collection arm area while the system is in operation.
» Never reach into the collection arm slot (see B in
Figure 5 on page 8) on the left side of the system.
Ultraviolet Light
» Ultraviolet (UV) light can injure your eyes. Always
turn off the system before removing the flow cell.
Never have the system turned on when the flow cell
is removed or when the retaining nut is loosened.
Leakage and Maintenance
» Always handle leakage immediately. If leakage is observed,
shut down the system, disconnect the power cord, and
follow the instructions in the “Leaks” section in the user
manual (P/N 416183). Do not wipe away any excess solvent
from the column surface as this can generate static charge.
» Follow all maintenance instructions in the “Maintenance”
chapter in the user manual (P/N 416183).
» There are potentially lethal voltages inside the
system. Do not remove the cover panels; there are
no user serviceable parts inside. If you experience
a problem with your system, please contact Biotage
1-Point Support immediately (www.biotage.com).
External Tubing and Fittings
» All tubing should be connected according to the instructions
in the user documentation supplied by Biotage.
» To prevent leakage, inspect all tubes before each run to
ensure that they do not show signs of wear or damage
and that they are properly connected and tightened.
» If using the optional secondary solvent containment,
ensure that the supplied drain tube is not damaged and
is safely connected to the drain port (see L in Figure 5
on page 8) and inserted into a waste reservoir.
» Use caution when finger tightening fittings to
prevent stripped threads or crushed ferrules.
» Use only tubing designed for the Selekt
system and supplied by Biotage.
Restriction of Hazardous
Substances (RoHS) Directive
The RoHS directive is a European Union-derived initiative
in which the elimination of certain hazardous substances
is the key objective. The elimination of these substances
will contribute to the protection of human health and the
environmentally sound recovery and disposal of equipment.
WEEE Compliance Statement
Valid for customers in EU countries
We are committed to being a good corporate
citizen. As part of that commitment, we strive to
maintain an environmentally conscious
manufacturing operation. The European Union (EU)
has enacted a directive on product recycling
(Waste Electrical and Electronic Equipment, WEEE).
Products falling under the scope of the WEEE Directive are
identified with a crossed over “wheelie bin” symbol on the
product label, as indicated to the left. To forward a product for
recycling or proper disposal, return them to Biotage Sweden AB.
Before forwarding a product for recycling or disposal, it should
be emptied of all liquid and cleaned from harmful residues.
When returning a product to Biotage, this should be done
in accordance with the material return procedures supplied
separately by Biotage.
